Six novel 2-substituted analogues of 1alpha,25-dihydroxy-19-norvitamin D(3), 6a,b-8a,b, were efficiently synthesized utilizing (-)-quinic acid as the A-ring precursor. The C2-modified A-rings were prepared as 4-alkylated (3R,5R)-3,5-dihydroxycyclohexanones 12-15 from (-)-quinic acid based on radical allylation at the C4 position of methyl (-)-quinicate. The new type of the CD-ring coupling partner 23 was synthesized from 25-hydroxy Grundmann's ketone 19 to apply to the modified Julia olefination to construct a diene unit between the A-ring and the CD-ring. The coupling yields, including a deprotection step, were 47-62%. After the separation of the diastereomers based on C2 stereochemistry, the structure (2alpha or 2beta) was determined by (1)H NMR experiments and compared to DeLuca's 2-methyl- and 2-ethyl-1alpha,25-dihydroxy-19-norvitamin D(3). Thus, the synthesized 2alpha-(3-hydroxypropyl)-1alpha,25-dihydroxy-19-norvitamin D(3) (8a) showed almost the same potency in binding to the bovine thymus vitamin D receptor (VDR) as the natural hormone 1, while its beta-isomer 8b had only a 3% affinity. Both 2alpha-allyl- and 2alpha-propyl-1alpha,25-dihydroxy-19-norvitamin D(3) (6a and 7a) and their 2beta-analogues (6b and 7b) possessed a weak affinity for the VDR. The strong VDR ligand 8a was ca. 36-fold more potent in induction of HL-60 cell differentiation than 1, and interestingly, even the weaker ligand 8b showed a 6.7-fold higher potency in the cell differentiation activity than that of 1.  相似文献

5-Phenyl-2-pentene (5Ph2P) was found to undergo monomer-isomerization polymerization with TiCl3–R3Al (R = C2H5 or i-C4H9, Al/Ti > 2) catalysts to give a polymer consisting of exclusively 5-phenyl-1-pentene (5Ph1P) unit. The geometric and positional isomerizations of 5Ph2P to its terminal and other internal isomers were observed to occur during polymerization. The catalyst activity of alkylaluminum examined to TiCl3 was in the following order: (C2H5)3Al > (i-C4H9)3Al > (C2H5)2AlCl. The rate of monomer-isomerization polymerization of 5Ph2P with TiCl3–(C2H5)3Al catalyst was influenced by both the Al/Ti molar ratio and the addition of nickel acetylacetonate [Ni(acac)2], and the maximum rate was observed at Al/Ti = 2.0 and Ni/Ti = 0.4 in molar ratios.  相似文献

Radical polymerization of N,N,N′,N′-tetraalkylfumaramides (TRFAm) bearing methyl, ethyl, n-propyl, isopropyl, and isobutyl groups as N-substituents (TMFAm, TEFAm, TnPFAm, TIPFAm, and TIBFAm, respectively) was investigated. In the polymerization of TEFAm initiated with 1,1′-azobiscyclohexane-1-carbonitrile (ACN) in benzene, the polymerization rate (Rp) was expressed as follows: Rp = k [ACN]0.28 [TEFAm]1.26, and the overall activation energy was 102.1 kJ/mol. The introduction of a bulky alkyl group into N-substituent of TRFAm decreased the Rp in the following order: TMFAm > TEFAm > TnPFAm > TIBFAm > TIPFAm ~ 0. The relative reactivities of these monomers were also investigated in radical copolymerization with styrene (St) and methyl methacrylate (MMA). In copolymerization of TRFAm (M2) with St (M1), monomer reactivity ratios were determined to be r1 = 1.07 and r2 = 0.20 for St–TMFAm, and r1 = 1.88 and r2 = 0.11 for St–TEFAm, from which Q2 and e2 values were estimated to be 0.35 and 0.44 for TMFAm, and 0.19 and 0.47 for TEFAm, respectively. The other TRFAm were also copolymerized with St, but copolymerization with MMA gave polymers containing a small amount of TRFAm units. The polymer from TRFAm consists of a less-flexible poly(N,N-dialkylaminocarbonylmethylene) structure. The solubility and thermal property of the polymers were also investigated.  相似文献

A perturbation theory for normal coordinates of nonadiabatic solvation is presented by means of the “string model” of chemical reactions. The dynamic normal coordinate is introduced for the perturbational treatment of the “intrinsic” normal coordinates that are orthogonal to the reaction path. The reaction is defined as the intrinsic reaction coordinate (IRC ) that is treated as a string. The string is thrown in the external force field that acts as a nonadiabatic source of perturbation. As an application of the present treatment, the effect of a water molecule for hydration reaction of formaldehyde is calculated. A second-order perturbation effect for the enhancement of the reaction rate is found.  相似文献

A study of the polymerization of methyl methacrylate initiated by the binary systems of some activated metals and organic halides has been made. It was found that the initiator activities of these systems were greatly dependent on the kind and the preparation or activation method of the metals (i.e., oxidation potential, surface area, and purity), and also on the kind of organic halides (i.e., bond-dissociation energy of their carbon–halogen bonds). From the kinetic studies of the polymerization at 60°C with the system reduced nickel–carbon tetrachloride, the rate of polymerization was found to be proportional to the monomer concentration and to the square root of concentration of both nickel and carbon tetrachloride at the lower concentration range of carbon tetrachloride, indicating that the system induced the radical polymerization. A similar conclusion was also obtained from the copolymerization with styrene with this system at 60°C, i.e., the resulting copolymer composition curve was in agreement with that obtained with azobisisobutyronitrile (AIBN). The apparent overall activation energy for the methyl methacrylate polymerization with this system was estimated to be 7.5 kcal/mole, which was considerably lower than that with AIBN. On the basis of the results obtained, an initiation mechanism for the polymerization with these initiator systems is presented and discussed.  相似文献

Abstract

Protons in alkaline-earth phosphate glasses give much higher mobility than those in silicate glasses. This is caused by the difference in O-H bonding state. Protons in P-OH form a strong hydrogen bonding with oxygen anion, resulting in formation of weak O-H bond.

The electrical conductivity of the glasses is proportional to the square of the proton concentration. The activation energy for the conduction is related to νOH, peak wavenumber of infrared absorption band (around~3000 cm?1), due to fundamental O-H stretching vibration. The mobility of protons increases with decreasing νOH, which depends on the species of ions included in the phosphate glasses.  相似文献

Using a highly sensitive flow‐type 27 MHz quartz crystal microbalance, we could detect a small mass change during stepwise and alternating one‐sugar transfer of glucuronic acid (GlcA) and N‐acetylgalactosamine (GalNAc) to an acceptor, catalyzed by chondroitin polymerase from Escherichia coli strain K4 (K4CP), and analyze the elongation mechanism of K4CP. K4CP was found to bind strongly to a chondroitin acceptor (Kd=0.97 μM ). Although the binding affinity and the catalytic rate constant for each monomer were considerably different, the apparent catalytic efficiency (kcat/Km) was similar (6.3×104 M ?1 s?1 for GlcA transfer and 3.4×104 M ?1 s?1 for the GalNAc transfer). This is reasonable for the smooth alternating elongation of GlcA and GalNAc on the acceptor. This is the first study to report the determination of kinetic parameters for enzymatic, alternated, sugar elongation.  相似文献

Covalently linked vancomycin dimers have attracted a great deal of attention among researchers because of their enhanced antibacterial activity against vancomycin‐resistant strains. However, the lack of a clear insight into the mechanisms of action of these dimers hampers rational optimization of their antibacterial potency. Here, we describe the synthesis and antibacterial activity of novel vancomycin dimers with a constrained molecular conformation achieved by two tethers between vancomycin units. Conformational restriction is a useful strategy for studying the relationship between the molecular topology and biological activity of compounds. In this study, two vancomycin units were linked at three distinct positions of the glycopeptide (vancosamine residue (V), C terminus (C), and N terminus (N)) to form two types of novel vancomycin cyclic dimers. Active NC‐VV‐linked dimers with a stable conformation as indicated by molecular mechanics calculations selectively suppressed the peptidoglycan polymerization reaction of vancomycin‐resistant Staphylococcus aureus in vitro. In addition, double‐disk diffusion tests indicated that the antibacterial activity of these dimers against vancomycin‐resistant enterococci might arise from the inhibition of enzymes responsible for peptidoglycan polymerization. These findings provide a new insight into the biological targets of vancomycin dimers and the conformational requirements for efficient antibacterial activity against vancomycin‐resistant strains.  相似文献

We designed bisnitroxide compounds where the radical sites are located close to each other in a molecule. Two new pincer-type bisnitroxide compounds have been synthesized, involving xanthene-4,5-diyl as a spacer and tert-butyl phenyl nitroxides as arms. From the X-ray crystal structure analysis, the shortest intramolecular interatomic N?O and O?O distances respectively are 5.074(6) and 5.258(6) Å for the m,m′-derivative and 3.624(3) and 3.771(3) Å for the p,p′-derivative. The N?O distance in the latter satisfies the empirical criterion for possible dimerization/degradation reaction accompanied by dia-/paramagnetic transition. However, the magnetic study clarified paramagnetic behavior in all the temperature range. According to a singlet-triplet model, antiferromagnetic couplings were characterized with 2J/kB = ?7.71(2) and ?8.83(4) K for the m,m′ and p,p′-derivatives, respectively. The present result suggests that a more flexible spacer is required for realization of possible dia-/paramagnetic transition.  相似文献
