Use of ultra-performance liquid chromatography/time-of-flight mass spectrometry with nozzle-skimmer fragmentation for comprehensive quantitative analysis of secondary metabolites in Arabidopsis thaliana

This study sought to develop techniques for LC/MS-based metabolomics and to verify that an MS/MS spectral tag (MS2T) could be used in practical secondary metabolite profiling. The retention time (RT), precursor ions, and fragment ions generated by nozzle-skimmer fragmentation were determined using ultra-performance liquid chromatography/time-of-flight mass spectrometry (UPLC/TOF-MS) and compared with the MS2T. A standard mix was analyzed with UPLC/TOF-MS under the same conditions as were used to construct the MS2T. The difference in RT for the standards was less than 0.15 min and the average RSD was about 2.8%, suggesting that the analysis was highly repeatable. Both precursor ions and fragment ions were observed when the cone voltage was 75 V. Experimental data and fragmentation pattern in the MS2T annotation list were highly similar. Wild-type and cas-1 mutant Arabidopsis thaliana samples treated with an elicitor were analyzed using UPLC/TOF-MS. Sixty-five peaks were successfully annotated. Fragment ions were observed with nozzle-skimmer fragmentation in 50 of 65 (77%) peaks. The reliability of annotation may have increased as a result of fragment ions. Results of multivariate analysis suggested that cas-1 was related to induction of the biosynthesis of these flavonoids. The devised method facilitated practical secondary metabolite profiling.     

Specific counterion effect on the adsorbed film of cationic surfactant mixtures at the air/water interface

To investigate the counterion effects, we employed dodecyltrimethylammonium bromide (DTABr)-dodecyltrimethylammonium tetrafluoroborate (DTABF(4)) mixed aqueous solutions and measured their surface tensions, then analyzed these data in a thermodynamic treatment. The tensiometry showed that DTABF(4) was more effective in lowering the surface tension of water. The phase diagram of adsorption demonstrated that the surface was enriched with BF(4)(-) ions, but the composition of Br(-) ions in the adsorbed film was slightly enhanced compared to the ideal mixing criteria. These were explained in terms of the size and polarizability of counterions. Moreover, the distribution of counterions of the DTABr-DTABF(4) mixtures in the adsorbed film is greatly different from that of the 1-hexyl-3-methylimidazolium bromide (HMIMBr)-1-hexyl-3-methylimidazolium tetrafluoroborate (HMIMBF(4)) mixtures, where a stronger hydrogen-bonding exists between BF(4)(-) and HMIM(+) ions. These findings suggest that the adsorption of counterions in electric double layers is likely subject to two factors: the nature of counterion and their interactions with surfactant ions.     

Metal-catalysed cleavage of carbon-carbon bonds

Reactions cleaving carbon-carbon bonds with the assistance of transition metals as the catalyst have provided various molecular transformations that are otherwise difficult to execute, opening a scenic avenue along organic synthesis. Construction of structural motifs like medium-sized carbocycles and chiral quaternary carbon centres have been set within an access of such paradoxical approaches in the past decade.     

Asymmetric Colliding-Pulse Mode-Locking in InGaAsP Semiconductor Lasers

Asymmetric colliding-pulse mode-locking was successfully demonstrated in an InGaAsP laser. This laser had a single saturable absorber section located partway along the cavity length, and showed mode-locking operation at harmonics of the cavity roundtrip frequency. The method verified that optical pulse repetition up to subterahertz rates can be generated with nearly 1-mm-long mode-locked InGaAsP lasers.     

Iridium-catalyzed enantioselective Pauson-Khand-type reaction of 1,6-enynes

Iridium-chiral diphosphine complex catalyzes an enantioselective intramolecular Pauson-Khand-type reaction to give various chiral bicyclic cyclopentenones. The enantioselective reaction proceeds more smoothly and enantioselectively under a lower partial pressure of carbon monoxide. Moreover, aldehyde can be used as a CO source in the enantioselective carbonylative coupling.     

Synthesis of 3‐Acyl‐4‐alkenylpyrrolidines by Platinum‐Catalyzed Hydrative Cyclization of Allenynes

A series of nitrogen‐tethered allenynes (‘5‐aza‐1,2‐dien‐7‐ynes’) 1 were transformed to the corresponding 3‐acyl‐4‐alkenylpyrrolidines 3 when treated with a catalytic amount of PtCl₂ in MeOH at 70°. Initial Pt‐promoted cyclization forms a nonclassical carbocationic intermediate. In contrast to the cycloisomerization in toluene, which produced the bicyclic cyclobutenes 2 , the intermediate is intercepted by addition of an oxygen nucleophile to achieve the formal hydrative cyclization.     

Miscibilities of methanol and ethanol with dodecylammonium chloride in the adsorbed film and micelle

The surface tension of the aqueous solutions of methanol– dodecylammonium chloride (DAC) and ethanol–DAC mixtures has been measured as a function of the total molality of the mixture m^ and the mole fraction of DAC X^₂ in the mixture at 298.15?K under atmospheric pressure. The compositions of the adsorbed film at 50, 40, and 30?mN?m^-1, and that of the micelle at the critical micelle concentration (CMC) have been evaluated by applying the thermodynamic equations derived previously and shown in the form of phase diagrams of adsorption and micelle formation. It has been found that (i) methanol is hardly miscible in M28.8nthe adsorbed film and micelle, and (ii) ethanol molecules are very slightly incorporated into adsorbed film of DAC at high-surface tension and into the micelle, although hardly miscible in the adsorbed film at low-surface tension. By comparing the corresponding phase diagrams of ethanol-2- (octylsulfinyl)ethanol (OSE) mixture, furthermore, it has been shown that there exists a difference in the miscibility of ethanol between DAC and OSE.
