Synthesis,surface accumulation,and micellar properties of amphiphilic block copolymers

Amphiphilic block copolymers, i.e., poly(methyl methacrylate)-b-poly(2-dimethylethylammoniumethyl methacrylate), were synthesized by the reaction between two prepolymers. Carboxyl-terminated poly(methyl methacrylate) and hydroxyl-terminated poly(2-dimethylaminoethyl methacrylate) were prepared by radical polymerization of the corresponding monomers in the presence of thioglycolic acid and 2-mercaptoethanol as a chain transfer agent, respectively. Two condensation methods, i.e., DCC and the acid chloride method, were used for the reactions of these prepolymers. The subsequent quarternization produced the amphiphilic block copolymers. Surface property of poly(methyl methacrylate) films containing this amphiphilic block copolymer was examined by measuring contact angles for water. The addition of only 0.5 wt% of the block copolymer was sufficient to make poly(methyl methacrylate) surfaces hydrophilic. The block copolymer formed a polymeric micelle in acetone–water mixed solvent.     

Phase-transfer catalytic activity of soluble polystyrenes containing crown ether moieties and hydroxyl groups adjacent to the macrorings

Soluble polystyrenes with crown ether structures and hydroxyl groups adjacent to the macrorings were prepared by the reaction of epoxide-containing polystyrenes with monoaza-15-crown-5 or monoaza-18-crown-6. Rate of formation of the polymer-bound alkali cation-crown alkoxide ion pair from the soluble polystyrenes and aqueous NaOH or KOH depended on the loading of crown ether and hydroxyl units and on the size of the macroring. The elimination of HCl from less reactive 2-chloroethylbenzene with aqueous NaOH or KOH in the presence of the soluble polystyrene catalysts under two-phase conditions was limited mainly by the intrinsic reaction rate. The elimination of HBr from more reactive 2-bromoethylbenzene in the presence of the less (11%) ring-substituted polymer catalyst with 18-crown unit was limited by the alkoxide formation rate. © 1994 John Wiley & Sons, Inc.     

Cu-Chelex as a selective adsorbent for o-nitroso derivatives of phenolic substances

Summary Chelex resins in several metal forms were tested for selective uptake of o-nitrosonaphthols and o-nitrosophenols from aqueous solution. Cu-Chelex was the most efficient resin over a wide pH range, and the taken-up nitroso compounds were effectively eluted with concentrated ammonia: acetonitrile (4:1, v/v) after washing the resin with acetonitrile to eliminate non-specific binding. The upper and lower limits of uptake capacity of the resin for 1-nitroso-2-naphthol were 0.5 mol/100 mg and 1.25 mol/100 mg, respectively. A preliminary recovery experiment using urine and cucumber juice showed that Cu-Chelex would be useful for screening of o-nitroso derivatives of phenolic substances in complicated samples such as biological fluids and foods.     

Structures of TMC-95A-D: novel proteasome inhibitors from Apiospora montagnei sacc. TC 1093

Four novel proteasome inhibitors, TMC-95A-D (1-4) have been isolated from the fermentation broth of Apiospora montagnei Sacc. TC 1093, isolated from a soil sample. All of the molecular formulas of 1-4 were established as C(33)H(38)N(6)O(10) by high-resolution FAB-MS. Their planar structures were determined on the basis of extensive analyses of 1D and 2D NMR, and degradation studies. Compounds 1-4 have the same planar structures to each other, and are unique highly modified cyclic peptides containing L-tyrosine, L-aspargine, highly oxidized L-tryptophan, (Z)-1-propenylamine, and 3-methyl-2-oxopentanoic acid units. The absolute configuration at C-11 and C-36 of 1-4 was determined based on chiral TLC and HPLC analyses of their chemical degradation products. The ROESY analysis along with (1)H-(1)H coupling constants clarified the absolute stereochemistry at C-6, -7, -8, and -14 of the cyclic moieties. These studies revealed the relationships of 1-4 to be diastereomers at C-7 and C-36.     

Comparative pharmacokinetics of 67Ga among the different routes of injection

The pharmacokinetic study of 67Ga-citrate (67Ga) following intravenous (i.v.), subcutaneous (s.c.) and intraperitoneal (i.p.) injection was performed in anesthetized rats using the repeated blood sampling method by cannulation technique into the external jugular vein. The disappearance of 67Ga from the blood following i.v. and s.c. injection was best fit a three-exponential equation. There was no significant difference between the areas under the curves following i.v. and s.c. injection of 67Ga. In the case of i.p. injection, the disappearance of 67Ga from the blood was described by a two-exponential equation. However, the maximum blood radioactivity was very low, and the disappearance rate of 67Ga from the blood was extremely slow compared to the other routes of injection. The conclusion from these results was that s.c. injection was as suitable as i.v. injection, but i.p. injection was not appropriate for the distribution study of 67Ga such as scintigraphy or autoradiography. However, i.p. route may be available for a special experiment which needs the long-time retention of 67Ga in the blood.     

Chemical release control: Carbamates of 3-vinylphenyl and 2-methacryloyloxyethyl isocyanates and perfume and herbicide alcohols

Polymerizable carbamates were synthesized from 3-vinylphenyl and 2-methacryloyloxyethyl isocyanates and perfume and herbicide alcohols, such as 2-phenethyl alcohol, citronellol, geraniol, 1-menthol, borneol, and 2-(2,4-dichlorophenoxy)- and 2-(2,4,5-trichlorophenoxy)ethyl alcohols. Copolymerization of these carbamate monomers and N-vinyl-2-pyrrolidone with AIBN in dioxane gave respective copolymers. Hydrolyses of both monomers and copolymers, however, required severe acid conditions, although different chemical structures gave different hydrolytic behaviors.     

A lithiation approach to cordycepin analogues variously substituted at the C-8 position

Several 8-substituted cordycepins were prepared via LDA lithiation of 2′,5′-bis-O-(t-butyldimethylsilyl)-cordycepin and successive reactions of its C-8 lithiated species with various types of electrophiles. Wittig reaction of the 8-formyl derivative was also examined.

Concomitant increase in putrescine incorporation with transferrin uptake into rat reticulocytes was found. [14C]Putrescine incorporation occurred in the presence of transferrin at 37 degrees C. The subcellular distribution of incorporated [14C]putrescine showed that the incorporation in the plasma membrane time dependently increased. Both 125I-labeled transferrin uptake and [14C]putrescine incorporation decreased when ethylene glycol bis(2-aminoethyl ether)-N,N,N',N'-tetraacetic acid was added, while it increased with the addition of A23187.
