TUMOR-LOCALIZING FLUORESCENT DIAGNOSTIC AGENTS WITHOUT PHOTOTOXICITY

In order to develop tumor-localizing fluorescent diagnostic agents without phototoxicity, various heterodimers linked by some spacers between a chlorine derivative and its Mn or Cu complex were synthesized. The representative agent of them was named HAT-D01 and has a molecular formula of m -phthalyl-{[13,17-bispropanoic acid-3-ethenyl-8-formylethylidene-7-hydroxy-2,7,12,18-tetramethyl-porphyrinate]-manganese (III)}-[3'-ethenyl-8'-formylethylidene-7'-hydroxy-2',7', 12', 18'-tetramethyl-porphine-13', 17'-bispropanoyl aspartic acid]-bishydrazone.     

Determination of cadmium in river water by sequential metal vapor elution analysis

Determination of cadmium in river water by sequential metal vapour elution analysis (column temperature; > 1500 K) with argon and hydrogen carrier gas and with atomic absorption spectrometric detection is described. The column is made of a molybdenum capillary tube (i.d. 1.22 mm) and the temperature is 1760 K. The cadmium vapor was separated from those of calcium, iron and sodium. The calibration graph was linear up to 15 μ/ml. Relative standard deviations of 0.8–4.3% were obtained in the range 1 to 15 μ/ml. Cadmium in spiked samples (river water) was determined. The results were in good agreement with the amount spiked.     

Stereoselective synthesis of procyanidin B3-3-O-gallate and 3,3″-di-O-gallate, and their abilities as antioxidant and DNA polymerase inhibitor

A simple method for the synthesis of procyanidin B3 substituted with a galloyl group at the 3 and 3″ position is described. Condensation of a benzylated catechin-3-O-gallate electrophile with a nucleophile, catechin and catechin-3-O-gallate, proceeded smoothly and stereoselectively to afford the corresponding dimer gallates, procyanidin B3-3-O-gallate and procyanidin B3-3,3″-di-O-gallate, in good yields. Further, their antioxidant activities on UV-induced lipid peroxide formation, DPPH radical scavenging activity and inhibitory activity of DNA polymerase were also investigated. Among three procyanidin B3 congeners (procyanidin B3, 3-O-gallate and 3,3″-di-O-gallate), the 3,3″-di-O-gallate derivative showed the strongest antioxidant and radical scavenging activity. Interestingly, the 3-O-gallate derivative was the strongest inhibitor of mammalian DNA polymerase with IC₅₀ value of 0.26 μM, although it showed the weakest antioxidant and radical scavenging activity. It became apparent that the presence of a galloyl group at the C-3 position in the proanthocyanidin oligomer was very important for biological activity, however, the antioxidant activity of these compounds was not parallel to the DNA polymerase inhibitory activity.     

Novel ring transformation reactions of xanthine derivatives

The novel ring transformation reactions were found in the reactions of 1,3,7,9-tetra-alkyl-8,9-dihydroxanthines and acetylenic compounds. The reaction of the dihydroxanthine with DMAD gave a propellane type compound and with methyl propiolate afforded the similar type compound and a pyrimido[4,5-b]diazepine derivative. The mechanism of these reactions was also discussed.     

Cyclic dithio-carbenium ion salts and their conversion to ketene dithioacetals

1,2-Ethanedithiol and 1,3-propanedithiol were treated with perchloric acid in acetic anhydride to give 2-methyl-1,3-dithiolan-2-yl and 2-methyl-1,3-dithian-2-yl perchlorates, respectively, which were converted to ketene dithioacetals by deprotonation with tertiary amines.     

A highly regio- and stereoselective synthesis of internal olefins via an elimination of trimethylstannyl group

Internal reaction of secondary alkyltin(IV) compounds having thionium ion gave internal trans olefins with high yield and regio- and stereoselectivity via 1,5- or 1,6-transfer of a hydride β to the trialkylstannyl group.     

Large-scale separation of metallic and semiconducting single-walled carbon nanotubes

In the applications of single-walled carbon nanotubes (SWNTs), it is extremely important to separate semiconducting and metallic SWNTs. Although several methods have been reported for the separation, only low yields have been achieved at great expense. We show a separation method involving a dispersion-centrifugation process in a tetrahydrofuran solution of amine, which makes metallic SWNTs highly concentrated to 87% in a simple way.     

A lithiation approach to cordycepin analogues variously substituted at the C-8 position

Several 8-substituted cordycepins were prepared via LDA lithiation of 2′,5′-bis-O-(t-butyldimethylsilyl)-cordycepin and successive reactions of its C-8 lithiated species with various types of electrophiles. Wittig reaction of the 8-formyl derivative was also examined.
