In order to quantify 1,3-dinitropyrene (DNP), 1,6-DNP and 1,8-DNP in soil, we developed an efficient clean-up procedure and a sensitive determination method using fluorescence detection. DNP isomers were efficiently cleaned by three stages of fractionation, i.e., a silica gel open column chromatography using stepwise elution and two further purification steps by high-performance liquid chromatography (PPLC) using a monomeric-type octadecylsilyl (ODS) column and a polymeric-type ODS column. The recoveries of DNPs during the whole clean-up process were 94% or more. The fraction corresponding to DNPs was injected into an analytical polymeric-type ODS column for HPLC to separate DNP isomers. The effluent from the analytical ODS column was directly introduced to a catalyst column, which was packed with 5 microns alumina coated with platinum and rhodium (Pt-Rh), in order to reduce DNPs to diamino compounds, and then the fluorescence of diaminopyrenes was detected. The immediate detection of diaminopyrene isomers after on-line reduction afforded a sensitive detection of DNP isomers. The detection limits for DNPs were in the range of 0.7 to 4 pg. These developed methods were applied to four soil samples collected at parks in residential areas.  相似文献

The static and dynamical local spin susceptibility of the nonstoichiometric organic conductor, κ-(BEDT-TTF)4Hg2.78Cl8, is studied by 13C NMR spectroscopy from room temperature down to 1.7 K. We observe a gradual growth of inhomogeneity in the spin state as a spectral broadening down to the lowest temperature, which is sustained even under 0.4 GPa but suppressed under 0.8 GPa. The emergence of the inhomogeneity is discussed in the light of the doped triangular-lattice Mott insulator.  相似文献

The Aconitum species (Ranunculaceae) are widely distributed in northern Asia and North America. Their roots are popularly used in herbal medicines in China and Japan. Many cases of accidental, suicidal and homicidal intoxication with this plant have been reported; some of these were fatal because the toxicity of Aconitum is very high. It is thus important to detect and quantify Aconitum alkaloids in body fluids, with high sensitivity. We have developed a simple and sensitive method for measuring four kinds of Aconitum alkaloids (aconitine, hypaconitine, jesaconitine and mesaconitine) by LC/electrospray (ESI)-time-of-flight (TOF)-MS. For all of them, only molecular ions were observed at an orifice voltage of 75 V; at 135 V, base peaks corresponding to [M - 60 + H]+ ions were observed. These four compounds and methyllycaconitine (internal standard) in human plasma samples were purified by solid-phase extraction. The four extracted compounds were completely separated in mass chromatograms; the calibration curves showed good linearity in the range 10-300 ng/ml, and the detection limits were estimated to be 0.2-0.5 ng/ml. Using our method, we also determined the amounts of these compounds in tuber samples. The present method is applicable in clinical and forensic toxicology.  相似文献

The inactivation effects of ultrasonic irradiation at 27.5 kHz and chlorination using sodium hypochlorite solution (NaOCl) on the growth of Saccharomyces cerevisiae (yeast cells) were investigated. In order to evaluate the effect of ultrasound on the growth of the yeast cells, calorimetric analysis was carried out in addition to colony counting. The heat evolution produced by the growth of yeast cells detected by calorimetry showed completely different patterns between sonication and chlorination. In case of sonication, the yeast cells were inactivated almost like a bactericidal effect, i.e. a quantitative change in cell number, at the beginning of sonication. It was similar to patterns obtained on simple dilution of yeast cells. In contrast, longer sonication increased the bacteriostatic effect, i.e. qualitative damage of the cell growth activity, together with the bactericidal effect. These results suggest that the cavitation caused by ultrasonic irradiation initially disrupted the cells located near the cavitation bubble which caused immediate cell death and the growth activity of the surviving cells was gradually damaged by further sonication. On the other hand, only a bacteriostatic effect was observed when the yeast cells were inactivated by chlorination.  相似文献

The Double Chooz experiment presents an indication of reactor electron antineutrino disappearance consistent with neutrino oscillations. An observed-to-predicted ratio of events of 0.944±0.016(stat)±0.040(syst) was obtained in 101 days of running at the Chooz nuclear power plant in France, with two 4.25 GW(th) reactors. The results were obtained from a single 10 m(3) fiducial volume detector located 1050 m from the two reactor cores. The reactor antineutrino flux prediction used the Bugey4 flux measurement after correction for differences in core composition. The deficit can be interpreted as an indication of a nonzero value of the still unmeasured neutrino mixing parameter sin(2)2θ(13). Analyzing both the rate of the prompt positrons and their energy spectrum, we find sin(2)2θ(13)=0.086±0.041(stat)±0.030(syst), or, at 90% C.L., 0.017相似文献

During evaporation, shape changes of nanoliter-scale (80-100 nL) water droplets were evaluated on two superhydrophobic surfaces with different random roughness (nm-coating, μm-coating). The square of the contact radius and the square of the droplet height decreased linearly with evaporation time. However, trend changes were observed at around 170 s (nm-coating) and around 150 s (μm-coating) suggesting a wetting mode transition. The calculated droplet radii for the wetting mode transition from the average roughness distance and the average roughness height of these surface structures were approximately equal to the experimental values at these trend changes. A certain level of correlation between the roughness size and droplet radius at the wetting mode transition was confirmed on surfaces with random roughness.  相似文献

Sel-P is considered to be the most important selenoprotein for evaluating the selenium (Se) status in the body. To isolate and determine Sel-P in plasma, we have developed an analytical method combining heparin affinity (AF) and size-exclusion column (SEC) high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICPMS). We used this method to validate the adsorption efficiency of selenoproteins on a heparin AF column, and to then determine the Se concentrations incorporated in proteins in human and mouse plasma. The adsorption efficiency of Sel-P on a heparin column was more than 90% for both human and mouse plasma. Tandem AF and SEC separation proved to be useful for determining the Se concentrations incorporated in Sel-P in mouse plasma, but not in human plasma, because of nonspecific adsorption of plasma-extracellular glutathione peroxidase (eGPx) and albumin on the heparin AF column. Ultimately, we used the tandem AF and SEC separation method for mouse plasma and SEC separation alone for human plasma. The Se concentration incorporated in selenoproteins determined by our method showed good agreements with the total Se concentration determined following acid digestion.  相似文献

To apply an irradiation technique to sterilize “Hybrid” biomedical materials including enzymes, we selected papain, a well-characterized plant endopeptidase as a model to examine durability of enzyme activity under the practical irradiation condition in which limited data were available for irradiation inactivation of enzymes. Dry powder and frozen aqueous solution of papain showed significant durability against 60Co-gamma irradiation suggesting that, the commercial irradiation sterilizing method is applicable without modification. Although irradiation of unfrozen aqueous papain solution showed an unusual change of the enzymatic activity with the increasing doses, and was totally inactivated at 15 kGy, we managed to keep the residual activity more than 50% of initial activity after 30-kGy irradiation, taking such optimum conditions as increasing enzyme concentration from 10 to 100 mg/ml and purging with N2 gas to suppress the formation of free radicals.  相似文献

Spores of Bacillus subtilis, containing plasmid (B. subtilis BD 366(pUB110, Trp C2 thr 5)), were prepared free of additives and the effect of water on their radiation sensitivity under aerobic condition was investigated. The moisture content of spores was controlled by (i) drying over CaCl2 (Aw = 0.0), (ii) equilibrating with the vapor of saturated aqueous NaCl solution (Aw = 0.7), or (iii) suspending in water (Aw = 1.0). The resistance of spores to radiation increased with increasing moisture content. The analysis of spore DNA by agarose gel electrophoresis showed that DNA damage (strand breaks) decreased with increasing water activity of the spores, a finding concomitant with the observed survival rates.  相似文献
