The potential use of a fluorescent tag system based on 7-(1H-1,2,3-triazol-4-yl)coumarin fluorophore having a fluorous moiety and a polyethylene glycol (PEG) spacer at opposite ends as a tool for a stepwise and comparative evaluation of the fabrication process of small molecule microarrays was illustrated by the qualitative analysis of the results of the fluorescence detection obtained from the microarray experiments using the tagged biotins and streptavidin-Cy3 (and avidin-Cy5) as the binding partners. 相似文献

ADMET (absorption, distribution, metabolism, excretion, and toxicity)‐related failure of drug candidates is a major issue for the pharmaceutical industry today. Prediction of PD‐PK‐T properties using in silico tools has become very important in pharmaceutical research to reduce cost and enhance efficiency. PaDEL‐DDPredictor is an in silico tool for rapid prediction of PD‐PK‐T properties of compounds from their chemical structures. It is free and open‐source software that, has both graphical user interface and command line interface, can work on all major platforms (Windows, Linux, and MacOS) and supports more than 90 different molecular file formats. By virtue of the electron‐donating and electron‐withdrawing properties of the thiazole ring, a new soluble rigid‐rod organometallic polymer containing electron‐donating and electron‐withdrawing trans‐[‐Pt‐(PBu3)2‐C≡C—R—C≡C—]n (R = bithiazolediyl) groups is prepared by CuI‐catalyzed dehydrohalogenation. The thermal properties and the optical absorption and photoluminescence spectra of the polymer are reported. The polymer is luminescent with a singlet emission peak at 539 nm and photoconducting in a single‐layer sandwich structure photocell. The optical gap of the polymer is reduced compared to that for the oligothienyl analogue. 相似文献

A series of rhenium containing polyimides was prepared by the polymerization between a tricarbonylrhenium diimine complex with various aromatic dianhydrides. Electronic absorption spectra of the polymers showed a very strong intramolecular charge transfer transition which obscures the transfer transition band due to the rhenium complex in the visible region. Dramatically shortened transverse relaxation times in transverse relaxation optimized spectroscopy (TROSY) result from interference between dipole–dipole interactions and the anisotropy of the chemical shift. Thus NMR spectroscopy becomes a suitable method for studying large biomolecules, with optimal performance when 1-GHz spectrometers become available. By using new phase cycles and data-processing methods, the sensitivity of the TROSY experiment was increased by a factor of √2, which is of considerable importance for applications in high-field NMR studies on large proteins. 相似文献

The present study focuses on the possible involvement of l-arginine-nitric oxide-cGMP-ATP-sensitive K+ channel pathway in the antinociceptive activity of a novel diarylpentanoid analogue, 2-benzoyl-6-(3-bromo-4-hydroxybenzylidene)cyclohexen-1-ol (BBHC) via a chemical nociceptive model in mice. The antinociceptive action of BBHC (1 mg/kg, i.p.) was attenuated by the intraperitoneal pre-treatment of l-arginine (a nitric oxide synthase precursor) and glibenclamide (an ATP-sensitive K+ channel blocker) in acetic acid-induced abdominal constriction tests. Interestingly, BBHC’s antinociception was significantly enhanced by the i.p. pre-treatment of 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (ODQ), a selective inhibitor of soluble guanylyl cyclase (p < 0.05). Altogether, these findings suggest that the systemic administration of BBHC is able to establish a significant antinociceptive effect in a mice model of chemically induced pain. BBHC’s antinociception is shown to be mediated by the involvement of l-arginine-nitric oxide-cGMP-ATP-sensitive K+ channel pathway, without any potential sedative or muscle relaxant concerns. 相似文献

We report a simple method for converting solid matrices into useful matrix solutions for matrix-assisted laser desorption/ionization (MALDI). This method is based on the dissolution of the solid matrix in a liquid support of low volatility such as glycerol. An appropriate solubilizing reagent was added to promote the dissolution of the matrix materials into the liquid support. Selection of the solubilizing reagent is empirically related to an acid-base relationship, i.e., an acidic solid matrix requires a basic organic compound to form a stable matrix solution in the liquid support and vice versa. A tenfold increase in the solubility can be obtained for many solid matrices when appropriate solubilizing reagents are added into the glycerol support. This solubility enhancement is tentatively attributed to the ion-pair formation in a polar nonvolatile liquid support. In addition, the hydrophobicity of the solid matrix seems to play an important role in the efficiency of the resulting matrix solution. By using glycerol as liquid support, a hydrophilic matrix, such as 2,5-dihydroxybenzoic acid (DHB), showed a substantial “peripheral effect,” in which good analyte ion signals could only be recorded at the peripheral region of the sample droplet. More hydrophobic matrices, such as α-cyano-4-hydroxycinnamic acid (α-CCA), exhibit better and more homogeneous responses at different regions of the droplets. The performance of these matrix solutions was evaluated in terms of the durability, reproducibility, sensitivity, high mass capability, and generality. A typical sample droplet can afford more than an hour of repeated sampling with excellent shot-to-shot reproducibility. A low picomole sensitivity was demonstrated using a luteinizing hormone releasing hormone (LHRH) in a Fourier transform ion cyclotron resonance mass spectrometer with a homemade external MALDI ion source. By using a commercial MALDI time-of-flight mass spectrometer, proteins with masses as high as 66,000 Da were successfully analyzed by using these matrix solutions. 相似文献

Preparation of highly uniform Ag/TiO2 and Au/TiO2 supported nanoparticle catalysts by photodeposition 总被引:1,自引:0,他引:1
Photodeposition of Ag nanoparticles on commercial TiO2 particles and nanoparticles was performed in order to provide direct visualization of the spatial distribution of photoactive sites on sub-micrometer-scale and nanoscale TiO2 particle surfaces and to create materials for potential catalytic applications. HRTEM (high-resolution transmission electron microscopy) and HAADF-STEM (high-angle annular dark-field scanning transmission electron microscopy) were used to characterize these materials. The size and spatial distributions of the Ag nanoparticles on the commercial TiO2 were not uniform; the concentration of Ag was higher on grain boundaries and at the edges of these submicrometer particles. In the case of TiO2 nanoparticles, the size distribution of the Ag nanoparticles deposited was relatively uniform and independent of irradiation time and photon energy. The amount of Ag deposited on TiO2 nanoparticles was at least 6 times higher than that on the commercial samples for comparable irradiation conditions. Compared to the case of Ag photodeposition, the difference in the amount of Au photodeposited on TiO2 particles and nanoparticles was even greater, especially at low precursor concentrations. Photodeposition on TiO2 nanoparticles is suggested as a potential method for the preparation of Au/TiO2 catalysts, as loadings in excess of 10 wt % of uniform 1 nm metal particles were achieved in this work. 相似文献
