2-DE separations of protein extracts sometimes have problems with poor resolution and streaking. This problem is particularly apparent with microorganisms, most notably those with a large cell wall. Here we describe a novel, rapid protocol for the extraction of microorganisms in acidic conditions, leading to increased resolution and 2-D gel quality. The efficiency of the protocol is demonstrated with extracts of bacteria, Escherichia coli and Bacillus subtilis; fungus, Trichoderma harzianum and yeast, Saccharomyces cerevisiae. We also demonstrate using a membrane centrifugal filtration, that large acidic molecules in excess of 100 kDa, probably including cell wall material, are responsible for the separation difficulties. A range of acidic extraction conditions were investigated, and it was found that optimal extraction is achieved using an extraction solution acidified to pH 3 by 80 mM citric acid. These findings have significant implications for the proteomic study of many medically, agriculturally and environmentally significant microorganisms, as the cell walls of these organisms are often considerably more complex than many commonly studied laboratory strains.  相似文献

Reaction of lithiated bis(diphenylphosphino)amine, [(C6H5)2P]2N? Li+, with K2PtCl4 or PdCl2 (in the presence of trimethylphosphine) yields the homoleptic bis[bis(diphenylphosphino)amide] complexes I and II, respectively. With NiCl2/(CH3)3P the chloro-bridged dinuclear complex III is obtained. A symmetrical bonding of the Ph2P-·N-·PPh2 anion to the metal through the phosphorus atoms is indicated for these diamagnetic, deep-yellow (I, II) or red (III) complexes by 31P NMR spectroscopy (J(PtP) 1812 Hz for I). I and II are dissolved in CF3COOH with protonation at the nitrogen atoms to give bis(diphenylphosphino)amine complexes IV and V, respectively (J(PtP) 2080 Hz for IV). IV and V are 1:2 electrolytes in CH3NO2. Methylation of I-III with CH3OSO2F leads to the bis(diphenylphos-phino)methylamine complexes VI-VIII, of which the palladium compound VII has been structurally characterized by single crystal X-ray diffraction. VII contains a planar CNP2PdP2NC skeleton and is thus based on planar ligand arrays both at Pd and at the two N atoms.  相似文献

Fluorescent amphiphilic benzoxazole derivatives were synthesized and used to produce photoactive phosphatidylcholine (PC) liposomes by reserve-phase evaporation. The dyes absorbed in the UV region and were fluorescent in the blue-green region (determined by solvent polarity). The alkyl chain length seemed to play a fundamental role in the photophysics of the benzoxazole fluorophore in reverse liposomes, and despite the same ESIPT core and phospholipid building block, each amphiphilic dye had a particular emission profile related to the dye location in the liposome. The fluorescence emission spectra from dye 5 showed that its fluorophore experienced a polar environment, due to the single normal emission, while dyes 6–7 had (in part) a normal emission, and the main fluorescent band ascribed to the ESIPT emission indicated a more hydrophobic environment. Despite the complex fluorescent profiles, the benzoxazole derivatives could be successfully introduced into the reverse liposome structure due to the interaction between the alkyl chain and PC bilayer.  相似文献

The present article deals with a variational formulation of a floating body in a perfect fluid. Since the considered energy functional depends on both stream function and floating body, these two minimizations are realized in two separated steps. By the use of free boundary methods, existence and Lipschitz continuity of the minimizing stream function are proved. In addition, it is shown that the corresponding free surface has Hölder boundary. The dependence on the floating body leads to an optimal shape problem which is treated by the use of Hausdorff convergence. Under the postulate of a bounded density parameter of the domain, the existence of an optimal floating body is shown.  相似文献

Deprotonation (NaH) of the phosphine ligand in 2a, with subsequent addition of [IrCl(CO)(PPh 3) 2] and Tl[PF 6], yields the neutral, zwitterionic complex [6,6,6-(CO) 3-4,7-mu-{Ir(H)(CO)(PPh 3) 2PPh 2}- closo-6,1-FeCB 8H 7] ( 3), which contains a B-P-Ir- B ring. Alternatively, deprotonation using NEt 3, followed by addition of HC[triple bond]CCH 2Br, affords [6,6,6-(CO) 3-7-(PPh 2CCMe)- closo-6,1-FeCB 8H 8] ( 4). Addition of [Co 2(CO) 8] to CH 2Cl 2 solutions of the latter gives [6,6,6-(CO) 3-7-(PPh 2-{(mu-eta (2):eta (2)-CCMe)Co 2(CO) 6})- closo-6,1-FeCB 8H 8] ( 5), which contains a {C 2Co 2} tetrahedron. In the absence of added substrates, deprotonation of the PHPh 2 group in compounds 2, followed by reaction of the resulting anions with CH 2Cl 2 solvent, affords [6,6,6-(CO) 3- n-(PPh 2CH 2Cl)- closo-6,1-FeCB 8H 8] [ n = 7 ( 6a) or 10 ( 6b)] plus [6,6-(CO) 2-6,7-mu-{PPh 2CH 2PPh 2}- closo-6,1-FeCB 8H 8] ( 7, formed from 2a), of which the latter species possesses an intramolecular B-P-C-P- Fe ring. Addition of Me 3NO to CH 2Cl 2 solutions of 2a causes loss of an Fe-bound CO ligand and formation of [6,6-(CO) 2-6,7-mu-{NMe 2CH 2PPh 2}- closo-6,1-FeCB 8H 8] ( 8), which incorporates a B-P-C-N- Fe ring. A similar reaction in the presence of ligands L yields [6,6-(CO) 2-6-L-7-(PPh 2CH 2Cl)- closo-6,1-FeCB 8H 8] [L = PEt 3 ( 9) or CNBu (t) ( 10)], in addition to 8.  相似文献

A sensitive and selective method for the simultaneous determination of a spectrum of trichothecenes in residuals of biogas production has been developed. It comprises sample clean-up by liquid/liquid partition for digested manure and solid phase extraction for digested solid phase. Quantification of A- and B-type trichothecenes as their trifluoroacetyl derivatives is performed by gas chromatography mass spectrometry (GC/MS), that of B-type trichothecenes alternatively by high-performance liquid chromatography (HPLC). Fluorescence detection (FLD) after post-column derivatisation using methylacetoacetate and ammonium acetate after alkaline decomposition of toxins was applied. Detection limits in digestates were between 1 and 30 microg/l and 20 and 50 microg/l for GC/MS and HPLC/FLD, respectively. Recovery rates were between 52 and 129% for GC/MS detection with the exception of T-2 tetraol with 22%, and between 56 and 123% for HPLC/FLD.  相似文献
