4-Fluorocyclohexa-2,5-dienones as new acceptors for the Hauser annulation

4-Fluorocyclohexa-2,5-dienones are introduced as new acceptors for the Hauser annulation. In cases where the corresponding methoxy analogs fail to undergo annulation, the former smoothly do so with phenylsulfonyl phthalides to furnish anthraquinones in good yields. Steric effects are implicated to explain the inertness of the methoxy analogs.     

Molecular docking studies on tetrahydroimidazo-[4,5,1-jk][1,4]-benzodiazepinone (TIBO) derivatives as HIV-1 NNRT inhibitors

At present, chemotherapy seems to be the main weapon in the arsenal of remedies for the ongoing crusade against AIDS. The mode of binding of the TIBO family of inhibitors has been of interest because these compounds do not fit the two-hinged-ring model as generally observed in the NNRTIs. Flexible docking simulations were performed with a series of 53 TIBO derivatives as NNRTIs. Binding preferences as well as the structural and energetic factors associated with them were studied. A good correlation (r ² = 0.849, q ² = 0.843) was observed between the biological activity and binding affinity of the compounds which suggest that the identified binding conformations of these inhibitors are reliable. Further screening of PubChem database yielded novel scaffolds. Our studies suggest that modifications to the TIBO group of inhibitors might enhance their binding efficacy and hence, potentially, their therapeutic utility.     

A facile method for the synthesis of steroidal and nonsteroidal 5-methyl pyrazolo[1,5-a]pyrimidines

The steroidal and nonsteroidal 5-methyl pyrazolo[1,5-a]pyrimidines were synthesized by the reaction of steroidal/nonsteroidal α,β-unsaturated ketones and 3-amino-1H-pyrazoles/5-amino-1H-pyrazoles in the presence of KO^tBu in ethanol under reflux condition.     

Regioselective synthesis of 1-hydroxycarbazoles via anionic [4+2] cycloaddition of furoindolones: a short synthesis of murrayafoline-A

Suitably N-protected furoindolones react regioselectively with a variety of Michael acceptors in the presence of LDA to give 1-hydroxycarbazoles in a single-pot process and in good yields. The methodology has been applied to the synthesis of murrayafoline-A.     

Asymptotic properties of the least squares estimators of the parameters of the chirp signals

Chirp signals are quite common in different areas of science and engineering. In this paper we consider the asymptotic properties of the least squares estimators of the parameters of the chirp signals. We obtain the consistency property of the least squares estimators and also obtain the asymptotic distribution under the assumptions that the errors are independent and identically distributed. We also consider the generalized chirp signals and obtain the asymptotic properties of the least squares estimators of the unknown parameters. Finally we perform some simulations experiments to see how the asymptotic results behave for small sample and the performances are quite satisfactory.     

Review of the aldol reaction

Aldol condensation is an important synthetic method widely used in organic synthesis. Development of catalytic methods that avoids the production of stoichiometric by-products while maintaining high levels of control available from stoichiometric processes provides an atom-economical alternative for these important transformations. Indeed, numerous catalysts for the aldol reaction have been reported in recent years, including enzymes, catalytic antibodies, organometals, organocatalysts, and small molecules. The direct aldol reaction is the most important reaction employed by synthetic chemists and is common in nature. Recently, various Lewis acids have been examined as catalysts for aldol reactions, but aldol condensation in a micellar medium has not been studied in detail so far. Because of stronger environmental concerns, organic reactions in green media, especially in water, have attracted more attention. It is believed that micelles act as nano reactors to enhance the reaction rates and give very good to excellent yields of end products.     

Stereodivergent Zweifel Olefination and its Mechanistic Dichotomy

Stereoselective Zweifel olefination using boronate complexes carrying two different reactive π-systems was achieved to synthesize vinyl heteroarenes and conjugated 1,3-dienes in good yield and up to 100 % stereoselectivity, which remains unexplored until now. Most importantly, we report the unprecedented formation of E vs. Z-vinyl heteroarenes for different heteroarenes under identical conditions. Density functional theory (DFT) investigations unveil the mechanistic dichotomy between olefin and heteroarene activation followed by 1,2-migration, leading to E or Z-vinyl heteroarenes respectively. We also report a previously unknown reversal of stereoselectivity by using 2,3-Dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) as an electrophile. The Zweifel olefination using a boronate complex that carries two different olefins was previously unexplored due to significant challenges associated with the site-selective activation of olefins. We have solved this problem and reported the site-selective activation of olefins for the stereoselective synthesis of 1,3-dienes.     

Studies on the interaction of diacetylcurcumin with calf thymus-DNA

Ongoing research on curcumin and its structural derivatives are a subject of growing interest because of their demonstrated biological properties. Diacetylcurcumin (DAC), a synthetic derivative of natural non-toxic curcumin has been shown to affect a host of activities ranging from wound healing to life threatening diseases like AIDS, cancer etc. The interaction of diacetylcurcumin (DAC) with calf thymus-DNA (ct-DNA) has been investigated by spectroscopic and viscometric techniques. The fluorescence intensity of DAC was quenched by ct-DNA. The mean binding constant obtained from the spectroscopic techniques was 3.97 ± 0.31 × 10⁵ M⁻¹. Circular dichroism studies did not reveal any unwinding of the DNA helix on interaction with DAC, implying no conformational changes. The binding mode was analyzed by competitive binding between ethidium bromide (EB) and DAC for ct-DNA and also by viscometric studies. DAC was found to be a minor groove binder with a preference for the A-T region compared to the G-C region. This was substantiated by displacement studies with Hoechst 33258, a known minor groove binder. Docking studies were found to corroborate the experimental results.
