Abstract— Erythrocytes from patients with erythropoietic protoporphyria contain large amounts of protoporphyrin bound to (hemo)globin. Irradiation of these cells causes a shift in fluorescence emission maximum and a decreased fluorescence intensity which is consistent with transfer of protoporphyrin from (hemo)globin to the cell membrane. When the erythrocytes were irradiated intermittently, nearly 70% of the protoporphyrin was released and the hemolysis was less than 3%. Giving the total light dose as a single pulse, resulted in 84% protoporphyrin release and 16% hemolysis.
In vivo the erythrocytes obtain small, repetitive light doses when circulating in the dermal capillaries. We suggest the possibility that in patients with erythropoietic protoporphyria these small light pulses could be sufficient to photodamage the binding place of protoporphyrin on (hemo)globin. In the dark, protoporphyrin can then move from (hemo)globin through the cell membrane and bind to albumin in the serum. Our findings indicate that if protoporphyrin is not present in the cell membrane during irradiation, no photohemolysis will occur. This may explain why patients with erythropoietic protoporphyria have no abnormal hemolysis. The effect of intermittent light pulses may also contribute to the understanding of the protoporphyrin release from erythrocytes in patients with erythropoietic protoporphyria.  相似文献

The effect of conformational preference on the chiroptical properties of α-substituted succinic acids and esters 2 has been investigated using variable-temperature CD in solvents of different polarity. All available evidence indicates that when R' in 2 is alkyl or halogen, the preferred conformation is 1e with the -CH2COOR group eclipsed by the CO bond, while when R' in 2 is hydroxy, methoxy or amino, the major conformer is 1d in which the heteroatom is eclipsed by the CO bond. In both cases, conformation 1f appears to be the least favored, mainly on steric grounds. The results when R' = chloro or bromo do not support previously proposed conclusions and an alternative explanation is advanced. In the resultant octant projection (Fig, 2) it is shown that the sign of the Cotton effect for 1 will be determined by the position of the groups X and Y in the back octants, and since one of these is always H in the two favored conformations, the sign is actually determined by the position of the other group. This octant projection predicts successfully the sign of the ellipticity of the n→π* transition for any α-substituted succinic acid or ester with the appropriate substituent R', and appears to apply also to simple alkanoic acids and esters with the same substituents.  相似文献

A kinetic method for the simultaneous determination of magnesium and calcium ions in solution is described. The method is based on the dissociation reactions of cryptand (2.2.2) complexes; a stopped-flow injection technique is used with spectrophotometric monitoring of the phthalein complexone complexes of the released metal ions. Analyses were done at a rate of 80 h?1, with injected sample volumes of 80 μl. A microcomputer system for data acquisition and control of the system is described.  相似文献

Microwave Spectra of 13CH212C80,78Se and 12CH213C80,78Se (selenoketene) are recorded. B 5-13C-1,2,3-selenadiazole all four species are formed. The 13C scrambling may take place via a three-membered ring, selenirene, but as expected its microwave spectrum was not observed.  相似文献

The molecular structure of gaseous OVF3 has been determined by electron diffraction to be: rg(V-O) = 1.570(5) Å, rg(V-F) = 1.729(2) Å and ∠α(OVF) = 107.5(4)°. A modified force field has been fitted to results from spectroscopic as well as diffractional studies. A similar attempt to determine the force field for OVCl3 was not as successful as for OVF3, probably because the Coriolis constants are less accurately determined for that molecule.  相似文献

Semiempirical and density functional electronic structure theory methods were used to study SWNT-X--R bond strengths, where the single-walled carbon nanotube (SWNT) had an armchair or zigzag structure, the link heteroatom X was O, N(H), or S and the hydrocarbon chain R was CH(2)CH(3), CH(OH)CH(3), CHCH(2), or CH(CF(3))CH(3). In all systems the hydrocarbon was bonded to the end of the nanotube. The SWNT-X--R bond (that is, the bond joining the link atom to the hydrocarbon) is more than 0.4 eV stronger for armchair than for zigzag nanotubes with the same diameters, irrespective of whether O, N, or S are used as link atoms or whether OH, C==C, or CF(3) groups are present in the hydrocarbon chain. This raises the possibility for selective manipulation of armchair/zigzag nanotubes using a variety of link atoms and hydrocarbon structures. The SWNT-O--CH(CF(3))CH(3) bond is weaker than the SWNT-O--CH(2)CH(3) bond (for both armchair and zigzag nanotubes), while inclusion of a double bond in the ethyl chain increases the bond strengths. Also, SWNT-S--CH(2)CH(3) and SWNT-N(H)--CH(2)CH(3) bonds are stronger than SWNT-O--CH(2)CH(3) bonds.  相似文献

The evidence for this is collisional activation spectra, using various precursors (including labelled analogues) under electron impact and field ionization conditions. The chemical ionization (CI) mass spectra of the 2-tert-butyl-substituted 1,3-cyclopentane- and 1,3-cyclohexanediol diacetates and dimethyl ethers have been determined using isobutane and methane as reagent gases. From the differences in the spectra of these compounds, it clearly follows that steric and conformational effects are expresssed in the CI mass spectra. The relative impact of these effects, however, is strongly dependent on diol derivatization and 2-alkyl substitution.  相似文献
