Aloe vera L., a member of Liliaceae, is a medicinal plant and has a number of curative properties. We describe here the development of tissue culture method for high-frequency plantlet regeneration from inflorescence axis-derived callus cultures of sweet aloe genotype. Competent callus cultures were established on 0.8% agar-gelled Murashige and Skoog’s (MS) basal medium supplemented with 6.0 mg l−1 of 2,4-dichlorophenoxyacetic acid (2,4-D) and 100.0 mg l−1 of activated charcoal and additives (100 mg l−1 of ascorbic acid, 50.0 mg l−1 each of citric acid and polyvinylpyrrolidone, and 25.0 mg l−1 each of l-arginine and adenine sulfate). The callus cultures were cultured on MS medium containing 1.5 mg l−1 of 2,4-D, 0.25 mg l−1 of Kinetin (Kin), and additives with 4% carbohydrate source for multiplication and long-term maintenance of regenerative callus cultures. Callus cultures organized, differentiated, and produced globular embryogenic structures on MS medium with 1.0 mg l−1 of 2,4-D, 0.25 mg l−1 of Kin, and additives (50.0 mg l−1 of ascorbic acid and 25.0 mg l−1 each of citric acid, l-arginine, and adenine sulfate). These globular structures subsequently produced shoot buds and then complete plantlets on MS medium containing 1.0 mg l−1 of 6-benzylaminopurine and additives. A hundred percent regenerated plantlets were hardened in the greenhouse and stored under an agro-net house/nursery. The regeneration system defined could be a useful tool not only for mass-scale propagation of selected genotype of A. vera, but also for genetic improvement of plant species through genetic transformation.  相似文献

Department of Atomic Energy, India, has a systematic environmental monitoring program to ensure that the impact of radionuclides on human and environment is well within the limits, stipulated by the regulatory body. Concentrations of radionuclides are estimated in air, water, soil, sediment and biota in the environment around the power plant site on a regular basis. The environmental migration pattern and impact of radionuclides in the environment depends upon not only on the total concentration but the physico-chemical nature of radionuclide also. This paper presents a brief review of the studies of environmental transportation of radionuclides and influence of speciation with special reference to Indian conditions. A fairly good amount of results are available on the influence of speciation of radionuclides on the wet and dry deposition and preliminary results are available on the speciation analysis in dietary items, in soil and in ground water. Initial studies are concentrated on tritium, 137Cs, Uranium, 90Sr and their chemical analogues.  相似文献

The problem of flame propagation in imperfectly premixed mixtures—mixtures of reactants with variable composition—is considered in this numerical study. We carry out two-dimensional direct numerical simulations of a flame propagating in a globally lean fuel-oxidizer mixture with imposed velocity and composition fluctuations of various intensities. The configuration adopted is that of a flame front interacting with spatially evolving fluctuations, and the characteristic scales of the domain and of the fluctuations imposed are significantly larger than the characteristic thickness of the flame, to account for important flame dynamics such as the hydrodynamic instability. One-step chemistry and Fick’s diffusion law are considered, along with unity Lewis number assumption for all the species. It is observed, in agreement with previous results, that relatively weak fluctuations in composition alone may lead to a large increase in flame length and burning rate. The hydrodynamic instability caused by gas expansion, catalyzed by the composition fluctuations interacting with the flame, is found to be responsible for the flame length enhancement. It is observed as well that the relative importance of this effect diminishes as the velocity fluctuations present become more intense, and that composition fluctuations have a small impact on flame length for these cases. It is additionally found that, with increasing intensity of composition fluctuations, there is eventually a reduction of burning rate per unit length of flame which leads, consequently, to a weak reduction of overall burning rate for the largest velocity fluctuation intensities covered by this study.  相似文献

We study the deterministic dynamics of a periodically driven particle in the underdamped case in a spatially symmetric periodic potential. The system is subjected to a space-dependent friction coefficient, which is similarly periodic as the potential but with a phase difference. We observe that frictional inhomogeneity in a symmetric periodic potential mimics most of the qualitative features of deterministic dynamics in a homogeneous system with an asymmetric periodic potential. We point out the need of averaging over the initial phase of the external drive at small frictional inhomogeneity parameter values or analogously low potential asymmetry regimes in obtaining ratchet current. We also show that at low amplitudes of the drive, where ratchet current is not possible in the deterministic case, noise plays a significant role in realizing ratchet current.  相似文献

We study a periodically driven (symmetric as well as asymmetric) double-well potential system at finite temperature. We show that mean heat loss by the system to the environment (bath) per period of the applied field is a good quantifier of stochastic resonance. It is found that the heat fluctuations over a single period are always larger than the work fluctuations. The observed distributions of work and heat exhibit pronounced asymmetry near resonance. The heat loss over a large number of periods satisfies the conventional steady-state fluctuation theorem.  相似文献

A priori and a posteriori studies for large eddy simulation of the compressible turbulent infinitely fast reacting shear layer are presented. The filtered heat release appearing in the energy equation is unclosed and the accuracy of different models for the filtered scalar dissipation rate and the conditional filtered scalar dissipation rate of the mixture fraction in closing this term is analyzed. The effect of different closures of the subgrid transport of momentum, energy and scalars on the modeling of the filtered heat release via the resolved fields is also considered. Three explicit models of these subgrid fluxes are explored, each with an increasing level of reconstruction and all of them regularized by a Smagorinsky-type term. It is observed that a major part of the error in the prediction of the conditional filtered scalar dissipation comes from the unsatisfactory modeling of the filtered dissipation itself. The error can be substantial in the turbulent fluctuation (rms) of the dissipation fields. It is encouraging that all models give good predictions of the mean and rms density in a posteriori LES of this flow with realistic heat release corresponding to large density change. Although a posteriori results show a small sensitivity to subgrid modeling errors in the current problem, extinction–reignition phenomena involving finite-rate chemistry would demand more accurate modeling of the dissipation rates. A posteriori results also show that the resolved fields obtained with the approximate reconstruction using moments (ARM) agree better with the filtered direct numerical simulation since the level of reconstruction in the modeled subfilter fluxes is increased.  相似文献

In this paper a new whole field technique for determination of characteristic parameters in integrated photoelasticity using phase-shifting methodology is proposed. Appropriate optical arrangements necessary for this are presented and the intensity equations are obtained by Jones calculus. The new methodology is verified experimentally for the problem of a circular disk under diametral compression viewed in oblique incidence.  相似文献

Direct numerical simulations of homogeneous turbulence in stably stratified shear flow have been performed to aid the understanding of turbulence and turbulent mixing in geophysical flow. Two cases are compared. In the first case, which has been studied in the past, the mean velocity has vertical shear and the mean density is vertically stably stratified. In the second case, which has not been studied systematically before, the mean velocity has horizontal shear and the mean density is again vertically stably stratified. The critical value of the gradient Richardson number, for which a constant turbulence level is obtained, is found to be an order of magnitude larger in the horizontal shear case. The turbulent transport coefficients of momentum and vertical mass transfer are also an order of magnitude larger in the horizontal shear case. The anisotropy of the turbulence intensities are found to be in the range expected of flows with mean shear with no major qualitative change in the range of Richardson numbers studied here. However, the anisotropy of the turbulent dissipation rate is strongly affected by stratification with the vertical component dominating the others. The crystal and molecular structure of3 has been determined by X-ray analysis. Thecrystal data are: triclinic, space group P'1,a = 12.999(2),b = 14.114(3),c = 16.132(3) Å, = 91.62(2), = 97.71(1), = 91.38(2)°,V = 2930.6(9) Å3,Z = 2,Dm = 1.046 g cm-3.Rotational disorder has been seen in the t-butyl groups whichwere refined isotropically. Least-squares refinementbased on F2, using 4827 observedreflections with I > 2(I) and 18 restraints,led to R = 0.123. The calix is found in a coneconformation. The crown ether part of the molecule isnot in a fully extended conformation but has foldsin the chain.
