Despite all the attempts to create advanced hemoglobin (Hb)‐based oxygen carriers (HBOCs) employing an encapsulation platform, major challenges including attaining a high Hb loading and long circulation times still need to be overcome. Herein, the fabrication, for the first time, of nanoparticles fully made of Hb (Hb‐NPs) employing the electrospray technique is reported. The Hb‐NPs are then coated by antioxidant and self‐polymerized poly(dopamine) (PDA) to minimize the conversion of Hb into nonfunctional methemoglobin (metHb). The PDA shell is further functionalized with poly(ethylene glycol) (PEG) to achieve stealth properties. The results demonstrate that the as‐prepared Hb‐NPs are hemo‐ and biocompatible while offering antioxidant protection and decreasing the formation of metHb. Additionally, decoration with PEG results in decreased protein adsorption onto the Hb‐NPs surface, suggesting a prolonged retention time within the body. Finally, the Hb‐NPs also preserve the reversible oxygen‐binding and releasing properties of Hb. All in all, within this study, a novel HBOCs with high Hb content is fabricated and its potential as an artificial blood substitute is evaluated.  相似文献

Roflumilast is a phosphodiesterase type 4 inhibitor that is administered orally as a long-term, in the clinical treatment of chronic obstructive pulmonary disease and asthma. Launched in 2010 for the European market, it currently does not have an official monograph. Here, a reproducible gradient RP-HPLC method was developed and validated for the separation and determination of Roflumilast in the presence of its six major degradation products. Separation was performed on a C18 analytical column (250?×?4.6?mm, 5?µm) with a mobile phase-A of ACN and a phase-B of ammonium acetate buffer (5?mM, pH 4.2) containing triethylamine (0.5% v/v). The most effective RP-HPLC gradient program was determined to be 0/80, 35/10, 36/80, 40/80 (time in minutes/% mobile phase-B). The flow rate was 1.0?ml/min and the column temperature was 25°C. The success of separation of the degradation products with different chemical characteristics was obtained by extending the time of the gradient, changing the proportion of the mobile phases and increasing the velocity of the flow. Two detectors were evaluated for the identification of degradation products and Roflumilast: a diode-arrary detector and a charged aerosol detector. The inability of the charged aerosol detector to dectect one of the six degradation products indicated that the method developed with RP-HPLC and the diode-array detector was more suitable for Roflumilast analysis. The method was validated according to specificity, linearity, LOD, LOQ, accuracy, precision and robustness.  相似文献

We discuss the two-dimensional isotropic antiferromagnet in the framework of gauge invariance. Gauge invariance is one of the most subtle useful concepts in theoretical physics, since it allows one to describe the time evolution of complex physical system in arbitrary sequences of reference frames. All theories of the fundamental interactions rely on gauge invariance. In Dirac’s approach, the two-dimensional isotropic antiferromagnet is subject to second-class constraints, which are independent of the Hamiltonian symmetries and can be used to eliminate certain canonical variables from the theory. We have used the symplectic embedding formalism developed by a few of us to make the system under study gauge invariant. After carrying out the embedding and Dirac analysis, we systematically show how second-class constraints can generate hidden symmetries. We obtain the invariant second-order Lagrangian and the gauge-invariant model Hamiltonian. Finally, for a particular choice of factor ordering, we derive the functional Schröodinger equations for the original Hamiltonian and for the first-class Hamiltonian and show them to be identical, which justifies our choice of factor ordering.  相似文献

This paper presents a biased random-key genetic algorithm for the resource constrained project scheduling problem. The chromosome representation of the problem is based on random keys. Active schedules are constructed using a priority-rule heuristic in which the priorities of the activities are defined by the genetic algorithm. A forward-backward improvement procedure is applied to all solutions. The chromosomes supplied by the genetic algorithm are adjusted to reflect the solutions obtained by the improvement procedure. The heuristic is tested on a set of standard problems taken from the literature and compared with other approaches. The computational results validate the effectiveness of the proposed algorithm.  相似文献

In a topological space, a family of continuous mappings is called universal if its action, in at least one element of the space, is dense. If the mappings are unitary or trace-preserving completely positive, the notion of universality is closely related to the notion of controllability in either closed or open quantum systems. Quantum controllability in infinite dimensions is discussed in this setting and minimal generators are found for full control universal families. Some of the requirements of the operators needed for control in infinite dimensions follow from the properties of the infinite unitary group. Hence, a brief discussion of this group and the appropriate mathematical spaces is also included.  相似文献

A useful application for copula functions is modeling the dynamics in the conditional moments of a time series. Using copulas, one can go beyond the traditional linear ARMA (p,q) modeling, which is solely based on the behavior of the autocorrelation function, and capture the entire dependence structure linking consecutive observations. This type of serial dependence is best represented by a canonical vine decomposition, and we illustrate this idea in the context of emerging stock markets, modeling linear and nonlinear temporal dependences of Brazilian series of realized volatilities. However, the analysis of intraday data collected from e‐markets poses some specific challenges. The large amount of real‐time information calls for heavy data manipulation, which may result in gross errors. Atypical points in high‐frequency intraday transaction prices may contaminate the series of daily realized volatilities, thus affecting classical statistical inference and leading to poor predictions. Therefore, in this paper, we propose to robustly estimate pair‐copula models using the weighted minimum distance and the weighted maximum likelihood estimates (WMLE). The excellent performance of these robust estimates for pair‐copula models are assessed through a comprehensive set of simulations, from which the WMLE emerged as the best option for members of the elliptical copula family. We evaluate and compare alternative volatility forecasts and show that the robustly estimated canonical vine‐based forecasts outperform the competitors. The equilibrium tilt angle profile in a cell limited by two concentric cylinders filled with nematic liquid crystals is determined for strong homeotropic anchoring at the surfaces. The anchoring condition is such that the nematic director is perpendicular to the cylinder axes and a radial nonuniform electric field is applied to investigate a Fréedericksz transition. The distortions induced by the field remain in the plane perpendicular to the cylinder axes, and a threshold field is analytically determined indicating a transition from a pure splay to a splay-bend conformation of the director. It is shown that this transition can be induced by the thickness of the region between the two cylinders, and can be detected even in the absence of an external field. If the maximum value of the tilt angle is assumed as an order parameter, its behavior near to the transition can be used to obtain the critical exponent, which is the same as the one obtained in the mean field approximation. These results are indications that nontrivial consequences may occur when complex fluids are subject to non-planar geometries.  相似文献

In the Regge kinematics the amplitude for gluon production off three scattering centers is found in the Lipatov effective action technique. The vertex for gluon emission with the reggeon splitting in three reggeons is calculated and its transversality is demonstrated. It is shown that in the sum of all contributions terms containing principal value singularities are canceled and substituted by the standard Feynman poles. These results may be used for calculation of the inclusive cross section for gluon production on two nucleons in the nucleus.  相似文献

To provide an application for the method of linear least squares to data collected in a laboratory, a beaker with water is heated in a microwave oven, and the water temperature is measured as a function of heating variables (time and oven setting). This procedure enables a student to obtain a regression line for each oven setting, and to evaluate the intercept and slope of this line and compare them with the initial temperature of the water and the heating versus oven setting relationship described in the microwaves manufacturers manual. MicroRNAs (miRs) have emerged as important clinical biomarkers with both diagnostic and prognostic value for relevant diseases, such as cancer. MiRs pose unique challenges for detection and are currently detected by northern blotting, real‐time PCR, and microarray techniques. These expensive, complicated, and time‐consuming techniques are not feasible for on‐site miR determination. In this study, amperometric magnetobiosensors involving RNA‐binding viral protein p19 as a selective biorecognition element were developed for miR quantification. The p19‐based magnetosensors were able to detect 0.4 fmol of a synthetic target and endogenous miR‐21 (selected as a model for its role in a wide variety of cancers) in only 2 h in total RNA extracted from cancer cells and human breast‐tumor specimens without PCR amplification and sample preprocessing. These results open up formidable perspectives for the diagnosis and prognosis of human cancers and for drug‐discovery programs.  相似文献
