A label-free immunosensor based on an aptamer-modified graphene field-effect transistor (G-FET) is demonstrated. Immunoglobulin E (IgE) aptamers with an approximate height of 3 nm were successfully immobilized on a graphene surface, as confirmed by atomic force microscopy. The aptamer-modified G-FET showed selective electrical detection of IgE protein. From the dependence of the drain current variation on the IgE concentration, the dissociation constant was estimated to be 47 nM, indicating good affinity and the potential for G-FETs to be used in biological sensors.  相似文献

The development of a new configuration of chemical ionization (CI)‐based ion source is presented. The ambient air containing the gaseous sample is sniffed into an enclosed ionization chamber which is of sub‐ambient pressure, and is subsequently mixed with metastable species in front of the ion inlet of the mass spectrometer. Metastable helium atoms (He*) are used in this study as the primary ionizing agents and are generated from a dielectric barrier discharge (DBD) source. The DBD is powered by an AC high‐voltage supply and the configuration of the electrodes is in such a way that the generated plasma is confined within the discharge tube and is not extended into the ionization chamber. The construction of the ion source is simple, and volatile compounds released from the bulky sample can also be analyzed directly by approaching the sample to the sampling nozzle. When combined with heated nitrogen or other desorption methods, its application can also be extended to non‐volatile compounds, and the consumption for helium can be kept minimum solely for maintaining the stable discharge and gas phase ionization. Applications to non‐proximate sample analysis, direct determination of active ingredients in drug tablets and the detection of trace explosive such as hexamethylene triperoxide diamine are demonstrated. Novel piperidine carboxylic acid derivatives of 10H-pyrazino[2,3-b][1,4]benzothiazine were prepared and evaluated for their inhibitory activity on the upregulation of adhesion molecules such as intercellular adhesion molecule-1 (ICAM-1). Replacement of the methanesulfonyl group on the piperidine ring of previously prepared derivatives with a carboxylic acid-containing moiety resulted in a number of potent adhesion molecule inhibitors. Of these, (anti) [3-(10H-pyrazino[2,3-b][1,4]benzothiazin-8-yl)methyl-3-azabicyclo[3.3.1]non-9-yl]acetic acid 2q (ER-49890), showed the most potent oral inhibitory activities against neutrophil migration in an interleukin-1 (IL-1) induced paw inflammation model using mice, and leukocyte accumulation in a carrageenan pleurisy model in the rat, and therapeutic effect on collagen-induced arthritis in rats.  相似文献

Fluorescence imaging is the most powerful technique currently available for continuous observation of dynamic intracellular processes in living cells. Suitable fluorescence probes are naturally of critical importance for fluorescence imaging, but only a very limited range of biomolecules can currently be visualized because of the lack of flexible design strategies for fluorescence probes. At present, design is largely empirical. Here we show that the carboxylic group of traditional fluorescein dyes, formerly considered indispensable, has been replaced with other substituents, affording various kinds of new fluoresceins. Further, by breaking out of the traditional structure of fluorescein, we developed the first and totally rational design strategy for novel fluorescence probes based on a strict photochemical basis. The value of this approach is exemplified by its application to develop a novel, highly sensitive, and membrane-permeable fluorescence probe for beta-galactosidase, which is the most widely used reporter enzyme.  相似文献

Cadmium sulfo-selenide Cd1+y(SxSe1?x) single crystals were grown by the sublimation method under controlled partial pressures of sulfur and selenium. The partial pressure control was achieved by amounts of sulfur and selenium introduced in a closed growth tube. To control the deviation from stoichiometry y for the specified compositions x, the sum of chalcogen partial pressures Pps(=pS2+PSe2) was regulated, holding the partial pressure ratio (PS2PSe2) constant. The comp osition x of single crystals, grown with appreciable growth rate, was found to be the same one as the source specimen but the electrical conductivity changed extremely from 1 to 10-12 (ω cm)-1 with increasing Pps. Hall effect and thermally stimulated current (TSC) were also measured on the high and low conductivity specimens respectively and found to depend upon the controlled Pps. From these experimental results, it was confirmed that the deviation from stoichiometry y was effectively controlled. The transport rate was also precisely measured as a function of controlled Pps.  相似文献

Resonant four-wave mixing (RFWM) based on the photo-induced refractive index change in erbium-doped optical fibers is demonstrated experimentally. The erbium-doped fibers act as a saturable absorber at 1.536 μm pump wavelength. Dependence of efficiency of RFWM on the pump power, the dopant concentration, the fiber length, and the signal modulation frequency are analyzed using three types of erbium-doped fibers differing in dopant concentration. It is found that a signal modulation frequency higher than 10 kHz is required for the correct measurement. The RFWM efficiency is improved with an increase in the input pump power in a range up to 1.5 mW and the highest reflectivity of 5.1% is attained. The highest RFWM efficiency is achieved at αgsL ∼ 5 dB, which is in good agreement with the theory.  相似文献

Al---Zr, Al---Nb, Al---Mo, Al---Ta and Al---W alloys were prepared by DC magnetron sputtering using an Al target with embedded heavy metals disks and substrate rotating around their own central axes and revolving around the central axes of the sputtering chamber. They were examined with X-ray diffraction (XD) of the Cu K, and Al---Ta alloys were also observed by transmission electron microscopy (TEM). Within the diffraction angles, 2τ = 20° ≈ 90°, XD patterns of the examined specimens were all typical of the amorphous structure. However, at the small angle region, 2τ < 10°, a number of sharp diffraction peaks appeared. The lattice spacing obtained from the diffraction peaks was linearly dependent on the inverse of the revolution period of the substrate during the sputter deposition. TEM images showed a layered structure which corresponded to the lattice spacing detected by XD. It is concluded that the superlattice-like structure appearing in the sputter-deposited alloy films arises from the compositional heterogeneity in the sputtered flux of atoms, which comes from the large non-uniformity of the target structure and sputtering conditions.  相似文献

An experimental study of high-order harmonic generation in nitrogen molecules (N2) has been made using intense visible (616 nm) dye-laser pulses, where the harmonic radiation up to the 21st order is observed. The harmonic distribution represents a plateau that is preceded by an intensity minimum at the 7th order. The harmonic generation characteristics were atomic-like. It has been found that there are some similarities in the high-order harmonic generation characteristics for N2 and Ar, including the highest-order harmonics, harmonic distributions, and the influence of the multiphoton ionization on the high-order harmonic generation. These similarities are reasonably attributed to the energetic correspondence of excited levels and ionization potentials. It is pointed out that the ac Stark shift of excited levels and ionization potentials plays an important role also in the high-order harmonic generation in N2.  相似文献

The stress relaxation under large step equibiaxial elongation for low‐density polyethylene with long‐chain branches revealed that the time‐strain separability holds in relaxation modulus GB(t, εB), and damping function hBB) exhibits weaker equibiaxial elongational strain εB dependence than that predicted by the Doi–Edwards theory without the independent alignment approximation. Dependencies of damping function h(γ) for step shear deformation and hBB) on stretch ratio α of polymer contour length and orientation of a polymer chain in direction of the maximum orientation were evaluated, and it was found that the α dependencies of h(γ) and hBB) are different, whereas dependencies of h(γ) and hBB) on the orientation coincide fairly well. These results indicate that the damping is dominated by the chain orientation rather than α. This implies that withdrawal of long‐chain branches into tube of a backbone chain occurs when the orientation of the long‐chain branches is large and friction force against the branch point withdrawal is small. © 2009 Wiley Periodicals, Inc.
