InCl3‐Driven Regioselective Synthesis of Functionalized/Annulated Quinolines: Scope and Limitations

The efficient, regioselective synthesis of functionalized/annulated quinolines was achieved by the coupling of 2‐aminoaryl ketones with alkynes/active methylenes/α‐oxoketene dithioacetals promoted by InCl₃ in refluxing acetonitrile as well as under solvent‐free conditions in excellent yields. This transformation presumably proceeded through the hydroamination–hydroarylation of alkynes, and the Friedländer annulation of active methylene compounds and α‐oxoketene dithioacetals with 2‐aminoarylketones. In addition, simple reductive and oxidative cyclization of 2‐nitrobenzaldehyde and 2‐aminobenzylalcohol, respectively, afforded substituted quinolines. Systematic optimization of the reaction parameters allowed us to identify two‐component coupling (2CC) conditions that were tolerant of a wide range of functional groups, thereby providing densely functionalized/annulated quinolines. This approach tolerates the synthesis of various bioactive quinoline frameworks from the same 2‐aminoarylketones under mild conditions, thus making this strategy highly useful in diversity‐oriented synthesis (DOS). The scope and limitations of the alkyne‐, activated methylene‐, and α‐oxoketene dithioacetal components on the reaction were also investigated.     

Microwave assisted highly efficient one-pot multi-component synthesis of novel 2-(tetrasubstituted-1H-pyrrol-3-yl)-4H-chroman-4-ones catalyzed by heterogeneous reusable silica gel supported polyphosphoric acid (PPA/SiO2)

A solvent-free, eco-friendly and facile approach for the synthesis of highly functionalized tetrasubstituted pyrroles has been reported through one-pot four-component reaction of aldehyde, amine, nitroalkane and 1,3-diketone using silica gel supported polyphosphoric acid (PPA–SiO₂) under microwave condition. The reaction occured through the in situ formation of β-keto enamine and nitrostyrene analog following Michael addition and finally intramolecular annulation affording the products in good yields. The key features of the present method include clean reaction, mild conditions, low catalyst loading, straightforward, high to excellent yields, short reaction time, avoiding use of harmful metal catalyst and organic solvent, environmentally friendly compared to the existing methods, recovery and reusability of catalyst and easy workup procedure.     

Enhancement of electrical conductivity,director relaxation frequency and slope of electro-optical characteristics in the composites of single-walled carbon nanotubes and a strongly polar nematic liquid crystal

In the present work, single-walled carbon nanotubes (SWCNTs) were dispersed in a room temperature nematic liquid crystal 4-pentyl-4′-cyanobiphenyl at the concentration of 0.02 and 0.05 wt%. Differential scanning calorimetry and temperature-dependent dielectric studies suggest decrease in clearing temperature of the composite materials as compared to the pure material. Ionic conductivity increases by two orders of magnitude due to the dispersion of such a low concentration (0.05 wt%) of SWCNTs. Dielectric studies also show that the presence of the SWCNTs decreases the effective longitudinal as well as transverse components of the dielectric permittivity. For homeotropic aligned samples, a relaxation mechanism has been detected in the lower MHz region both for the pure as well as dispersed samples. Presence of SWCNTs increases the relaxation frequency corresponding to flip-flop motion of molecules around their short axes. From frequency-dependent dielectric studies, important dielectric parameters such as relaxation frequency, dielectric strength and distribution parameters have been determined. Electro-optical experiments show that the threshold voltage decreases and the steepness of the transmission voltage curve improves due to the dispersion of SWCNTs.     

Long-term neprilysin gene transfer is associated with reduced levels of intracellular Abeta and behavioral improvement in APP transgenic mice

Background  

Proteolytic degradation has emerged as a key pathway involved in controlling levels of the Alzheimer's disease (AD)-associated amyloid-β (Aβ) peptide in the brain. The endopeptidase, neprilysin, has been implicated as a major Aβ degrading enzyme in mice and humans. Previous short and intermediate term studies have shown the potential therapeutic application of neprilysin by delivering this enzyme into the brain of APP transgenic mice using gene transfer with viral vectors. However the effects of long-term neprilysin gene transfer on other aspects of Aβ associated pathology have not been explored yet in APP transgenic mice.     

An improved turbidimetric procedure for the determination of sulphate in plants and soils

A rapid turbidimetric procedure for the determination of sulphate in soils and plants has been developed with sorbitol as stabilizing agent. When used in a concentration of 20% in the final volume, the sorbitol regulates the size of the BaSO(4) particles formed and stabilizes them against sedimentation, growth and aggregation for long periods of time. The conditions of precipitation are not rigorous, as they are in similar procedures. Linearity of the calibration curve up to 50 ppm of sulphate and good reproducibility are special attributes of the procedure.     

Titration of thiol groups in non-aqueous solvents

Thiols are titrated in acetone or dimethylformamide with sodium methoxide, employing visual end-point detection with Thymol Blue, Victoria Blue, p-hydroxyazobenzene or Azo Violet. Aromatic thiols are titrated in the presence of aliphatic thiols in acetone, with Thymol Blue as indicator.     

Self-aggregation of reverse bis peptide conjugate derived from the unstructured region of the prion protein

A novel bis scaffold containing a pentapeptide PHGGG, derived from the prion octarepeat, forms fibrils of nanometric dimensions, thus indicating that certain segments in the unstructured region of the prion protein may facilitate an initial fibrillation event.     

Rheological microscopy: local mechanical properties from microrheology

We demonstrate how tracer microrheology methods can be extended to study submicron scale variations in the viscoelastic response of soft materials; in particular, a semidilute solution of lambda-DNA. The polymer concentration is depleted near the surfaces of the tracer particles, within a distance comparable to the polymer correlation length. The rheology of this microscopic layer alters the tracers' motion and can be precisely quantified using one- and two-point microrheology. Interestingly, we found this mechanically distinct layer to be twice as thick as the layer of depleted concentration, likely due to solvent drainage through the locally perturbed polymer structure.     

Versatile and facile synthesis of diverse semisynthetic tetracycline derivatives via Pd-catalyzed reactions

A diverse collection of tetracycline derivatives has been synthesized utilizing Heck, Suzuki, and other palladium-coupling reactions via tetracycline arenediazonium and iodoarene salts. Large numbers of tetracyclines are now possible via these reactions, including numerous upper periphery derivatives of doxycycline, minocycline, sancycline, and methacycline modified at positions C7, C9, and C6-C13 on the tetracycline naphthacene ring. Application of palladium-coupling reactions to the tetracyclines has yielded new tetracycline classes with differing structural attributes, greatly increasing the structural diversity of this family of antibiotics, one of the last of the early antibiotic families to be expanded by organic and medicinal chemistry.
