本合成了3种不同载体蛋白的雌二醇抗原,研究了载体蛋白免疫原性对雌二醇抗体产生的影响。应用酶联免疫吸附分析法测定了血清中雌二醇,浓度范围为5-160ng/ml(1.8×10^-^8-5.9×10^-^7mol/L).RSD%小于10.%,检测限为1.97ng/ml,相当于98.5pg/孔(按空白平均值的二倍标准偏差计算)。另外,本还建立了一种用考马斯亮蓝G-250测定半抗原与蛋白结合比的方法,提  相似文献

Unlabeled fluoroimmunoassay with a europium chelate is described. The recommended method is simple because the complicated labeling process is omitted. The recovery of some samples ranges from 91 to 111%. The relative standard deviation of the method is less than 10%. The detection limit is about 10−10mol liter−1antigen.  相似文献

The relative ability of peroxidase-like metallotetrakis(N-methylpyridiniumyl)porphyrins [Me-TMPyP, Me = Mn(III), Fe(III), Co(III), Ni(II), Cu(II), and Zn(II)] to catalyse the hydrogen peroxide oxidation of homovanillic acid to a fluorescent dimer has been studied. The complexes of Mn, Fe and Co are effective catalysts in the reaction, but the complexes of Ni, Cu and Zn are not. The catalytic behaviour of Mn-TMPyP, Fe-TMPyP and Co-TMPyP has been compared with that of HRP in both enzymatic and kinetic analysis. The sequence of peroxidase-like catalytic activity is Mn-TMPyP> Co-TMPyP> Fe-TMPyP. The catalytic activity of Mn-TMPyP is 84% of that of HRP. These Me-TMPyP (Me = Mn, Fe, and Co) compounds are good substitutes for HRP in enzymatic analysis. Traces of hydrogen peroxide and glucose can be determined with the Me-TMPyP systems.  相似文献

N,N′-Dicyanomethyl-o-phenylenediamine was synthesized with a 90% yield by a reaction ofo-phenylenediamine with chloroacetonitrile in triethylamine. Our experimental results showed that it was the effective fluorogenic substrate for horseradish peroxidase (HRP) and hemin. TheKmfor the HRP system was 48 μM,and that for hemin was 1.3 μM.Properties of the substrate were evaluated from the detection limits of enzymes and H2O2. The linear ranges for the determination of HRP and hemin were 21–150 pMand 2–20 nM,respectively. The linear range for the determination of H2O2using HRP or hemin was 18–140 and 60–1000 nM,respectively. The structural elucidation of the fluorescent product using NMR and mass spectral techniques was proposed to be 1,2-dihydro-2-imido-imidazo[1,2-a] quinoxaline. Based on the product structure and earlier reports, the possible reaction mechanism of HRP and the substrate was also proposed, i.e., two steps for ring closures, one step of isomerization, and a final step of oxidative dehydrogenation.  相似文献

Traces of hydrogen peroxide (8.5 × 10?8–2.5 × 10?6 mol/l) and, indirectly, glucose (3–44 × 10?6 mol/l) can be determined by the fluorescence reaction between homovanillic acid and hydrogen peroxide. Mn-TPPS4 is found to have very similar catalytic properties to horse peroxidase.  相似文献

Based on the interaction between Eriochrome Blue Black R (EBBR) and proteins, which causes a strong light-scattering signal with the maximum scattering peak located at 398 nm, a simple, rapid, sensitive and selective method is developed for the determination of proteins by the light-scattering technique using a common spectrofluoremeter. Under proper experimental conditions, the protein determination can be performed in the range of 0.1-25, 0.1-20 and 0.25-25 μg ml−1 for bovine serum albumin (BSA), human serum albumin (HSA) and human immunoglobulin G (IgG), respectively. The detection limit, calculated as 3 times the S.D. of nine blank measurements, are 33 μg l−1 for BSA, 25 μg l−1 for HSA and 38 μg l−1 for IgG. Moreover, there is no significant difference among the scattering signals yielded by HSA, IgG and BSA, and almost no interference of many amino acids and metal ions. The method has been satisfactorily applied to the direct determination of the total protein in human serum, saliva and urine samples. The results obtained from the studies on the binding characteristics of EBBR to BSA indicated that an electrostatic force existed in the binding system, and the binding constant (K) and the number of the binding sites (n) at 25 °C are 1.69×105 l mol−1 and 0.946, respectively.  相似文献

As the hardware of FLIM technique becomes mature, the most important criterion for FLIM application is the correct interpretation of its data. In this research, first of all, a more orthogonal phasor approach, called as Modified Phasor Approach (MPA), is put forward. It is a way to calculate the lifetime of the complex fluorescent process, and a rule to measure how much the fluorescence process deviates from single exponential decay. Secondly, MPA is used to analysis the time-resolved fluorescence processes of the transfected CHO-K1 Cell lines expressing adenosine receptor A1R tagged by CYP and YFP, measured in the channel of the acceptor. The image of the fluorescence lifetime and the multiplication of the fluorescence lifetime and deviation from single exponential decay reveal the details of the Homo-FRET. In one word, MPA provides the physical meaning in its whole modified phasor space, and broadens the way for the application of the fluorescence lifetime imaging.  相似文献

The nervous system is a significant part of the human body, and peripheral nerve injury caused by trauma can cause various functional disorders. When the broken end defect is large and cannot be repaired by direct suture, small gap sutures of nerve conduits can effectively replace nerve transplantation and avoid the side effect of donor area disorders. There are many choices for nerve conduits, and natural materials and synthetic polymers have their advantages. Among them, the nerve scaffold should meet the requirements of good degradability, biocompatibility, promoting axon growth, supporting axon expansion and regeneration, and higher cell adhesion. Polymer biological scaffolds can change some shortcomings of raw materials by using electrospinning filling technology and surface modification technology to make them more suitable for nerve regeneration. Therefore, polymer scaffolds have a substantial prospect in the field of biomedicine in future. This paper reviews the application of nerve conduits in the field of repairing peripheral nerve injury, and we discuss the latest progress of materials and fabrication techniques of these polymer scaffolds.  相似文献
