Dynamics of folding in semiflexible filaments

The dynamics of a single semiflexible filament under the action of a compressing force is simulated. We find that the filament folds asymmetrically with a folding length which depends only on the bending stiffness kappa and the applied force f. It is shown that this behavior can be attributed to the exponentially falling tension profile in the filament. While the folding time tau(0) depends on the initial configuration, the distance moved by the terminal point of the filament and the length of the fold scales as tau(1/2) at tau>tau(0) and is independent of the initial configuration.     

Determination of aliphatic amines by gas chromatography–mass spectrometry after in-syringe derivatization with pentafluorobenzoyl chloride

A simple and highly sensitive gas chromatographic method has been developed for the determination of low molecular weight short-chain aliphatic amines (SCAAs) after their simultaneous extraction and in-syringe derivatization with pentafluorobenzoyl chloride (PFBOC). Derivatization of the low molecular weight aliphatic amines in bicarbonate buffer of pH 10.5 with PFBOC was followed by immersed solvent microextraction. Derivatization conditions, including reagent concentration, reaction pH, ionic concentration of buffer, reaction time, stirring rate, reaction temperature and extraction solvent, have been investigated for method optimization. Linearity was studied within range of 0.15 pg ml⁻¹–50 ng ml⁻¹. The correlation coefficients were between 0.9934 and 0.9999. Detection limit of derivatized amines proved to be in the range of 0.117–1.527 pg ml⁻¹, and the intraday and interday relative standard deviation (RSD) values were less than 8% with respect to peak area. The method was applied for analysis of lake, river and industrial waste water. The recoveries of extraction from lake, river and industrial waste water samples, which have been spiked with different levels of aliphatic amines, were in the range of 68–99%, 63–102% and 62–105%, respectively.     

Synthesis of Some 2-(3-Alkyl/aryl-5-trifluoromethylpyrazol-1-yl)-4-(coumarin-3-yl)thiazoles as Novel Antibacterial Agents

Abstract

Herein, we describe a one-pot synthesis of some novel 2-(3-alkyl/aryl-5-trifluoromethylpyrazol-1-yl)-4-(coumarin-3-yl)thiazoles (6) involving the reaction of 3-alkyl/aryl-5-hydroxy-5-trifluoromethyl-4,5-dihydropyrazole-1-thiocarboxamides (3) with 3-bromoacetylcoumarins (5) in the presence of sodium carbonate in ethanol. Reaction of 3 with 5 in the absence of sodium carbonate, however, resulted in the formation of 2-(3-alkyl/aryl-5-hydroxy-5-trifluoromethyl-4,5-dihydropyrazol-1-yl)-4-(coumarin-3-yl)thiazoles, which were subsequently dehydrated to 6 by refluxing in ethanol in the presence of sodium carbonate. The structure of the synthesized compounds (6) was confirmed by infrared (IR), mass, ¹H NMR, and ¹³C NMR spectra and elemental analysis data. Newly synthesized compounds (6) showed moderate to good activity against Gram-positive bacteria.     

Nanoindentation as a Probe for Mechanically‐Induced Molecular Migration in Layered Organic Donor–Acceptor Complexes

Nanoindentation and scratch experiments on 1:1 donor–acceptor complexes, 1 and 2 , of 1,2,4,5‐tetracyanobenzene with pyrene and phenanthrene, respectively, reveal long‐range molecular layer gliding and large interaction anisotropy. Due to the layered arrangements in these crystals, these experiments that apply stress in particular directions result in the breaking of interlayer interactions, thus allowing molecular sheets to glide over one another with ease. Complex 1 has a layered crystal packing wherein the layers are 68° skew under the (002) face and the interlayer space is stabilized by van der Waals interactions. Upon indenting this surface with a Berkovich tip, pile‐up of material was observed on just one side of the indenter due to the close angular alignment of the layers with the half angle of the indenter tip (65.35°). The interfacial differences in the elastic modulus (21 %) and hardness (16 %) demonstrate the anisotropic nature of crystal packing. In 2 , the molecular stacks are arranged in a staggered manner; there is no layer arrangement, and the interlayer stabilization involves C? H???N hydrogen bonds and π???π interactions. This results in a higher modulus (20 %) for (020) as compared to (001), although the anisotropy in hardness is minimal (4 %). The anisotropy within a face was analyzed using AFM image scans and the coefficient of friction of four orthogonal nanoscratches on the cleavage planes of 1 and 2 . A higher friction coefficient was obtained for 2 as compared to 1 even in the cleavage direction due to the presence of hydrogen bonds in the interlayer region making the tip movement more hindered.     

MYMIV-AC2, a Geminiviral RNAi Suppressor Protein, Has Potential to Increase the Transgene Expression

Gene silencing is one of the limiting factors for transgene expression in plants. But the plant viruses have learnt to suppress gene silencing by encoding the protein(s), called RNA silencing suppressor(s) (RSS). Hence, these proteins could be used to overcome the limitation for transgene expression. The RNAi suppressors, namely HC-Pro and P19, have been shown to enhance the transgene expression but other RSS proteins have not been screened for similar role. Moreover, none of RSSs from the DNA viruses are known for enhancing the expression of transgenes. The Mungbean Yellow Mosaic India Virus (MYMIV) belonging to the genus Begomovirus within the family of Geminiviridae encodes an RSS called the AC2 protein. Here, we used AC2 to elevate the expression of the transgenes. Upon introduction of MYMIV-AC2 in the silenced GFP transgenic tobacco lines, by either genetic hybridisation or transgenesis, the GFP expression was enhanced several fold in F1 and T0 lines. The GFP-siRNA levels were much reduced in F1 and T0 lines compared with those of the initial parental silenced lines. The enhanced GFP expression was also observed at the cellular level. This approach was also successful in enhancing the expression of another transgene, namely topoisomeraseII.     

Synthesis and characterization of block copolymers from 2‐vinylnaphthalene by anionic polymerization

The anionic polymerization of 2‐vinylnaphthalene (2VN) has been studied in tetrahydrofuran (THF) at ?78 °C and in toluene at 40 °C. 2VN polymerization in THF, toluene, or toluene/THF (99:1 v/v) initiated by sec‐butyllithium (sBuLi) indicates living characteristics, affording polymers with predefined molecular weights and narrow molecular weight distributions. Block copolymers of 2VN with methyl methacrylate (MMA) and tert‐butyl acrylate (tBA) have been synthesized successfully by sequential monomer addition in THF at ?78 °C initiated by an adduct of sBuLi–LiCl. The crossover propagation from poly(2‐vinylnaphthyllithium) (P2VN) macroanions to MMA and tBA appears to be living, the molecular weight and composition can be predicted, and the molecular weight distribution of the resulting block copolymer is narrow (weight‐average molecular/number‐average molecular weight < 1.3). Block copolymers with different chain lengths for the P2VN segment can easily be prepared by variations in the monomer ratios. The block copolymerization of 2VN with hexamethylcyclotrisiloxane also results in a block copolymer of P2VN and poly(dimethylsiloxane) (PDMS) contaminated with a significant amount of homo‐PDMS. Poly(2VN‐b‐nBA) (where nBA is n‐butyl acrylate) has also been prepared by the transesterification reaction of the poly(2VN‐b‐tBA) block copolymer. Size exclusion chromatography, Fourier transform infrared, and ¹H NMR measurements indicate that the resulting polymers have the required architecture. The corresponding amphiphilic block copolymer of poly(2VN‐b‐AA) (where AA is acrylic acid) has been synthesized by acidic hydrolysis of the ester group of tert‐butyl from the poly(2VN‐b‐tBA) copolymer. © 2002 Wiley Periodicals, Inc. Synthesis of dihydropyrazolo[3′,4′:3,4]pyrrolo[1,2-a]indoles and spiro-[3H-indole-3,3′-[Δ-1,2,4]-triazolin]-2-ones via intra and intermolecular 1,3-dipolar cycloadditions

A new class of dihydropyrazolo[3′,4′:3,4]pyrrolo[1,2-a]indoles and spiro[3H-indole-3,3′-[Δ²-1,2,4]-triazoline]-2-ones were synthesised via intra and intermolecular 1,3-dipolar cycloaddition reactions in good yields.
