A unique peptide based search algorithm for identification of protein mixture using PMF is proposed. The proposed search algorithm utilizes binary search and heapsort programs to generate frequency chart depicting the unique peptides corresponding to all proteins in a proteome. The use of binary search program significantly reduces the time for frequency chart preparation to ~2 s for a proteome comprising ~23 000 proteins. The algorithm was applied to a three‐protein mixture identification, host cell protein (HCP) analysis, and a simulation‐generated data set. It was found that the algorithm could identify at least one unique peptide of a protein even in the presence of fourfold higher concentration of another protein. In addition, two HCPs that are known to be difficult to remove were missed by MS/MS approach and were exclusively identified using the presented algorithm. Thus, the proposed algorithm when used along with standard proteomic approaches present avenues for enhanced protein identification efficiency, particularly for applications such as HCP analysis in biopharmaceutical research, where identification of low‐abundance proteins are generally not achieved due to dynamic range limitations between the target product and HCPs.  相似文献

The conversion of aldehydes into carboxylic acids using oxidizing agents is a common protocol in transformation chemistry. An efficient oxidation strategy of transformation of pyrazole‐4‐aldehydes to the corresponding acids using vanadium catalysts in the presence of 30% H2O2 as an oxidant is described. The catalytic technology was successfully applied to a range of various 4‐formylpyrazoles, and plausible mechanism is also discussed.  相似文献

The development of organic photoluminescent materials, which show promising roles as catalysts, sensors, organic light-emitting diodes, logic gates, etc., is a major demand and challenge for the global scientific community. In this context, a photoclick polymerization method is adopted for the growth of a unique photoluminescent three-dimensional (3D) polymer film, E, as a model system that shows emission tunability over the range 350–650 nm against the excitation range 295–425 nm. The DFT analysis of energy calculations and π-stacking supports the spectroscopic observations for the material exhibiting a broad range of emission owing to newly formed chromophoric units within the film. Full polarization spectroscopic Mueller matrix studies were employed to extract and quantify the molecular orientational order of both the ground (excitation) and excited (emission) state anisotropies through a set of newly defined parameters, namely the fluorescence diattenuation and fluorescence polarizance. The information contained in the recorded fluorescence Mueller matrix of the organic polymer material provided a useful way to control the spectral intensity of emission by using pre- and post-selection of polarization states. The observation was based on the assumption that the longer lifetime of the excited dipolar orientation is attributed to the compactness of the film.  相似文献

This article focuses on the synthesis and the crystal chemistry of six members of a series of rare-earth metal based germanides with general formula RELiGe(2) (RE = La-Nd, Sm, and Eu). The structures of these compounds have been established by single-crystal X-ray diffraction (CaLiSi(2) structure type, space group Pnma, Z = 4, Pearson symbol oP16). The chemical bonding within this atomic arrangement can be rationalized in terms of anionic germanium zigzag chains, conjoined via chains of edge-shared LiGe(4) tetrahedra and separated by rare-earth metal cations. The structure can also be viewed as an intergrowth of AlB(2)-like and TiNiSi-like fragments, or as the result of the replacement of 50% of the rare-earth metal atoms by lithium in the parent structure of the REGe monogermanides. Except for LaLiGe(2) and SmLiGe(2), the remaining four RELiGe(2) phases exhibit Curie-Weiss paramagnetism above about 50 K. In the low temperature regime, the localized 4f electrons in CeLiGe(2), PrLiGe(2), and SmLiGe(2) order ferromagnetically, while antiferromagnetic ordering is observed for NdLiGe(2) and EuLiGe(2). The calculated effective magnetic moments confirm RE(3+) ground states in all cases excluding EuLiGe(2), in which the magnetic response is consistent with Eu(2+) configuration (J = S = 7/2). The experimental results have been complemented by tight-binding linear muffin-tin orbital (TB-LMTO) band structure calculations.  相似文献

Magnesium aluminate nanoparticles with different chromium concentration (0–12%) have been synthesized by a citrate–nitrate sol–gel route. X-ray diffraction studies confirmed the formation of single-phase cubic spinel structure excluding the presence of any secondary phase. Crystallite size of the synthesized nanoparticles was found to increase from 8.5 to 19.8 nm with the increase in Cr concentration. Fourier transformed infrared spectroscopic studies confirmed the presence of AlO6 group which led to the formation of MgAl2O4 spinel structure. Surface morphology of the sintered pellets was investigated with the help of a field emission scanning electron microscope which revealed the existence of both grain and grain boundary along with their aggregates. The dielectric constant, dielectric loss and ac conductivity were studied as a function of frequency of the applied electric field for different composition and their nature of variation with frequency has been elucidated on the basis of Maxwell–Wagner interfacial model. Impedance spectroscopy technique has been used to study the effect of grain and grain boundary on the electrical properties of this spinel oxide. All the electrical parameters showed strong dependence on the nanostructural properties and were found to vary consistently with the increase of doping concentration.  相似文献

Reported here are details on the discovery of a phosphine-catalyzed isomerization of hemiketals and subsequent reactions of the cyclic keto enol ether products. The new cycloisomerization complements a previously reported amine-catalyzed process that gave oxepinones from the same hemiketal starting materials. In the absence of functionality (R(2)) on the cyclic keto enol ether, a rapid and facile dimerization occurs, giving spiroketal products. When the enone is substituted (i.e., R(2) = Ph), the cyclic keto enol ether is sufficiently stable so that it can be isolated; it can then be further reacted in the same pot to provide the corresponding dihydropyrazoles. Both the spiroketal and dihydropyrazole products arise by a tandem reaction that begins with the novel cycloisomerization. The method allows for the rapid introduction of complexity in the products from relatively simple starting materials. It should find application in the synthesis of natural product-like molecules.  相似文献

This review narrates the electron transfer reactions of various nickel(III) and nickel(IV) complexes reported during the period 1981 until today. The reactions have been categorized mainly with respect to the type of nickel complexes. The reactivity of nickel(III) complexes of macrocycles, 2,2′-bipyridyl and 1,10-phenanthroline, peptides and oxime–imine, and of nickel(IV) complexes derived from oxime–imine, oxime and miscellaneous ligands with various organic and inorganic electron donors have been included. Kinetic and mechanistic features associated with such interactions have been duly analyzed. The relevance of Marcus cross-relation equations in the delineation of the electron transfer paths has also been critically discussed.  相似文献
