Comprehensive two-dimensional HPLC (2DLC) has been used very successfully in proteomics applications for over a decade. Increasingly we are seeing online, comprehensive 2DLC used for non-proteomic applications. This article gives an overview of the state of the art of this technique, with emphasis on current trends in theory and practice that are exciting and hold the most promise for advancing the performance of 2DLC. Specifically, the recently commercialized small, superficially porous packing materials are very well suited to use in the second dimension of online 2DLC where analysis speed is critical and largely dictates the performance of the 2DLC system. Further, the recent development of optimization schemes and associated software that support the use of different second-dimension elution modes in a single 2DLC analysis will improve the flexibility and effectiveness of 2DLC separations. Excluding separations of peptides, proteins, and polymers, 2DLC systems utilizing reversed-phase separation in one dimension, and either normal phase or reversed-phase in the other dimension continue to be the most popular. Although these systems have been applied mostly to complex biological materials, we are beginning to see applications in the analysis of pharmaceutical materials for ingredient purity and degradation profiling. The general lack of robust, easy-to-use commercially available software is arguably the greatest impediment to wider application of 2DLC methods. This situation is improving slowly, however, and at least two commercially available software packages have been described in the peer-reviewed literature. This is an exciting time in the development of online 2DLC.  相似文献

The paper describes a study of the kinetics and mechanism of the coagulation of two types of immunoassays using sensitized latexes. The positive response to the first test is based on the aggregation of the gamma globulin (IgG)-coated polystyrene latexes in the presence of IgM rheumatoid antigen. The second test is relative to the heteroaggregation of two types of sensitized latexes induced by the presence of human chorionic gonadotropin (HCG). In the latter test, two identical polystyrene latexes bearing carboxylic acid surface groups were sensitized by covalent coupling of monoclonal antibodies specific for the αHCG determinant on one type of latex and for the βHCG determinant on the other type. Using the Coulter Counter method, the aggregate size distribution c(n) was determined as a function of the number n of elementary constituents, thus enabling calculation of the number N(t) and weight S(t) average sizes of the aggregates. The temporal variations of the average sizes were compared with typical situtions of reaction-limited aggregation processes in order to characterize the mechanism of aggregation induced by antibody–antigen reactions.  相似文献

Effect of Substituents and Anions on the Fluorescence Properties of Mixed-Ligand (Amine-triphenylphosphane) Copper(I) Halides Mixed-ligand complexes analyzing CuHal · PΦ3 · Py? R where Hal = Cl?, Br?, I? and where R are various substituents at the pyridine (Py) exhibit an intense solid state fluorescence depending on substituent and anion when excited with uv radiation (365 nm). Starlike poly (oxypropylene) molecules (M=2630 resp. 710 g/mol), each with three OH-end-groups, are crosslinked with 4,4-diphenylmethane diisocyanate in one stoichiometric and several nonstoichiometric reactions. Dynamic mechanical (frequency 10–4 to 102 Hz) and also some dielectric (10–2 to 106 Hz) measurements were made on these networks in a wide temperature range. The time-temperature-superposition principle was used to obtain master curves. Two large relaxation processes were detected (separated by many decades of frequency in some samples). The high frequency process seems to correspond to the glass-rubber transition in linear polymers, the low frequency process is probably due to the relaxation of dangling chains.  相似文献
