Summary The kinetics of oxidation of TeIV by CoIII have been studied in aqueous HClO4. A mechanism presuming [Co(OH2)5(OH)]2+ to be the reactive species has been proposed, which leads to the rate-equation shown. Rate=–d[CoIII]/dt=2kKK h 2 [CoIII] t 2 [TeIV]/[H+]2 Kb is the hydrolysis constant of CoIII, K is the formation constant of the complex between CoIII and TeIV and k is the rate of decomposition of that complex. Ea and S are 95.0±2.1 kJ mol–1 and 28.3±7.1 JK–1 mol–1, respectively.  相似文献

Cytochrome P450 2D6 (CYP2D6) is used to develop an approach for predicting affinity and relevant binding conformation(s) for highly flexible binding sites. The approach combines the use of docking scores and compound properties as attributes in building a neural network (NN) model. It begins by identifying segments of CYP2D6 that are important for binding specificity, based on structural variability among diverse CYP enzymes. A family of distinct, low-energy conformations of CYP2D6 are generated using simulated annealing (SA) and a collection of 82 compounds with known CYP2D6 affinities are docked. Interestingly, docking poses are observed on the backside of the heme as well as in the known active site. Docking scores for the active site binders, along with compound-specific attributes, are used to train a neural network model to properly bin compounds as strong binders, moderate binders, or nonbinders. Attribute selection is used to preselect the most important scores and compound-specific attributes for the model. A prediction accuracy of 85+/-6% is achieved. Dominant attributes include docking scores for three of the 20 conformations in the ensemble as well as the compound's formal charge, number of aromatic rings, and AlogP. Although compound properties were highly predictive attributes (12% improvement over baseline) in the NN-based prediction of CYP2D6 binders, their combined use with docking score attributes is synergistic (net increase of 23% above baseline). Beyond prediction of affinity, attribute selection provides a way to identify the most relevant protein conformation(s), in terms of binding competence. In the case of CYP2D6, three out of the ensemble of 20 SA-generated structures are found to be the most predictive for binding.  相似文献

Reactive carbonyl species (RCS) may originate from the oxidation of unsaturated fatty acids and sugar in conditions of pathology. They are known to have high reactivity towards DNA as well as nucleophilic sites of proteins, resulting in cellular dysfunction. It has been considered that various pathological conditions are associated with an increased level of RCS and their reaction products. Thus, regulating the levels of RCS may be associated with the mitigation of various metabolic and neurodegenerative disorders. In order to perform a comprehensive review, various literature databases, including MEDLINE, EMBASE, along with Google Scholar, were utilized to obtain relevant articles. The voluminous review concluded that various synthetic and natural agents are available or in pipeline research that hold tremendous potential to be used as a drug of choice in the therapeutic management of metabolic syndrome, including obesity, dyslipidemia, diabetes, and diabetes-associated complications of atherosclerosis, neuropathy, and nephropathy. From the available data, it may be emphasized that various synthetic agents, such as carnosine and simvastatin, and natural agents, such as polyphenols and terpenoids, can become a drug of choice in the therapeutic management for combating metabolic syndromes that involve RCS in their pathophysiology. Since the RCS are known to regulate the biological processes, future research warrants detailed investigations to decipher the precise mechanism.  相似文献

The Curcuma longa plant is endowed with multiple traditional and therapeutic utilities and is here explored for its phytochemical constituents and cytotoxic potential. Turmeric rhizomes were extracted from three different solvents and screened for the presence of different phytochemical constituents, observation of which indicated that the polar solvents favoured extraction of greater versatile phytochemical constituents. These extracts were investigated for their cytotoxic potential by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay on three different of cell lines including SCC-29B (oral cancer cell line), DU-145 (prostate cancer cell line) and the Vero cell line (healthy cell line/non-cancerous cell line). This assay was performed by taking three extracts from isolated curcuminoids and a pure bioactive compound bisdemethoxycurcumin (BD). Bisdemethoxycurcumin was isolated from curcuminoids and purified by column and thin-layer chromatography, and its structural characterisation was performed with different spectroscopic techniques such as FTIR, NMR (1H Proton and 13C Carbon-NMR) and LC-MS. Amongst the extracts, the ethanolic extracts exhibited stronger cytotoxic potential against the oral cancer cell line (SCC-29B) with an IC50value of 11.27 μg/mL, and that this was too low of a cytotoxicity against the Vero cell line. Although, curcuminoids have also shown a comparable cytotoxic potential against SCC-29B (IC50 value 16.79 μg/mL), it was not as potent against the ethanolic extract, and it was even found to be cytotoxic against healthy cell lines at a very low dose. While considering the isolated compound, bisdemethoxycurcumin, it also possessed a cytotoxic potential against the prostate cancer cell line (DU-145) (IC50 value of 93.28 μg/mL), but was quite safe for the healthy cell line in comparison to doxorubicin.  相似文献

Size effect of silver nano particles on the photophysical properties of 2,3-bis(chloromethyl)anthracene-1,4,9,10-tetraone (BCMAT) have been investigated using optical absorption and fluorescence emission techniques. Silver NPs of different sizes have been prepared by two different methods. Quenching of fluorescence of BCMAT has been found to increase with decrease in the size of the silver NPs. Convenient synthesis of a new square planar nickel(II) naphthaldehyde thiosemicarbazone complex has been described. The composition of the complex has been established by elemental analysis, spectral methods, and single crystal X-ray crystallography. The new complex acts as an active homogeneous catalyst for the Mizoroki–Heck reaction of electron deficient (activating) and electron rich (deactivating) aryl bromides with various olefins under optimized conditions.  相似文献

The ligational behavior of meso-arylsapphyrins and rubyrins toward Rh(I) is investigated. Sapphyrins form monometallic complexes with coordination of one imine and amine type nitrogens of the bipyrrole unit in an eta2 fashion. The Rh(I) coordination is completed by the presence of two ancillary carbon monoxide ligands. Rubyrins form both monometallic and bimetallic complexes. Two types of bimetallic complexes have been isolated. In the first type, both rhodium atoms are projected above the mean rubyrin plane, while in the second type, one rhodium atom is projected above and the other below the mean plane. Detailed 1H and 2D NMR spectral analyses along with IR and UV-visible spectra of the complexes confirm the proposed binding modes for the rhodium complexes. Furthermore, the single-crystal X-ray analysis of one of the bimetallic complexes of rubyrin shows a bowl-shaped symmetric structure where both Rh(I) atoms are projected above the mean rubyrin plane at an angle of 71.73 degrees. The geometry around each rhodium center is approximately square planar [N1-Rh1-N2, 80.38(9) degrees; C15-Rh1-C16, 86.95(14) degrees; N1-Rh1-C15, 97.13(12) degrees; and N2-RH1-C16, 94.97(12) degrees ]. The omicronbserved distance of 4.313 A between the two rhodium centers reveals very little interaction between the two rhodium atoms. This type of metal binding is accompanied by a 180 degrees ring flip of the heterocyclic ring connecting the two bipyrrole units. In dioxarubyrin, where one of the pyrrole rings of the bipyrrole unit is inverted, Rh(I) binds at the periphery to the pyrrole nitrogen, leaving the rubyrin cavity empty. The absence of one amino and one imino nitrogen on the dipyrromethene subunits in the sapphyrins and rubyrins described here forces Rh(I) to bind to bipyrrole nitrogens.  相似文献

The synthesis and characterization of five hydrogen-bonded ferroelectric liquid crystal complexes (HBFLCs) prepared from mesogenic p-n-alkoxy benzoic acids and non-mesogenic propionic/butyric acids with different chiral centres are reported. Complementary intermolecular hydrogen bonding is confirmed through IR study. HBFLCs are found to exhibit chiral nematic (N*), smectic C* (SmC*) and smectic G* (monotropic) phases in their cooling profiles during polarizing thermal microscopy and differential scanning calorimetry. Phase coexistence regions are observed above the IN* transition. The chiral nematic to smectic C* transition is found to be of first order. The temperature variation of spontaneous polarization exhibited by these HBFLC complexes in their SmC* phase is presented. The effect of non-covalent interaction imparted by the soft hydrogen bonding in these LC complexes on enhanced or induced thermal stability of tilted LC phases is discussed.  相似文献

A double substrate uncatalyzed bromate oscillator is reported. Oscillations occur for several minutes, the color of the solution alternating between dark brown and reddish yellow. The oscillatory wave form (Pt electrode) has a distinct pattern, with a progressive increase in the amplitude and time interval. The presence of both substrates namely 8-hydroxyquinoline and malonic acid is essential. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献
