We present a new chronoamperometric methodology for the indirect determination of pepsin activity in proteic solutions. The high-sensitive polyphenol oxidase (PPO) carbon paste bioelectrodes, prepared with 150–200 U of PPO adsorbed on the carbon paste surface and trapped behind a dialysis membrane were able to detect free -tyrosine ( -Tyr) as well as -Tyr-containing peptides, the product of pepsin hydrolysis on haemoglobin samples. The electroactive enzymatic products were determined by their reduction current at −0.050 V, a working potential low enough for preventing electrochemical interferences. The external diffusional barrier posed by the dialysis membrane and the high PPO-superficial concentrations allowed us to prevent electrode surface fouling and to reach stationary diffusional currents after 3 min. The electrochemical -Tyr concentrations measured with proper internal standards, were correlated to those determined by the spectrophotometric reference method (Folin–Ciocalteu), reaching a linear relationship with a slope of 1.05±0.04 and a linear regression coefficient of 0.9951. These results as well as a recovery assay of 98% and the detection limit lower than that of the spectrophotometric method indicate that the proposed methodology is very satisfactory and potentially useful for indirect determination of pepsin in biological samples. The technique was also applied for the determination of pepsin activity in hydrolysed immuno globulin solutions of different composition.  相似文献

The influence of pulse duration on the laser-induced damage in undoped or infrared-absorbing-dye doped thin triazenepolymer films on glass substrates has been investigated for single, near-infrared (800 nm) Ti:sapphire laser pulses with durations ranging from 130 fs up to 540 fs and complementarily for infrared (1064 nm) Nd:YAG ns-laser single-pulse irradiation. The triazenepolymer material has been developed for high resolution ablation with irradiation at 308 nm. Post-irradiation optical microscopy observations have been used to determine quantitatively the threshold fluence for permanent laser damage. In contrast to our previous studies on a triazenepolymer with different composition [J. Bonse, S.M. Wiggins, J. Solis, T. Lippert, Appl. Surf. Sci. 247 (2005) 440], a significant dependence of the damage threshold on the pulse duration is found in the sub-picosecond regime with values ranging from ∼500 mJ/cm2 (130 fs) up to ∼1500 mJ/cm2 (540 fs). Other parameters such as the film thickness (50 nm and 1.1 μm samples) or the doping level show no significant influence on the material behavior upon irradiation. The results for fs- and ns-laser pulse irradiation are compared and analyzed in terms of existent ablation models.  相似文献

We have identified addimer chain structures as metastable precursors to compact epitaxial islands on the (2 × n) reconstructed SiGe wetting layer, using polarity-switching scanning tunneling microscopy (STM). These chain structures are comprised of 2-12 addimers residing in the troughs of neighboring substrate dimer rows. The chain structures extend along equivalent 〈1 3 0〉 directions across the substrate dimer rows in a zigzag fashion, giving rise to kinked and straight segments. We measure a kink-to-straight ratio of nearly 2:1. This ratio corresponds to a free energy difference of 17 ± 4 meV, favoring the formation of kinked segments. The chain structures convert to compact epitaxial islands at elevated temperatures (?90 °C). This conversion suggests that the chain structures are a precursor for compact island formation on the SiGe wetting layer. We digitally process filled- and empty-state STM images to distinguish chain structures from compact islands. By monitoring the populations of both species over time, the chain-to-island conversion rates are measured at substrate temperatures ranging from 90 to 150 °C. The activation energy for the conversion process is measured to be 0.7 ± 0.2 eV with a corresponding pre-exponential factor of 5 × 104±2 s−1.  相似文献

Micelles, vesicles, and films composed of two species of incompatible heterogeneous molecules exhibit full internal segregation of the component species. This macroscopic segregation can be inhibited by oppositely charging the two different molecular species. The degree of compatibility achieved by the charges leads to either fully homogenous mixtures or to local segregation and the possible formation of regular patterns. We investigate the induction of periodic surface patterns by the presence of opposite charges in flat films and cylindrical micelles. In the strong segregation limit the incompatibility between species can be described by a line tension parameter gamma. The size of the patterns formed is of the order of a characteristic size L approximately (gamma/sigma(2))(1/2), where sigma is the surface charge density. The pattern symmetry on flat surfaces is function only of the fraction of area covered by the components, f: lamellar for 0.34相似文献

Femtosecond laser pulse temporal shaping techniques have led to important advances in different research fields like photochemistry, laser physics, non-linear optics, biology, or materials processing. This success is partly related to the use of optimal control algorithms. Due to the high dimensionality of the solution and control spaces, evolutionary algorithms are extensively applied and, among them, genetic ones have reached the status of a standard adaptive strategy. Still, their use is normally accompanied by a reduction of the problem complexity by different modalities of parameterization of the spectral phase. Exploiting Rabitz and co-authors’ ideas about the topology of quantum landscapes, in this work we analyze the optimization of two different problems under a deterministic approach, using a multiple one-dimensional search (MODS) algorithm. In the first case we explore the determination of the optimal phase mask required for generating arbitrary temporal pulse shapes and compare the performance of the MODS algorithm to the standard iterative Gerchberg–Saxton algorithm. Based on the good performance achieved, the same method has been applied for optimizing two-photon absorption starting from temporally broadened laser pulses, or from laser pulses temporally and spectrally distorted by non-linear absorption in air, obtaining similarly good results which confirm the validity of the deterministic search approach.  相似文献

Two‐dimensional hybrid perovskites are used as absorbers in solar cells. Our first‐generation devices containing (PEA)2(MA)2[Pb3I10] ( 1 ; PEA=C6H5(CH2)2NH3+, MA=CH3NH3+) show an open‐circuit voltage of 1.18 V and a power conversion efficiency of 4.73 %. The layered structure allows for high‐quality films to be deposited through spin coating and high‐temperature annealing is not required for device fabrication. The 3D perovskite (MA)[PbI3] ( 2 ) has recently been identified as a promising absorber for solar cells. However, its instability to moisture requires anhydrous processing and operating conditions. Films of 1 are more moisture resistant than films of 2 and devices containing 1 can be fabricated under ambient humidity levels. The larger bandgap of the 2D structure is also suitable as the higher bandgap absorber in a dual‐absorber tandem device. Compared to 2 , the layered perovskite structure may offer greater tunability at the molecular level for material optimization.  相似文献

The standard topological censorship theorems require asymptotic hypotheses which are too restrictive for several situations of interest. In this paper we prove a version of topological censorship under significantly weaker conditions, compatible, e.g., with solutions with Kaluza–Klein asymptotic behavior. In particular we prove simple connectedness of the quotient of the domain of outer communications by the group of symmetries for models which are asymptotically flat, or asymptotically anti-de Sitter, in a Kaluza–Klein sense. This allows one, e.g., to define the twist potentials needed for the reduction of the field equations in uniqueness theorems. Finally, the methods used to prove the above are used to show that weakly trapped compact surfaces cannot be seen from Scri. Gregory J. Dimethyl fumarate is a cytoprotective and immunomodulatory drug used in the treatment of multiple sclerosis. We performed a bibliometric study examining the characteristics and trends of the top 100 cited articles that include dimethyl fumarate in the title. On 21 September 2020 we carried out an electronic search in the Web of Science (WOS), seeking articles that include the following terms within the title: dimethyl fumarate, BG-12, or Tecfidera. To focus our investigation on original research, we refined the search to include only articles, early access, others, case report, and clinical trials. We obtained a total of 1115 items, which were cited 7169 times, had a citation density of 6.43 citations/item, and an h-index of 40. Around 2010, there was a jump in the number of published articles per year, rising from 5 articles/year up to 12 articles/year. We sorted all the items by the number of citations and selected the top 100 most cited (T100). The T100 had 4164 citations, with a density of 37 citations/year and contained 16 classic research articles. They were published between 1961 and 2018; the years 2010–2018 amassed nearly 80% of the T100. We noted 17 research areas with articles in the T100. Of these, the number one ranking went to neurosciences/neurology with 39 articles, and chemistry ranked second on the T100 list with 14 items. We noticed that the percentage of articles belonging to different journals changed depending on the time period. Chemistry held the highest number of papers during 1961–2000, while pharmacology andneurosciences/neurology led the 2001–2018 interval. A total of 478 authors from 145 institutions and 25 countries were included in the T100 ranking. The paper by Gold R et al. was the most successful with 14 articles, 1.823 citations and a density of 140.23 citations/year. The biotechnological company Biogen led the T100 list with 20 articles. With 59 published articles, the USA was the leading country in publications. We concluded that this study analyzed the use of and research on dimethyl fumarate from a different perspective, which will allow the readership (expert or not) to understand the relevance of classic and recent literature on this topic.  相似文献

We consider a parallel fiber‐reinforced periodic elastic composite that present an imperfect contact of spring type between the fiber and the matrix. We use the elliptic integral of Cauchy type for solving the plane strain local problems that arise from the asymptotic homogenization method. Several general conditions are assumed, which include that the fibers are disposed of arbitrary manner in the local cell, that all fibers present contact perfect with different constants of imperfection, and that their cross section is a smooth closed arbitrary curve. We find that there are infinity solutions for these problems, and we find relations between these solutions and effective coefficients of the composite. We consider a model for periodic patterns of charges constrained over a cylindrical surface. In particular we focus on patterns of chiral helices, achiral rings, or vertical lamellae, with the constraint of global electroneutrality. We study the dependence of the patterns' size and pitch angle on the radius of the cylinder and salt concentration. We obtain a phase diagram by using numerical and analytic techniques. For pure Coulomb interactions, we find a ring phase for small radii and a chiral helical phase for large radii. At a critical salt concentration, the characteristic domain size diverges, resulting in an achiral macroscopic phase-segregated structure. We discuss possible consequences and generalizations of our model.  相似文献
