Reactive nitrogen species such as peroxynitrite can nitrate specific amino acids, whether free or protein bound, and 3-nitrotyrosine is believed to be one marker of this reaction. To examine the significance of this pathway in biological systems we have developed an accurate, sensitive, and specific assay for 3-nitrotyrosine based on combined liquid chromatography tandem mass spectrometry. Our approach allowed simultaneous analysis of both tyrosine and 3-nitrotyrosine and employs isotopomer standards (i.e., [15N1, 13C9]-tyrosine and [13C6]-3nitrotyrosine). Calibration curves were linear (r2 = 0.999) across the range 0.5-100 pg/microL (i.e., 2.2-442 fmol/microL), and the detection limit for standard samples was 0.5 pg/microL (2.2 fmol/microL, or 10 fmol on column; S/N = 5) or 1 pg/microL (4.4 fmol/microL) for extracted (biological) samples. As a component of this study we have undertaken an extensive investigation of artifactual formation of 3-nitrotyrosine under conditions that exist during sample extraction and derivatization. Our studies show that under appropriate conditions (low pH, elevated temperatures, and in the presence of a vast excess of the two substrates, tyrosine and the nitrate anion), 3-nitrotyrosine can readily be formed as an artifact.  相似文献

Cationic complexes of the type fac-[(L(2))Pt(IV)Me(3)(pyr-X)][OTf] (pyr-X = 4-substituted pyridines; L(2) = diphosphine, viz., dppe = bis(diphenylphosphino)ethane and dppbz = o-bis(diphenylphosphino)benzene; OTf = trifluoromethanesulfonate) undergo C-C reductive elimination reactions to form [L(2)Pt(II)Me(pyr-X)][OTf] and ethane. Detailed studies indicate that these reactions proceed by a two-step pathway, viz., initial reversible dissociation of the pyridine ligand from the cationic complex to generate a five-coordinate Pt(IV) intermediate, followed by irreversible concerted C-C bond formation. The reaction is inhibited by pyridine. The highly positive values for DeltaS()(obs) = +180 +/- 30 J K(-1) mol(-1), DeltaH(obs) = 160 +/- 10 kJ mol(-1), and DeltaV()(obs) = +16 +/- 1 cm(3) mol(-1) can be accounted for in terms of significant bond cleavage and/or partial reduction from Pt(IV) to Pt(II) in going from the ground to the transition state. These cationic complexes have provided the first opportunity to carry out detailed studies of C-C reductive elimination from cationic Pt(IV) complexes in a variety of solvents. The absence of a significant solvent effect for this reaction provides strong evidence that the C-C reductive coupling occurs from an unsaturated five-coordinate Pt(IV) intermediate rather than from a six-coordinate Pt(IV) solvento species.  相似文献

The Rh(III) complexes of substituted ethylenediamines are observed to be catalytically active at the DME and studies of the effects of human serum proteins on this activity are reported. Rh(III) complex waves are shown to respond to six different proteins, albumin, α-, β-, γ-globulin, mercaptoalbumin, and glycoprotein at trace concentration levels. The effect gives linear calibration for all proteins in the range 0.1–0.8 μg ml?1 with sensitivity in the order glycoprotein >albumin>γ-globulin > β-globulin > α-globulin > mercaptoalbumin at pH 8. Important factors controlling sensitivity are buffer composition and pH, and Rh complex structure and concentration, and at pH 9.2, almost equal response of proteins is obtained. The method for protein analysis is compared to the Brdicka method and found to have several advantages in terms of sensitivity, pH range, response of differing proteins and wave resolution.  相似文献

Insertion of molecular oxygen into a palladium(II) hydride bond to form an (eta1-hydroperoxo)palladium(II) complex is reported. The hydroperoxo palladium(II) product has been crystallographically characterized. A second-order rate law (first-order in palladium and first-order in oxygen) is observed for the reaction and a large kinetic isotope effect implicates Pd-H bond cleavage in the rate-determining step. The results of studies with radical inhibitors and light suggest that the reaction does not proceed by a radical chain mechanism.  相似文献

Red-black [HIPTN3N]Cr (1) ([HIPTN3N]3- = [(HIPTNCH2CH2)3N]3- where HIPT = 3,5-(2,4,6-i-Pr3C6H2)2C6H3 = HexaIsoPropylTerphenyl) can be prepared from CrCl3, while green-black [HIPTN3N]Cr(THF) (2) can be prepared from CrCl3(THF)3. Reduction of {1|2} (which means either 1 or 2) with potassium graphite in ether at room temperature yields [HIPTN3N]CrK (3) as a yellow-orange powder. There is no evidence that dinitrogen is incorporated into 1, 2, or 3. Compounds that can be prepared readily from {1|2} include red [HIPTN3N]CrCO (4), blood-red [HIPTN3N]CrNO (6), and purple [HIPTN3N]CrCl (7, upon oxidation of {1|2} with AgCl). The dichroic (purple/green) Cr(VI) nitride, [HIPTN3N]CrN (8) was prepared from Bu4NN3 and 7. X-ray studies have been carried out on 4, 6, and 7, and on two co-crystallized compounds, 7 and [HIPTN3N]CrN3 (65:35) and [HIPTN3N]CrN3 and 8 (50:50). Exposure of a degassed solution of {1|2} to an atmosphere of ammonia does not yield "Cr(NH3)" as a stable and well-behaved species analogous to Mo(NH3). An attempt to reduce dinitrogen under conditions described for the catalytic reduction of dinitrogen by [HIPTN3N]Mo compounds with 8 yielded a substoichiometric amount (0.8 equiv) of ammonia, which suggests that some ammonia is formed from the nitride but none is formed from dinitrogen.  相似文献

Head-to-tail cyclic peptides have been reported to bind to multiple, unrelated classesof receptor with high affinity. They may therefore be considered to beprivileged structures. This review outlines the strategies by which bothmacrocyclic cyclic peptides and cyclic dipeptides or diketopiperazines havebeen synthesised in combinatorial libraries. Ammonia-borane (NH(3)BH(3), AB) has garnered interest as a hydrogen storage material due to its high weight percent hydrogen content and ease of H(2) release relative to metal hydrides. As a consequence of dehydrogenation, B-N-containing oligomeric/polymeric materials are formed. The ability to control this process and dictate the identity of the generated polymer opens up the possibility of the targeted synthesis of new materials. While precious metals have been used in this regard, the ability to construct such materials using earth-abundant metals such as Fe presents a more economical approach. Four Fe complexes containing amido and phosphine supporting ligands were synthesized, and their reactivity with AB was examined. Three-coordinate Fe(PCy(3))[N(SiMe(3))(2)](2) (1) and four-coordinate Fe(DEPE)[N(SiMe(3))(2)](2) (2) yield a mixture of (NH(2)BH(2))(n) and (NHBH)(n) products with up to 1.7 equiv of H(2) released per AB but cannot be recycled (DEPE = 1,2-bis(diethylphosphino)ethane). In contrast, Fe supported by a bidentate P-N ligand (4) can be used in a second cycle to afford a similar product mixture. Intriguingly, the symmetric analogue of 4 (Fe(N-N)(P-P), 3), only generates (NH(2)BH(2))(n) and does so in minutes at room temperature. This marked difference in reactivity may be the result of the chemistry of Fe(II) vs Fe(0).  相似文献

Cytostatic agents that interfere with specific cellular components to prevent cancer cell growth offer an attractive alternative, or complement, to traditional cytotoxic chemotherapy. Here, we describe the synthesis and characterization of a new binuclear RuII–PtII complex [Ru(tpy)(tpypma)Pt(Cl)(DMSO)]3+ (tpy=2,2′:6′,2′′‐terpyridine and tpypma=4‐([2,2′:6′,2′′‐terpyridine]‐4′‐yl)‐N‐(pyridin‐2‐ylmethyl)aniline), VR54, which employs the extended terpyridine tpypma ligand to link the two metal centres. In cell‐free conditions, VR54 binds DNA by non‐intercalative reversible mechanisms (Kb=1.3×105 M ?1) and does not irreversibly bind guanosine. Cellular studies reveal that VR54 suppresses proliferation of A2780 ovarian cancer cells with no cross‐resistance in the A2780CIS cisplatin‐resistant cell line. Through the preparation of mononuclear RuII and PtII structural derivatives it was determined that both metal centres are required for this anti‐proliferative activity. In stark contrast to cisplatin, VR54 neither activates the DNA‐damage response network nor induces significant levels of cell death. Instead, VR54 is cytostatic and inhibits cell proliferation by up‐regulating the cyclin‐dependent kinase inhibitor p27KIP1 and inhibiting retinoblastoma protein phosphorylation, which blocks entry into S phase and results in G1 cell cycle arrest. Thus, VR54 inhibits cancer cell growth by a gain of function at the G1 restriction point. This is the first metal‐coordination compound to demonstrate such activity.  相似文献
