Previous mode of action studies identified methionine aminopeptidase 2 (MetAP-2) as the target of the antiangiogenic natural product fumagillin and its drug candidate analog, TNP-470. We report here that TNP-470-mediated MetAP-2 inhibition blocks noncanonical Wnt signaling, which plays a critical role in development, cell differentiation, and tumorigenesis. Consistent with this finding, antisense MetAP-2 morpholino oligonucleotide injection in zebrafish embryos phenocopies gastrulation defects seen in noncanonical Wnt5 loss-of-function zebrafish mutants. MetAP-2 inhibition or depletion blocks signaling downstream of the Wnt receptor Frizzled, but upstream of Calmodulin-dependent Kinase II, RhoA, and c-Jun N-terminal Kinase. Moreover, we demonstrate that TNP-470 does not block the canonical Wnt/beta-catenin pathway. Thus, TNP-470 selectively regulates noncanonical over canonical Wnt signaling and provides a unique means to explore and dissect the biological systems mediated by these pathways. 相似文献

The effects of flame retardants such as hydrated aluminium oxide, antimony trioxide and chloroparaffin on the thermal properties and flammability of sulphur vulcanizates of butyl and halogenated butyl elastomers were studied. The thermoanalytical curves of the elastomers were interpreted. Greater tendencies to thermal degradation were observed for halogenated butyl elastomers than for the original butyl rubber elastomer. This was confirmed by elastomer combustibility studies. The use of these flame retardants allowed the formation of self-extinguishing vulcanizates of the investigated elastomers. 相似文献

Orientation dependent wetting of InP substrates by In- and InxSn1−x melts saturated with InP was studied using the sessile drop method. The contact angle was found to depend on the substrate orientation as follows: It is shown that a tin admixture to the indium melt improves the wetting behaviour. Additional to the dependence on substrate orientation it was found an anisotropic wetting on all investigated faces. 相似文献
