Technical improvements in protein microsequencing

Improved techniques for preparing samples and sequencing reagents have been developed to achieve highly sensitive protein sequencing. These improvements include a new sequencing buffer system which gives extremely low chromatographic background, a method for preparing polybrene, a key chemical in gas-phase protein microsequencing, and techniques for preparing protein samples suitable for microsequencing.     

Extremal points and optimal solutions for general capacity problems

This paper studies the infinite dimensional linear programming problems in the integration type. The variable is taken in the space of bounded regular Borel measures on compact Hausdorff spaces. It will find an optimal measure for a constrained optimization problem, namely a capacity problem. Relations between extremal points of the feasible region and optimal solutions of the optimization problem are investigated. The necessary/sufficient conditions for a measure to be optimal are established. The algorithm for optimal solution of the general capacity problem onX = Y = [0, 1] is formulated.     

Multiplicity Free Actions of Quantum Groups and Generalized Howe Duality

Noncommutative associative algebras are constructed which have the structure of module algebras over tensor products of pairs of quantized universal enveloping algebras. These module algebras decompose into multiplicity free direct sums of irreducible modules, yielding quantum analogues of generalized Howe dualities.     

A New Kind of Trivariate C 1 Macro-element

We propose a construction of a trivariate C ¹ macro-element over a special tetrahedral partition and compare our construction with known C ¹ macro-elements which are summarized in this paper. Also, we propose an improvement of the Alfeld construction of a C ¹ quintic macro-element such that the new scheme is able to reproduce all polynomials of total degree ≤5.     

Chiral resolution,absolute configuration determination,and stereo-activity relationship study of IDO1 inhibitor NLG919

NLG919 (1) with two chiral carbon atoms on its chemical structure is a potent indoleamine 2,3-dioxygenase 1 (IDO1) inhibitor. We developed an effective way to prepare all stereoisomers of 1, the key step being the chiral resolution of racemic intermediate 2. The optimal resolution solvent system was identified as dichloromethane and n-pentane or petroleum ether. Using (?)-di-p-toluoyl-d-tartaric acid as resolution reagent, optical pure (R)-2 (e.e.?>?99%, yield?=?70%) was obtained. The mechanism of chiral resolution was clarified through single-crystal X-ray diffraction of the diastereomeric salt. The absolute configurations of four stereoisomers of 1 were established through electronic circular dichroism spectra, quantum chemical calculation and transition metal method. Their IDO1 inhibitory activity was assessed by pharmacological experiments in vitro and in mouse, demonstrating that S configuration of C5 played an important role on the inhibition of IDO1, while the stereochemistry on C2′ exerted little effect on the IDO1 inhibitory activity in mouse.
