Ten new bridged dimers of oxo-centered triruthenium clusters with CO and 4-(dimethylamino)pyridine (dmap), pyridine (py), or 4-cyanopyridine (cpy) as terminal ligands and pyrazine-d(4) (d(4)-pz), 2,5-dimethylpyrazine (dmpz), 2-methylpyrazine (mpz), and 2-chloropyrazine (clpz) as bridging ligands were prepared. The carbonyl stretching frequency, nu(CO), was used as a probe for infrared spectroelectrochemical measurements. In the neutral and doubly reduced states, a single band was observed for each of the dimers, with a shift in frequency due to the oxidation state of the triruthenium clusters. In the singly reduced state, a range of nu(CO) line shapes was observed, depending on the nature of the ligands, from two bands centered at the frequencies of the bands of the neutral and doubly reduced species to one broad band at the average of these two frequencies. By synthesizing new combinations of bridging and ancillary ligands, electronic communication between two bridged triruthenium clusters was effectively tuned, and electron-transfer rates were estimated by IR spectral line-shape analysis. In dimers bridged by the asymmetric ligand mpz, it was possible through selective isotope labeling of one CO ligand to observe "mixed-valence isomers," the two alternate charge distributions of a mixed-valence complex.  相似文献

The anthrax lethal toxin (LeTx) is composed of two proteins, protective antigen and lethal factor, which bind and enter the cell through a host receptor termed the anthrax toxin receptor (ATR). In the cell, LeTx targets p38, part of the MAP kinase signaling pathway. The toxin appears to initiate an apoptotic pathway in infected cells, indicating additional downstream targets of the toxin. We have applied a proteomics approach to investigate these downstream targets and the affected processes. In this study we have used an improved strategy for fractionation based on protein pI, off-gel electrophoresis, employed in conjunction with relative quantitation using the mass labeling approach. In our survey, 67 proteins were observed and quantified from the cytosol of RAW 264.7 cells with respect to control versus toxin-treated cells. Many of these proteins are involved in the oxidative stress response, as well as apoptosis, and thus likely to be relevant to the effects of anthrax in infected cells. Our results indicate that the tumor necrosis factor-alpha-mediated pathway is compromised in intoxicated cells. The knowledge of such changes and the pathways leading to the changes should be of great value in understanding and combating this disease.  相似文献

Starting from 1-tert-butyloxycarbonyl-3-methyl-4-ethoxycarbonyl-1,2-diaza-1,3-diene and β,β,β and α,β-substituted enamines a careful choice of solvents and temperatures allows the divergent synthesis of 5,6-dihydro-4H-pyridazines, 2-(1-N-boc-hydrazono-ethyl)-4-pyrrolidin-1-yl-but-3-enoic acid ethyl ester, and 1-amino-pyrroles. Moreover, some interesting conclusions about the mechanism(s) of the reaction have been drawn by careful analysis of products' structure and distribution. Thus, the reaction may proceed through a stereospecific [4+2] cycloaddition mechanism giving rise to 5,6-dihydro-4H-pyridazines or by simple addition or domino addition/cyclization pathways affording, respectively, 2-(1-N-boc-hydrazono-ethyl)-4-pyrrolidin-1-yl-but-3-enoic acid ethyl ester and 1-amino-pyrroles (formally the [3+2] cycloaddition product).  相似文献

Acrylate functionalized ionic liquids based on tetraalkylammonium salts with terminal acrylates- and methylacrylates were synthesized. Melting points and ionic conductivity of twenty compounds in six groups were determined. Within one group the effect of three different counterions was investigated and discussed. The groups differ in cationic structure elements because of their functional groups such as acrylate and methacrylate, alkyl residues at the nitrogen and number of quaternary ammonium atoms within the organic cation. The effect of these cationic structure elements has been examined concerning the compiled parameters with a view to qualifying them as components for solid state electrolytes. The newly synthesized ionic liquids were characterized by NMR and FTIR analysis. The exchange of halide ions like bromide as counter ions to weakly coordinating [PF?]?, [OTf]? or [TFSI]? reduces the melting points significantly and leads to an ion conductivity of about 10?? S/cm at room temperature. In the case of the dicationic ionic liquid, an ion conductivity of about 10?3 S/cm was observed.  相似文献

A series of epothilone B and D analogues bearing isomeric quinoline or functionalized benzimidazole side chains has been prepared by chemical synthesis in a highly convergent manner. All analogues have been found to interact with the tubulin/microtubule system and to inhibit human cancer cell proliferation in vitro, albeit with different potencies (IC50 values between 1 and 150 nM ). The affinity of quinoline‐based epothilone B and D analogues for stabilized microtubules clearly depends on the position of the N‐atom in the quinoline system, while the induction of tubulin polymerization in vitro appears to be less sensitive to N‐positioning. The potent inhibition of human cancer cell growth by epothilone analogues bearing functionalized benzimidazole side chains suggests that these systems might be conjugated with tumor‐targeting moieties to form tumor‐targeted prodrugs.  相似文献

The behaviour of 95Zr and 95Nb isotopes on TEVA® resin column was studied. The influence of hydrofluoric, hydrochloric, oxalic and sulphuric acids on the separation and chemical yield of zirconium and niobium was tested. The method is suitable for the separation of 93Zr and 94Nb isotopes from hydrofluoric, hydrochloric and sulphuric acid by using different concentrations of the acid for the load or elutes solutions. Samples from reactor and fuel assemblies such as the control-rod connection rod, shielding/absorber parts of fuel rods, neutron in-core measurement channels, pressure vessel basic construction material and internal cladding, core barrel and reactor protection tube unit were analysed.  相似文献

The natural phenomenon of drug resistance is a widespread issue that hampers the performance of drugs in many major clinical indications. Antibacterial and antifungal drugs are affected, as well as compounds for the treatment of cancer, viral infections, or parasitic diseases. Despite the very diverse set of biological targets and organisms involved in the development of drug resistance, the underlying molecular mechanisms have been identified to understand the emergence of resistance and to overcome this detrimental process. Detailed structural information on the root causes for drug resistance is nowadays frequently available, so next‐generation drugs can be designed that are anticipated to suffer less from resistance. This knowledge‐based approach is essential for fighting the inevitable occurrence of drug resistance.  相似文献
