Bleomycin is a clinically used antitumor antibiotic long thought to function therapeutically at the level of DNA cleavage. Recently, it has become clear that bleomycin can also cleave selected members of all major classes of RNA. Using the computer program COMPARE to search the database established by the Anticancer Drug Screening Program of the National Cancer Institute, a possible mechanism-based correlation was found between onconase, an antitumor ribonuclease currently being evaluated in phase III clinical trials, and the chemotherapeutic agent bleomycin. Following these observations, experimentation revealed that bleomycin caused tRNA cleavage and DNA-independent protein synthesis inhibition in rabbit reticulocyte lysate and when microinjected into Xenopus oocytes. The correlation of protein synthesis inhibition to the previously reported site-specific RNA cleavage caused by bleomycin supports the thesis that RNA cleavage may constitute an important element of the mechanism of action of bleomycin. 相似文献

Europium-doped lanthanide oxide RE(2)O(3):Eu(3+) (RE = Y or Gd) luminescent beads, with a spherical shape and a diameter of 150 ± 15 nm, have been modified by reaction with 3-aminopropyltriethoxysilane (APTES), in order to introduce reactive amine groups at their surfaces. The direct silanation has resulted in the formation of a nanometric layer at the surface of the beads, with an optimum grafting rate of 0.055 ± 0.005 mol APTES/mol RE(2)O(3). Fourier transform infrared (FTIR) and X-ray photoelectron (XPS) spectroscopies confirmed the condensation of an organosilane layer, made of cross-linked -O-Si-O-Si- and of groups -O-Si-R (with R = (CH(2))(3)NH(2) or O-Et). Titration of the accessible amine groups has been performed by simultaneously measuring the luminescence of grafted fluorescein isothiocyanate and that of core particles: there are about 2.3 × 10(4) (2.8 × 10(4)) -NH(2) per Y(2)O(3):Eu(3+) (Gd(2)O(3):Eu(3+)) bead. The isoelectronic point was shifted by one pH unit after APTES modification. The surface modification by APTES at least preserved (for Gd(2)O(3):Eu(3+)) or improved (for Y(2)O(3):Eu(3+)) the red emission of the beads. 相似文献

We study the shear jamming of athermal frictionless soft spheres, and find that in the thermodynamic limit, a shear-jammed state exists with different elastic properties from the isotropically-jammed state. For example, shear-jammed states can have a non-zero residual shear stress in the thermodynamic limit that arises from long-range stress-stress correlations. As a result, the ratio of the shear and bulk moduli, which in isotropically-jammed systems vanishes as the jamming transition is approached from above, instead approaches a constant. Despite these striking differences, we argue that in a deeper sense, the shear jamming and isotropic jamming transitions actually have the same symmetry, and that the differences can be fully understood by rotating the six-dimensional basis of the elastic modulus tensor. 相似文献

A colorimetric method based on silver nanoparticles was developed for the determination of melamine in milk. Silver nanoparticles were synthesized without any stabilizer, using sodium borohydride as the reducing agent. Optimization of the variables for the formation of the nanoparticles was performed by factorial design, resulting in stable colloidal silver nanoparticles with a mean diameter of 14.0?±?2.7?nm. Spectrophotometric measurements performed at 475?nm showed a linear range from 0.033 to 1.50?mg?L?1 of melamine with limits of detection and quantification of 0.009 and 0.031?mg?L?1, respectively. Modeling the consequences of crystalline defects requires efficient interaction sampling. Empirical potentials can identify relevant pathways if the energetics and configurations of competing defects are captured. Here, we develop such a potential for an alloy of arbitrary point defect concentration, body-centered cubic alpha-Fe supersaturated in C. This potential successfully calculates energetically favored defects, and predicts formation energies and configurations of multicarbon-multivacancy clusters that were not attainable with existing potentials or identified previously via ab initio methods. 相似文献

Thermal conduction modes in a nanocolloid (nanofluid) are quantitatively assessed by combining linear response theory with molecular dynamics simulations. The microscopic heat flux is decomposed into three additive fluctuation modes, namely, kinetic, potential, and collision. For low volume fractions (<1%) of nanosized platinum clusters which interact strongly with xenon host liquid, a significant thermal conductivity enhancement results from the self correlation in the potential flux. Our findings reveal a molecular-level mechanism for enhanced thermal conductivity in nanocolloids with short-ranged attraction and offer predictions that can be experimentally tested. 相似文献
