In the presence of an (R)-MOP-Pd2(dba)3 catalyst, the reaction of ortho-tert-butylaniline with 2-bromophenyl arylethynyl ketone proceeded via a tandem amination (1,4-addition of aniline to an ynone and subsequent intramolecular Buchwald–Hartwig amination) to afford axially chiral N-(2-tert-butylphenyl)-2-aryl-4-quinolinone derivatives with moderate enantioselectivity (up to 72% ee).  相似文献

Allopurinol (ALP), an inhibitor of the xanthine oxidase enzime, is reported to provide protection against free-radical mediated damage by various mechanisms, including free-radical scavenging and metal chelation (i.e. Cu(II)). To obtain a wider insight into the molecular aspects of the beneficial action of ALP, free ALP and the Cu(II)- ALP system were investigated by radiation chemical and spectroscopic studies. Pulse radiolysis experiments show that ALP is a good · OH scavenger (1.8 × 109 and 5.4 × 109 M-1 s-1 at pH 6.0 and 11.0, respectively), leading to · OH-adducts and transient semi-oxidized species, such as phenoxyl radicals. The latter are also formed by the reaction of ALP with some specific one-electron oxidizing radicals (i.e. N3 and SO4 -). The semi-oxidized species are stabilized by their resonance properties and scarcely react with oxygen. In addition, the chelation of Cu(II) by ALP does not significantly affect the reactivity of the drug towards ·OH (2.5×09 M-1 s-1). Raman and the IR spectra support the good chelating ability of ALP, indicating the formation of two Cu(II)- ALP complexes with a slightly different structure. Depending on the metal/ligand ratio, pyrimidine nitrogens may take part to the Cu(II) co-ordination in addition to the N pyrazolic atoms and the C O groups of some ALP molecules. These results suggest that ALP may inhibit oxidative damage both through the direct radical scavenging and the copper-chelation mechanism. In fact, both the conversion of a harmful radical, such as ·OH, into a less reactive transient species, and the capture of copper ions, which play a relevant role in metal-catalysed generation of reactive oxygen species, will prove beneficial for the cell protection.  相似文献

The hydrolysis of 2-phenylethyl β-d-glucopyranoside (3) was found to be partially inhibited by feeding with 2-phenyl-N-glucosyl-acetamidiumbromide (8), a β-glucosidase inhibitor, resulting in a decrease in the diurnal emission of 2-phenylethanol (2) from Rosa damascena Mill. flowers. Detection of [1,1,2,2′,3′,4′,5′,6′-2H8]-2 and [1,2,2′,3′,4′,5′,6′-2H7]-2 from R. ‘Hoh-Jun’ flowers fed with [1,1,2,2′,3′,4′,5′,6′-2H8]-3 suggested that β-glucosidase, alcohol dehydrogenase, and reductase might be involved in scent emission. Comprehensive GC-SIM analyses revealed that [1,2,2,2′,3′,4′,5′,6′-2H8]-2 and [1,2,2,2′,3′,4′,5′,6′-2H8]-3 must be biosynthesized from [1,2,2,2′,3′,4′,5′6′-2H8] l-phenylalanine ([2H8]-1) with a retention of the deuterium atom at α-position of [2H8]-1.  相似文献

A catalytic enantioselective synthesis of β-amino secondary amides was achieved using vinyl azides as the enamine-type nucleophile and chiral N-Tf phosphoramide as the chiral Brønsted acid catalyst through a five-step sequential transformation in one pot. The established sequential transformation involves an enantioselective [4+2] cycloaddition reaction of vinyl azides with N-acyl imines as the key stereo-determining step that is efficiently accelerated by a chiral N-Tf phosphoramide catalyst in a highly enantioselective manner in most cases. Further generation of the iminodiazonium ion intermediate through ring opening of the cycloaddition product and subsequent skeletal rearrangement involving Schmidt-type 1,2-aryl group migration followed by recyclization of the resulting nitrilium ion were also initiated by the same acid catalyst. Final acid hydrolysis of the recyclized products in the same pot gave rise to enantioenriched β-amino amides through C−C bond formation at the α-position of the secondary amides.  相似文献

A novel series of C12-keto-type saxitoxin (STX) derivatives bearing an unusual nonhydrated form of the ketone at C12 has been synthesized, and their NaV-inhibitory activity has been evaluated in a cell-based assay as well as whole-cell patch-clamp recording. Among these compounds, 11-benzylidene STX ( 3 a ) showed potent inhibitory activity against neuroblastoma Neuro 2A in both cell-based and electrophysiological analyses, with EC50 and IC50 values of 8.5 and 30.7 nm , respectively. Interestingly, the compound showed potent inhibitory activity against tetrodotoxin-resistant subtype of NaV1.5, with an IC50 value of 94.1 nm . Derivatives 3 a – d and 3 f showed low recovery rates from NaV1.2 subtype (ca 45–79 %) compared to natural dcSTX ( 2 ), strongly suggesting an irreversible mode of interaction. We propose an interaction model for the C12-keto derivatives with NaV in which the enone moiety in the STX derivatives 3 works as Michael acceptor for the carboxylate of Asp1717.  相似文献

The oxidative degradation of cresols smoothly proceeded toward inorganic end products when a gaseous plasma generated by means of dc glow discharge was sustained in contact with the surface of aqueous solution containing organic compounds. In order to get mechanistic insight, the monohydroxylation products from each isomeric cresol were closely examined as primary intermediates to reveal that the aromatic hydroxylation preferentially occurred at the position para to the hydroxyl group of each starting material. It was also established that the degradation of cresols strictly followed the first-order rate law. On the basis of the orientational analysis and the kinetical consideration including the effects of Fe ions added on the reaction rate, it was concluded that hydroxyl radical, which might result from the homolytic cleavage of water molecules by the action of plasma, was the most likely reagent responsible for the mineralization of cresols.  相似文献

Flavonoids are ingested by the general population as anti-oxidant and anti-inflammatory agents. In this study, we investigated the effects of myricitrin, a flavonoid rich in Myrica rubra leaf, upon anti-inflammatory action. Myrica rubra leaf extracts inhibited pro-inflammatory TNFα production in a macrophage cell line, Raw264.7 cells. We observed that the serum IgE levels in the leaf extract-treated DO11.10, a mouse allergy model, were down-regulated. HPLC was performed to demonstrate that M. rubra leaf extracts contain a large amount of myricitrin. We observed an inhibitory effect of HPLC-purified myricitrin on TNFα production in Raw264.7 cells. Thus, myricitrin may be of potential interest in the management of inflammatory conditions.  相似文献

The effects of water content on water transport and electro-osmosis in a representative polymer electrolyte membrane, Nafion, are investigated in detail by means of first-principles molecular dynamics (MD) simulations in the presence of a homogeneous electric field. We have directly evaluated electro-osmotic drag coefficients (the number of water molecules cotransported with proton conduction) from the trajectories of the first-principles MD simulations and also explicitly evaluated factors that contribute to the electro-osmotic drag coefficients. In agreement with previously reported experiments, our calculations show virtually constant values ( approximately 1) of the electro-osmotic drag coefficients for both low and high water content states. Detailed comparisons of each factor contributing to the drag coefficient reveal that an increase in water content increases the occurrence of the Grotthuss-like effective proton transport process, whose contribution results in a decrease in the electro-osmotic drag coefficient. At the same time, an environment that is favorable for the Grotthuss-like effective proton transport process is also favorable for the transport of water arising from water transport occurring beyond the hydration shell around the protons, whose contribution results in an increase in the electro-osmotic drag coefficient. Conversely, an environment that is not favorable for proton conduction is also not favorable for water transport. As a result, the electro-osmotic drag coefficient shows virtually identical values with respect to change in the water content.  相似文献

A tungsten trioxide (WO(3)) coated aluminium (Al) substrate exhibits a reversible color change without light irradiation or an electric power supply. The sample turns blue upon exposure to an aqueous acid solution, but exposure to oxygen restores the original light yellow color.
