Histone deacetylases (HDACs) are key enzymatic regulators of many cellular processes such as gene expression, cell cycle, and tumorigenesis. These enzymes are attractive targets for drug development. However, very few simple methods for monitoring HDAC activity have been reported. Here, we have developed a fluorogenic probe, K4(Ac)-CCB, which consists of the histone H3 peptide containing acetyl-Lys and a coumarin fluorophore with a carbonate ester. By the simple addition of the probe to a HDAC solution, enzyme activity was clearly detected through spontaneous intramolecular transesterification, which renders the probe fluorescent. In addition, K4(Ac)-CCB can be applied to the evaluation of HDAC inhibitor activity. This is the first report to demonstrate the monitoring of HDAC activity by using a one-step procedure. Thus, our novel fluorogenic probe will provide a powerful tool for epigenetic research and the discovery of HDAC-targeted drugs. 相似文献

Stroke, a deleterious cerebrovascular event, is caused by a critical reduction in the blood flow to the brain parenchyma that leads to brain injury and loss of brain functions. The inflammatory responses following ischemia often aggravate the neurological damage. Several pro‐inflammatory mediators released after stroke are closely related to the metabolism of phospholipids. In this study we directly profiled the changes in phospholipids in the infarcted rat cerebral cortex 24 hours after middle cerebral artery occlusion (MCAO) using matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). Several phosphatidylcholine (PC) species and sphingomyelin (SM) were significantly decreased after infarction. The cationization pattern of the remaining PCs showed a prominent shift from a mostly potassiated or protonated form to a predominantly sodiated pattern. Stroke also elevated the lysophosphatidylcholines (LPCs) and heme in tissue. The isobaric pairs in PC and LPC classes were resolved by masses through their respective alkali metal adducts in the presence of CsCl. The major fatty acyl LPC species were also structurally confirmed by MALDI‐MS/MS. Overall, the results described the changes in PC and LPC species in the infarcted rat cortex. The elevated tissue levels of LPCs and heme signify the ongoing pathological lipid breakdown and the state of parenchymal inflammation. The elevated LPC level in tissue suggests a means of intervention through lysophospholipid metabolism that could potentially benefit the management of stroke and other acute neurological injuries. We report a photoluminescence detected anticrossing of the energy levels in an undoped asymmetric coupled-double-quantum-well buried in a p-i-n structure. Due to the built-in electric field, the quantum wells are tilted in such a way that the symmetric energy level is higher than that of the antisymmetric one in the conduction band. Keeping the laser excitation energy below the barrier, with increasing laser power, the level anticrossing and the quantum confined Stark effect were observed due to decreasing built-in electric field by the photogenerated electron and hole pairs. 相似文献

A single-longitudinal-mode selection is realized in a linear-cavity fiber laser construction using a loop-back optical circulator and a partial reflectance fiber Bragg grating as the cavity ends. At 1551 nm wavelength, the measured signal-to-noise-ratio is 56 dB and with a line width less than 1 MHz obtained using multiple subring cavities as mode filters. Li (lithium) isotope analysis using MC-ICP-MS is a very powerful tracer measurement method. This is widely used for identification of Li isotopes in many fields of study. This useful method, however, has an effect on the natural Li isotope background. This is impacted by the instrument matrix. In this study, we show that the MC-ICP-MS condition is characterized by both a low baseline background and a high-sensitivity distance at Ar plasma condition. In addition, the Li isotope ratio was measured by the use of experimental conditions that were superior to both the general plasma condition and those used in other studies. The samples were subjected to both acid leaching and a cation exchange resin (Bio-Rad AG 50 W-X8 200–400 mesh) modified for seawater samples. The isotope variations were corrected using the bracket method, the measured Li isotope ratio of sample, and the mean ratios of the L-SVEC standard (NIST L-SVEC Li2CO3) measured before and after the sample run. The isotope variation was presented as the deviation (per mil) of the measured ratio from that of the recommended value. 相似文献

Geometric and conformational changes of zwitter‐type ionic liquids (ZILs) due to hydrogen‐bonding interactions with water molecules are investigated by density functional theory (DFT), two‐dimensional IR correlation spectroscopy (2D IR COS), and pulsed‐gradient spin‐echo NMR (PGSE NMR). Simulation results indicate that molecular structures in the optimized states are strongly influenced by hydrogen bonding of water molecules with the sulfonate group or imidazolium and pyrrolidinium rings of 3‐(1‐methyl‐3‐imidazolio)propanesulfonate ( 1 ) and 3‐(1‐methyl‐1‐pyrrolidinio)propanesulfonate ( 2 ), respectively. Concentration‐dependent 2D IR COS reveals kinetic conformational changes of the two ZIL–H2O systems attributable to intermolecular interactions, as well as the interactions of sulfonate groups and imidazolium or pyrrolidinium rings with water molecules. The dramatic changes in the 1H self‐diffusion coefficients elucidate the formation of proton‐conduction pathways consisting of ZIL networks. In ZIL domains, protons are transferred by a Grotthuss‐type mechanism through formation, breaking, and restructuring of bonds between ZILs and H2O, leading to an energetically favorable state. The simulation and experimental investigations delineated herein provide a perspective to understanding the interactions with water from an academic point of view as well as to designing ILs with desired properties from the viewpoint of applications. 相似文献

A method to determine 8 polychlorinated biphenyls (PCBs), 23 organochlorine pesticides (OCPs) and 16 polycyclic aromatic hydrocarbons (PAHs) was described using dispersive liquid–liquid microextraction (DLLME) of a small amount of plasma or serum sample and gas chromatography–tandem mass spectrometry (GC–MS/MS). The appropriate selection of the extraction solvent and dispersing solvent contributes to a high extraction yield and a clean extract. To verify the developed method, the interference, linearity of the calibration curve, detection limit, precision and accuracy were evaluated. The calibration curves were linear by 2–3 orders of magnitude with correlation coefficients above 0.997 in all cases. The LODs of PCBs, OCPs and PAHs were measured in the ranges of 0.0006–0.0029, 0.001–0.029 and 0.0002–0.012 ng/mL. The intraday precision achieved by this method was 2.19–10.3% (PCBs), 1.65–14.3% (OCPs) and 0.91–12.8% (PAHs), and the intraday accuracy 1.56–7.37% (PCBs), 2.34–19.6% (OCPs) and 1.49–15.7% (PAHs). The advantage of this method is that the analysis of PCBs, OCPs, and PAHs can be performed in a single chromatographic run, and the low detection limit enables monitoring of target substances in low exposure general public samples, and the analysis procedure is relatively simple and fast. 相似文献
