Pyrolysis–GC/mass spectrometry experiments reveal that naphthalene groups attached to maleated polyethylene as the 1-naphthylethyl ester are stable for relatively long periods of time at 170°C. Decomposition can be detected for samples heated for 2.0 min at 200°C, but even at that temperature, the extent of decomposition is very small. At higher temperatures, two of the decomposition products from the labeled polymer are readily understood: 1-vinylnaphthalene and 1-naphthylethanol can form by reactions that are well-precedented in the organic chemistry literature. At 200°C, only naphthalene is formed, which requires scission of the bond between the naphthyl ring and the C1 carbon of the ethyl group. With the widespread clinical use of drug combinations, the incidence of drug–drug interactions (DDI) has significantly increased, accompanied by a variety of adverse reactions. Drug transporters play an important role in the development of DDI by affecting the elimination process of drugs in vivo, especially in the pathological state. Tubulointerstitial fibrosis (TIF) is an inevitable pathway in the progression of chronic kidney disease (CKD) to end-stage renal disease. Here, the dynamic expression changes of eleven drug transporters in TIF kidney have been systematically investigated. Among them, the mRNA expressions of Oat1, Oat2, Oct1, Oct2, Oatp4C1 and Mate1 were down-regulated, while Oat3, Mrp2, Mrp4, Mdr1-α, Bcrp were up-regulated. Pearson correlation analysis was used to analyze the correlation between transporters and Creatinine (Cr), OCT2 and MATE1 showed a strong negative correlation with Cr. In contrast, Mdr1-α exhibited a strong positive correlation with Cr. In addition, the pharmacokinetics of cimetidine, ganciclovir, and digoxin, which were the classical substrates for OCT2, MATE1 and P-glycoprotein (P-gp), respectively, have been studied. These results reveal that changes in serum creatinine can indicate changes in drug transporters in the kidney, and thus affect the pharmacokinetics of its substrates, providing useful information for clinical use. 相似文献

为了提高胆碱传感器的灵敏度和抗干扰性,以纳米碳管修饰铂电极为基础电极,采用溶胶-凝胶法固定胆碱氧化酶(ChOx),构建了电流型胆碱检测生物传感器,对纳米碳管修饰电极的电化学特征进行分析,得知纳米碳管的引入不仅使电极对H2O2的催化电流增大,同时降低了电催化所需的恒定电位。讨论了缓冲液介质、pH值、酶负载量对传感器响应的影响。研究表明,所制备的传感器在pH 7.2、电位为0.15V条件下对氯化胆碱的线性响应范围为5.0×10-6~1.0×10-4mol/L;检出限为5.0×10-7mol/L;灵敏度为9.48μA/mmol/L。传感器的稳定性好,经过1个月,仍可保持初始电流的85%。抗干扰能力有很大提高。用于人体血清中的胆碱浓度测定,结果令人满意。 相似文献

采用反相高效液相色谱-电喷雾质谱法(LC-ESI/MS)测定人体血浆中盐酸舍曲林。以盐酸丙咪嗪为内标,按内标法定量。血浆样品经pH 10.00碳酸钠溶液碱化,加入甲基叔丁基醚萃取,离心分离,取上清液吹干,用流动相溶解进样。色谱柱:RP-Extend-C18(150 mm×4.6 mm,5μm),柱温:40℃;流动相:pH 3.00的三氟乙酸溶液-甲醇(40∶60,V/V),流速0.6 mL/m in。质谱采用选择离子监控模式,检测离子的核质比(m/z)分别是281(丙咪嗪)和159(舍曲林)。舍曲林和丙咪嗪的保留时间分别是5.4 m in和3.8 m in;舍曲林标准曲线线性范围为1~80μg/L;检出限为1.0μg/L(S/N>10);日间、日内相对标准差均小于6.0%,相对回收率为90%~106%;提取回收率在75%~93%范围内。此法适合人体药代动力学的血浆样品的分析,结果准确、可靠。 相似文献
