Background: Poria cocos (P. cocos) is an important medicinal fungus in traditional Chinese medicine. Poria acid (PA), a triterpenoid compound, is an effective component of traditional Chinese medicine P. cocos. This experiment investigated the anti-gastric cancer biological activity of PA in vitro. Methods: The effect of PA on the viability of gastric cancer cells was detected by the thiazolyl blue (MTT) assay. Cell adhesion assays were used to detect changes in the adhesion of cells treated after PA (0, 20, 40, and 80 µmol/L). The ability of cell invasion and migration were detected by Transwell assays and wound healing assays. A high-content imaging system was used to dynamically record the motility of the gastric cancer cells after PA (0, 20, 40, and 80 µmol/L) treatment. Western blotting was used to detect the expression of epithelial–mesenchymal transformation (EMT), invasion and migration related proteins. Results: The MTT assay showed that the proliferation of gastric cancer cells was significantly inhibited after PA treatment. Cell adhesion experiments showed that the adhesion of gastric cancer cells was significantly decreased after PA treatment. Compared with the control group, the wound healing area of the gastric cancer cells treated with different concentrations of PA decreased. The Transwell assay showed that the number of gastric cancer cells passing through the cell membrane were significantly reduced after PA treatment. In addition, after PA treatment, the cells’ movement distance and average movement speed were significantly lower than those of the control group. Finally, PA can significantly alter the expression of EMT-related proteins E-cadherin, N-cadherin, and Vimentin and decreased the expressions of metastasis-related proteins matrix metalloproteinase (MMP) 2, MMP-9 and tissue inhibition of matrix metalloproteinase (TIMP)1 in the gastric cancer cells. Conclusions: Triterpenoids from P. cocos have significant biological activity against gastric cancer, and the mechanism may be involved in the process of epithelial–mesenchymal transformation. 相似文献

夹心式换能器应用极为广泛,但当其横向尺寸过大时,存在耦合振动,影响其辐射面的位移分布.本文通过在大尺寸夹心式换能器的前盖板中加工周期排列的槽,来形成一种二维声子晶体结构.随后,采用有限元法对基于二维声子晶体的大尺寸夹心式换能器的振动传输特性、共振频率以及发射电压响应进行仿真模拟,讨论了开槽高度和开槽宽度对其带隙、共振与反共振频率、带宽以及辐射面位移分布的影响.研究结果表明,通过在大尺寸夹心式换能器中应用声子晶体结构可对其进行优化设计.当大尺寸夹心式换能器的工作频率位于其带隙范围内时,二维声子晶体结构能有效地抑制其横向振动,从而改善换能器辐射面位移分布的均匀程度.此外,在大尺寸夹心式换能器的前盖板中加工二维声子晶体结构,能有效提升换能器的带宽,进而拓宽大尺寸夹心式换能器的工作频带. 相似文献

在液体石蜡中采用微波技术原位合成了油溶性环烷酸铅(LN)和十二烷基水杨酸铅(LAS),在高速低负荷(r=1500±10rpm,P=196-392N)和低速高负荷(r=300±10rpm,P=800N)两种条件下,用四球摩擦磨损试验对LN,LAS和对应的羧酸进行了摩擦学性能评价,用往复式摩试验机考察了LN和LAS抗磨减摩性能,结果表明:LN具有良好的抗磨减磨性能和中等的极压性能,且各项摩擦学性能指标均好于LAS。为弄清其作用机理,从分子结构分析了产生摩擦学性能差异的原因,并用SEM及XPS研究了磨斑表面,结果发现:摩擦过程中,LN和LAS都能在摩擦副表面形成吸附膜且部分吸附膜发生摩擦化学反应产生了铅氧化物转化膜,但所形成的吸附膜和转化膜厚度不同。 相似文献

The antibacterial activity of propolis has long been of great interest, and the chemical composition of propolis is directly dependent on its source. We recently obtained a type of propolis from China with a red color. Firstly, the antibacterial properties of this unusual propolis were determined against Staphylococcus aureus and methicillin-resistant Staphylococcus aureus (MRSA). Studies on its composition identified and quantified 14 main polyphenols of Chinese red propolis extracts (RPE); quantification was carried out using liquid chromatography triple quadrupole tandem mass spectrometry (LC-QQQ-MS/MS) and RPE was found to be rich in pinobanksin, pinobanksin-3-acetate, and chrysin. In vitro investigations of its antibacterial activity revealed that its activity against S. aureus and MRSA is due to disruption of the cell wall and cell membrane, which then inhibits bacterial growth. Despite its similar antibacterial activities against S. aureus and MRSA, metabolomic analysis further revealed the effects of RPE on bacteria metabolism were different. The untargeted metabolomic results showed that a total of 7 metabolites in 12 metabolic pathways had significant changes (Fold change > 2, p < 0.05 *) after RPE treatment in S. aureus, while 11 metabolites in 9 metabolic pathways had significant changes (Fold change > 2, p < 0.05 *) after RPE treated on MRSA. Furthermore, RPE downregulated several specific genes related to bacterial biofilm formation, autolysis, cell wall synthesis, and bacterial virulence in MRSA. 深入研究了测试图案对测量畸变的影响。提出"一靶测试法"进行畸变测量与校正,拍摄了自行设计、制作的"综合点阵靶板"的畸变图像,图像处理得到其畸变点位置,并与理想点位置进行对应比较。建立多项式模型确定畸变点与理想点的位置校正关系,理想点位置由理想成像得到,并运用最小二乘拟合算法求得多项式系数,标定整个CCD摄像系统的畸变。"综合点阵靶板"采用灰色圆点黑色间带图案,方便了点按序标记排列,从而确定畸变点与理想点的一一对应。对2.6 mm焦距的广角CCD镜头摄像系统进行了畸变测量与校正,其重复测量,校正精度达到0.3%。 相似文献

Nearly 5% of the Shenqi Fuzheng Injection’s dry weight comes from the secondary metabolites of Radix codonopsis and Radix astragali. However, the chemical composition of these metabolites is still vague, which hinders the authentication of Shenqi Fuzheng Injection (SFI). Ultra-high performance liquid chromatography with a charged aerosol detector was used to achieve the profiling of these secondary metabolites in SFI in a single chromatogram. The chemical information in the chromatographic profile was characterized by ion mobility and high-resolution mass spectrometry. Polygonal mass defect filtering (PMDF) combined with Kendrick mass defect filtering (KMDF) was performed to screen potential secondary metabolites. A total of 223 secondary metabolites were characterized from the SFI fingerprints, including 58 flavonoids, 71 saponins, 50 alkaloids, 30 polyene and polycynes, and 14 other compounds. Among them, 106 components, mainly flavonoids and saponins, are contributed by Radix astragali, while 54 components, mainly alkaloids and polyene and polycynes, are contributed by Radix codonopsis, with 33 components coming from both herbs. There were 64 components characterized using the KMDF method, which increased the number of characterized components in SFI by 28.70%. This study provides a solid foundation for the authentification of SFIs and the analysis of its chemical composition. 相似文献

提出了一种新的算法—高阶辛时域有限差分法(SFDTD(3,4):symplectic finite-difference time-domain)求解含时薛定谔方程.在时间上采用三阶辛积分格式离散,空间上采用四阶精度的同位差分格式离散,建立了求解含时薛定谔方程的高阶离散辛框架;探讨了高阶辛算法的稳定性及数值色散性.通过理论上的分析及数值算例表明:当空间采用高阶同位差分格式时,辛积分可提高算法的稳定度;SFDTD(3,4)法和FDTD(2,4)法较传统的FDTD(2,2)法数值色散性明显改善.对二维量子阱和谐振子的仿真结果表明:SFDTD(3,4)法较传统的FDTD(2,2)法及高阶FDTD(2,4)法有着更好的计算精度和收敛性,且SFDTD(3,4)法能够保持量子系统的能量守恒,适用于长时间仿真. 相似文献
