A spectrofluorimetric method has been developed for the determination of trace Cu(II) in real samples with 4,5-dihydroxy-l,3-benzenedisulfonic acid (Tiron) as a fluorimetric reporter. Tiron is very soluble in water and is a good fluorimetric reagent. However, as Tiron was complexed with Cu(II), the fluorescence intensity decreased proportionally to the concentration of Cu(II) by a static quenching effect. The excitation wavelength and the fluorescence wavelength of Tiron were 294 and 350 nm, respectively, as it was caused by a quenching effect from Cu(II) at pH 8.0. The highest sensitivity was shown at Tiron concentration of 5.0x10(-5) M. To enhance the quenching effect, the Cu(II)-Tiron complex solution was heated up to 80 degrees C for 90 min. As for Cu(II), the interference by Co(II) was very serious, which was eliminated by oxalate ion. The linear response to Cu(II) was shown at the concentration range between 5.0x10(-7) and 1.0x10(-5) M. With this proposed method, the detection limit of Cu(II) was 3.83(+/-0.09)x10(-7) M. Recoveries of Cu(II) in the diluted brass samples and the stream water samples were almost 100%. Based on results from the experiment, this proposed technique could be applied to the practical determination of Cu(II) in real samples.  相似文献

Two series of new poly(amide imide)s having (n‐alkyoxy)phenyloxy side branches with various lengths, poly{p‐phenyleneiminoterephthaloylimino‐p‐phenylene[3,6‐di(n‐alkyloxy)phenyloxy]pyromellitimide}s ( PC m TA s, m = 4, 8, 12) and poly{p‐phenyleneiminosebacoylimino‐p‐phenylene[3,6‐di(n‐alkyloxy)‐phenyloxy]‐ pyromellitimide}s ( PC m SeA s, m = 4, 8, 12), were prepared by condensation of terephthalamide‐N,N′‐4,4′‐dianiline ( TA ) and sebacamide‐N,N′‐4,4′‐dianiline ( SeA ) with 3,6‐di[4‐(n‐alkyloxy)phenyloxy]pyromellitic dianhydrides , respectively. The inherent viscosities of the polymers were in the 0.82–1.20 dL/g range. The polymers were highly soluble in N‐methylpyrolidinone (NMP), even at room temperature and soluble in other polar aprotic solvents on heating. The PC m TA s, which have aromatic backbones, were thermally more stable (431–442 °C) than the PC m SeA s, which have an octamethylene unit in the main chain (407–409 °C). Degradation of weight up to 900 °C corresponded with the loss of side chain contents. The PC m TA s exhibited no phase transition, whereas two endothermic peaks were observed for each of the PC m SeA s. Wide‐angle X‐ray diffractometer investigations revealed that both polymers are amorphous and the n‐alkyloxy side chains are present in a layered structure. © 2001 John Wiley & Sons, Inc. Five‐member cyclic dithiocarbonates were synthesized by the reactions of carbon disulfide with benzoic, p‐anisic, p‐chlorobenzoic, 1‐naphthalenecarboxylic, p‐nitrobenzoic, and p‐(tert‐butyl)benzoic glycidyl esters, and their cationic ring‐opening polymerizations were carried out with methyl trifluoromethane sulfonate and trifluoromethane sulfonic acid as initiators at room temperature to 80 °C. Polymers with number‐average molecular weights of 3400–24,900 were obtained in high yields, and their structures were estimated by NMR and IR spectroscopy. The monomers showed a clear difference in the polymerization rate according to the substituents. The rate of polymerization decreased in the order of p‐chlorobenzoic ≥ benzoic > 1‐naphthalenecarboxylic > p‐nitro‐benzoic > ptert‐butylbenzoic > p‐anisic. The data of the reaction kinetics, NMR studies, and molecular orbital calculations proved a plausible mechanism involving the participation of p‐substituted benzoyloxymethyl groups to stabilize the cationic propagating end. The polymers showed decomposition temperatures with 5% weight loss ranging from 200 to 260 °C. No glass‐transition temperatures for the polymers were observed below 200 °C by differential scanning calorimetry. © 2001 John Wiley & Sons, Inc. A sensitive and specific high-performance liquid chromatographic (HPLC) assay was developed for the quantitation of mitoxantrone in plasma using electrochemical detection. Bisantrene was chosen as the internal standard. A reversed-phase, 10-microns muBondapak C18 analytical column (30 cm X 3.9 mm) with an isocratic mobile phase of 28% acetonitrile in 80 mM sodium formate buffer (pH 3.0) was used. The eluent was monitored by both electrochemical detection at an applied potential of +0.75 V vs. Ag/AgCl and visible absorbance at 660 nm. Only electrochemical detection was able to quantitate the internal standard and provided ten times higher sensitivity than visible absorbance for mitoxantrone with a detection limit as low as 0.1 ng/ml. Calibration curves in the range 0.1-1000 ng/ml showed good linearity (r = 0.998) and precision (coefficient of variation less than 10%). This HPLC method utilized a reproducible and inexpensive liquid-liquid extraction procedure. Using methylene chloride, the extraction efficacy of mitoxantrone from plasma was 85.3% with a coefficient of variation less than 2.1%. This new assay was then applied to measure mitoxantrone concentrations in plasma obtained from two leukemic patients receiving 12 mg/m2 mitoxantrone as a 1-h infusion.  相似文献

The studies with proteins are necessary to understand the biological effects of atmospheric pressure plasma (APP). Among proteins, those with transient metal ions play key roles in many biological events and they are very sensitive to environmental redox states. Iron-containing hemoglobin (Hb) is investigated in this study, after APP treatments under two environmental gas conditions of pure N2 and N2?+?O2 mixture. Monitoring the intensity change for absorption spectra could lead to a quantitative assessment of the effect of discharge plasma on Hb. Redox states of Hb are classified into five states including O2-bound Hb (oxy-Hb), deoxy-Hb, met-Hb, NO-bound Hb (NO-Hb), and hemichrome. Chemically generated reactive species and some scavengers are applied to understand the chemical reactions. Our experimental results confirm the complex chemical reactions of APP and suggest the possible use of Hb as a model protein for the visualization of APP biological effects.  相似文献

Highly ordered titanium dioxide (TiO2) nanotubes film was successfully synthesized via anodic oxidation of a Ti foil in an ammonium fluoride-based ethylene glycol solution. The electrogenerated chemiluminescence (ECL) behavior of the resulting TiO2 nanotubes film was subsequently studied. Strong ECL emission was observed at −1.40 V (vs. Ag/AgCl) and the ECL spectrum displayed three emission peaks which were bathochromatically shifted by ca. 140 nm as compared to its corresponding photoluminescence (PL) emission peaks, indicating that the surface state plays an important role in the emission process. The ECL emission can also occur in a deareated solution attributing to the surface adsorbed O2 molecules. The ECL emission intensity was quenched by dopamine and greatly enhanced in the presence of dissolved O2 and H2O2, making it possible to detect these analytes. The TiO2 nanotubes film has been successfully applied to determine the dissolved O2 content in river and pond water samples, the H2O2 concentration in commercial disinfectant samples and the dopamine concentration in commercial dopamine injections with satisfactory results. The plausible ECL mechanisms of TiO2 nanotubes film in aqueous solution are discussed.  相似文献

Alkali metal cations easily form complexes with proteins in biological systems; understanding amino acid clusters with these cations can provide useful insight into their behaviors at the molecular level including diagnosis and therapy of related diseases. For the purpose of characterization of basic interaction between amino acids and alkali metal, each of the 20 naturally occurring amino acids were ionized in the presence of lithium, sodium and potassium cations by electrospray ionization, and the resulting product ions were analyzed. We focus our attention on the gas phase alkali metal ion-proton exchanged complexes in current study, specifically complexes with serine, threonine, asparagine and glutamine, which share characteristic pattern unlike other amino acids. All amino acids generated [M + H](+) and [M + Na](+) ions, where M stands for the neutral amino acid. Serine, threonine, asparagine and glutamine generated cluster ions of [nM - nH + (n + 1)Na](+) and [nM - (n - 1)H + (n - 1)Na + K](+) , where n = 1-7. While the (M - H + Li) and (M - H + K) species were not observed, the neutral (M - H + Na) species formed by proton-sodium cation exchange had a highly stable cyclic structure with ketone and amine ligand sites, suggesting that (M - H + Na) serves as a building block in cluster ion formation. Cluster ion intensity distributions of [nM - nH + (n + 1)Na](+) and [nM - (n - 1)H + (n - 1)Na + K](+) showed a magic number at n = 3 and 4, respectively. Extensive B3LYP-DFT quantum mechanical calculations were carried out to elucidate the geometry and energy of the cluster ions, and they provided a reasonable explanation for the stability and structure of the cluster ions.  相似文献

Trace detection and physicochemical characterization of protein aggregates have a large impact in understanding and diagnosing many diseases, such as ageing-related neurodegeneration and systemic amyloidosis, for which the formation of protein aggregates is one of the pathological hallmarks. Here we demonstrate an innovative label-free method for detecting and characterizing small amounts of early stage protein aggregates using a Raman active nanofluidic device. Sub-micrometre channels formed by a novel elastomeric collapse technique enable the separation and concentration of matured protein aggregates from small protein molecules. The Raman enhancement by gold nanoparticle clusters fixed below a micro/nanofluidic junction allows characterization of intrinsic properties of protein aggregates at concentration levels (~fM) much lower than can be done with traditional analytical tools. With our device we show for the first time the concentration dependence of protein aggregation over these low concentration ranges. We expect that our method could facilitate definitive diagnosis and possible therapeutics of diseases at early stages.  相似文献
