Production of galactose epimerase by anKluyveromyces sp. isolated from Kefir (dairy product) was investigated in batch culture. The microorganism was cultured in media with 1%
galactose, 1% xylose, or 0.5% xylose plus 0.5% galactose, in Erlenmeyer flasks shaken at 200 rpm and maintained at 30°C. After
48 h, the biomass was harvested by centrifugation and permeated with 80% ethanol. Permeated cells were suspended in 0.1M sodium
phosphate buffer pH 6.5. A part of this suspension was shaken for 17h at 140 rpm. The supernatant, free of cells, was separated.
Partial characterization ofKluyveromyces sp. epimerase was carried out in the cellular suspension and the supernatant solution. Enzymatic activity, using galactose
as substrate, was measured. The product of this reaction was measured by the use of glucose oxidase. The results indicated:
(1) there was a strong effect of xylose on induction of epimerase activity; (2) the epimerase obtained was independent of
the energetic cell activity; (3) the epimerase activity in whole cells was similar to the activity obtained from the supernatant;
(4) epimerase showed a typical substrate-inhibition curve and dependence on magnesium; and (5) the best pH range was between

Systematic monitoring of Rn in soil, background gamma levels and meteorological measurements, have been conducted at the site of a Nuclear Research Center. The data cover a time period of several years. The radon in soil gas fluctuations are influenced by meteorological seasonal changes and by geophysical local parameters and they range between 1000 Bq/m3 and 18000 Bq/m3. The observed gamma levels show geographic stability; some seasonal fluctuations were observed. Data analysis indicates that no significant changes occurred at the site over the measuring time period and radiation levels and meteorological changes are moderate inducing no ecological impact on the surroundings. 相似文献

Total cyclic-3'-5'-adenosine monophosphate (cAMP) levels were measured in the gametophyte of the red macroalga Porphyra leucosticta under different light conditions in order to study its regulation by phytochrome or photosynthesis. cAMP levels were relatively low when samples were incubated in darkness, or exposed to red or far-red light. Irradiation with red+far-red light induced a moderate increase in cAMP levels, while white light induced a pronounced increase in cAMP levels. When incubated under increasing white light irradiance, cAMP levels closely followed the increase in photosynthetic oxygen evolution rate, suggesting a direct relationship between photosynthesis and cAMP accumulation. cAMP levels were not dependent on cellular ATP concentration, as inhibitors of ATP synthesis did not significantly affect cAMP levels in light. We conclude that cAMP depends on photosynthetic activity regardless of ATP synthesis and concentration or phytochrome activity. 相似文献

To implement and validate an analytical method by ultra‐performance liquid chromatography–tandem mass spectrometry (UPLC MS/MS) to quantify mycophenolic acid (MPA) in kidney transplant patients. Quantification of MPA was performed in an ACQUITY UPLC H Class system coupled to a Xevo TQD detector and it was extracted from plasma samples by protein precipitation. The chromatographic separation was achieved through an ACQUITY HSS C18 SB column with 0.1% formic acid and acetonitrile (60:40 vol/vol) as mobile phase. The pharmacokinetic parameters were calculated by non‐compartmental analysis of MPA plasma concentrations from 10 kidney transplant patients. The linear range for MPA quantification was 0.2–30 mg/L with a limit of detection of 0.07 mg/L; the mean extraction recovery was 99.99%. The mean intra‐ and inter‐day variability were 2.98% and 3.4% with a percentage of deviation of 8.4% and 6.6%, respectively. Mean maximal concentration of 10 mg/L at 1.5 h, area under the concentration–time curve of 36.8 mg·h/L, elimination half‐life of 3.9 h, clearance of 0.32 L/h/kg and volume of distribution of 1.65 L/kg were obtained from MPA pharmacokinetics profiles. A simple, fast and reliable UPLC–MS/MS method to quantify MPA in plasma was validated and has been applied for pharmacokinetic analysis in kidney transplant patients. 相似文献
