A brief historical background of the development of the theranostic approach in nuclear medicine is given and seven theranostic pairs of radionuclides, namely 44gSc/47Sc, 64Cu/67Cu, 83Sr/89Sr, 86Y/90Y, 124I/131I, 152Tb/161Tb and 152Tb/149Tb, are considered. The first six pairs consist of a positron and a β?-emitter whereas the seventh pair consists of a positron and an α-particle emitter. The decay properties of all those radionuclides are briefly mentioned and their production methodologies are discussed. The positron emitters 64Cu, 86Y and 124I are commonly produced in sufficient quantities via the (p,n) reaction on the respective highly enriched target isotope. A clinical scale production of the positron emitter 44gSc has been achieved via the generator route as well as via the (p,n) reaction, but further development work is necessary. The positron emitters 83Sr and 152Tb are under development. Among the therapeutic radionuclides, 89Sr, 90Y and 131I are commercially available and 161Tb can also be produced in sufficient quantity at a nuclear reactor. Great efforts are presently underway to produce 47Sc and 67Cu via neutron, photon and charged particle induced reactions. The radionuclide 149Tb is unique because it is an α-particle emitter. The present method of production of 152Tb and 149Tb involves the use of the spallation process in combination with an on-line mass separator. The role of some emerging irradiation facilities in the production of special radionuclides is discussed.  相似文献

Ruthenium nanoparticles prepared in nitrile-functionalised ionic liquids (ILs) display unusual selectivities toward the hydrogenation of nitrile containing aromatic compounds. In particular, a selective catalytic hydrogenative coupling of nitriles was observed. In this transformation, nitrile groups are exclusively hydrogenated in the presence of arenes, which are typically hydrogenated by ruthenium nanoparticles in non-functionalised ILs. The catalyst material was characterised by means of TEM and EDS analysis. Furthermore, molecular species formed during the catalytic process were characterised by MS-analysis of the gaseous phase and the ionic liquid phase by ESI/MS Q-TOF.  相似文献

Mass spectrometry has proven to be an indispensable tool for protein identification, characterization, and quantification. Among the possible methods in quantitative proteomics, stable isotope labeling by using reductive dimethylation has emerged as a cost-effective, simple, but powerful method able to compete at any level with the present alternatives. In this review, we briefly introduce experimental and software methods for proteome analysis using dimethyl labeling and provide a comprehensive overview of reported applications in the analysis of (1) differential protein expression, (2) posttranslational modifications, and (3) protein interactions.  相似文献

The scope of the work was to investigate the influence of selenate fertilisation and the addition of symbiotic fungi (mycorrhiza) to soil on selenium and selenium species concentrations in garlic. The selenium species were extracted from garlic cultivated in experimental plots by proteolytic enzymes, which ensured liberation of selenium species contained in peptides or proteins. Separate extractions using an aqueous solution of enzyme-deactivating hydroxylamine hydrochloride counteracted the possible degradation of labile selenium species by enzymes (such as alliinase) that occur naturally in garlic. The selenium content in garlic, which was analysed by ICP–MS, showed that addition of mycorrhiza to the natural soil increased the selenium uptake by garlic tenfold to 15 μg g−1 (dry mass). Fertilisation with selenate and addition of mycorrhiza strongly increased the selenium content in garlic to around one part per thousand. The parallel analysis of the sample extracts by cation exchange and reversed-phase HPLC with ICP–MS detection showed that γ-glutamyl-Se-methyl-selenocysteine amounted to 2/3, whereas methylselenocysteine, selenomethionine and selenate each amounted to a few percent of the total chromatographed selenium in all garlic samples. Se-allyl-selenocysteine and Se-propyl-selenocysteine, which are selenium analogues of biologically active sulfur-containing amino acids known to occur in garlic, were searched for but not detected in any of the extracts. The amendment of soil by mycorrhiza and/or by selenate increased the content of selenium but not the distribution of detected selenium species in garlic. Finally, the use of two-dimensional HPLC (size exclusion followed by reversed-phase) allowed the structural characterisation of γ-glutamyl-Se-methyl-selenocysteine and γ-glutamyl-Se-methyl-selenomethionine in isolated chromatographic fractions by quadrupole time-of-flight mass spectrometry.  相似文献

To understand how proteins perform their function, knowledge about their structure and dynamics is essential. Here we use a combination of an efficient chemical lysine acetylation reaction and nanoLC-MALDI tandem mass spectrometry to probe the accessibility of every lysine residue in a protein complex. To demonstrate the applicability of this approach, we studied the interaction between the DNase domain of Colicin E9 (E9) and its immunity protein Im9. Free E9 and E9 in complex with Im9 were rapidly acetylated, followed by proteolytic digestion and analysis by LC-MALDI-TOF/TOF MS/MS. Acetylated peptides could be filtered out of the complex peptide mixtures using selective ion chromatograms of the specific immonium marker ions. Additionally, isobaric acetylated peptides, acetylated at different sites, could be separated by their LC retention times. The combination of LC and MALDI-TOF/TOF MS/MS provided information about the amount of acetylation on each individual lysine even for peptides containing several lysine residues. In general, our data agree well with those derived from the crystal structure of E9 and the E9:Im9 complex. Interestingly, next to in the binding interface expected lysines, K89 and K97, two from the crystal structure data unexpected lysines, K81 and K76, were observed to become less exposed upon Im9 binding. Moreover, K55 and K63, positioned in the predicted DNA binding region, were also found to be less accessible upon Im9 binding. These findings may illustrate some of the described differences in the solution-phase structure of the E9:Im9 complex compared with the crystal structure.  相似文献

Cleaning of paper objects represents one of the most complex cases of laser ablation, since low volumes of dispersed material phases are evaporated while a sensitive and fragile fibrous organic matrix has to be preserved. Conventional chemical and mechanical cleaning methods suffer from the common phenomenon that the foreign matter is diluted into the substrate rather than removed. The application of a laser beam allows highly localized and optically specific interaction. However, the occurrence of extreme temperatures and light intensities may cause irreversible alteration of the paper matrix. Further, incomplete removal and/or chemical conversion of contaminations may result in insufficient cleaning or affect the ageing behaviour. Laser treatments were performed by Q-switched Nd:YAG lasers at three wavelengths (355 nm, 532 nm, and 1064 nm). Papers contaminated with inks and adhesive-tape remnants served as model samples. The radiochemical separation of radiogallium from radiogermanium was studied using ion-exchange chromatography (Amberlite IR-120) and solvent extraction (Aliquat 336 in o-xylene). Both Amberlite IR-120 and Aliquat 336 in o-xylene have been used for the first time in separations involving radiogallium and radiogermanium. For tracer studies the radionuclides 68Ge (t 1/2 = 270.8 days), 69Ge (t 1/2 = 39 h) and 67Ga (t 1/2 = 78.3 h) were used. They were produced by the nuclear reactions natGa(p,xn)68,69Ge and natZn(p,xn)67Ga, respectively, and separated from their target materials in no-carrier-added form. Several factors affecting the separation of radiogallium from radiogermanium were studied and for each procedure the optimum conditions were determined. The solvent extraction using Aliquat 336 was found to be better. The separation yield of radiogallium was >95%, the time of separation short, the contamination from radiogermanium <0.008% and the final product was obtained in 0.5 M KOH. This method was adapted to the separation of n.c.a. 68Ga from its parent n.c.a. 68Ge. The quality of the product thus obtained is discussed.  相似文献
