Oxygenase‐Catalyzed Desymmetrization of N,N‐Dialkyl‐piperidine‐4‐carboxylic Acids

γ‐Butyrobetaine hydroxylase (BBOX) is a 2‐oxoglutarate dependent oxygenase that catalyzes the final hydroxylation step in the biosynthesis of carnitine. BBOX was shown to catalyze the oxidative desymmetrization of achiral N,N‐dialkyl piperidine‐4‐carboxylates to give products with two or three stereogenic centers.     

Kinetics and mechanisms of hydrolysis and aminolysis of thioxocephalosporins

The effect of replacing the beta-lactam carbonyl oxygen in cephalosporins by sulfur on their reactivity has been investigated. The second-order rate constant for alkaline hydrolysis of the sulfur analogue is 2-fold less than that for the natural cephalosporin. The thioxo derivative of cephalexin, with an amino group in the C7 side chain, undergoes beta-lactam ring opening with intramolecular aminolysis by a reaction similar to that for cephalexin itself. However, the rate of intramolecular aminolysis for the S-analogue is 3 orders of magnitude greater than that for cephalexin. Furthermore, unlike cephalexin, intramolecular aminolysis in the S-analogue occurs up to pH 14 with no competitive hydrolysis. The rate of intermolecular aminolysis of natural cephalosporins is dominated by a second-order dependence on amine concentration, whereas that for thioxocephalosporins shows only a first-order term in amine. The Bronsted beta(nuc) for the aminolysis of thioxo-cephalosporin is +0.39, indicative of rate-limiting formation of the tetrahedral intermediate with an early transition state with relatively little C-N bond formation.     

CH-stretching overtone spectroscopy of 1,1,1,2-tetrafluoroethane

We have recorded the vibrational absorption spectrum of 1,1,1,2-tetrafluoroethane (HFC-134a) in the fundamental and first five CH-stretching overtone regions with the use of Fourier transform infrared, dispersive long-path, intracavity laser photoacoustic, and cavity ringdown spectroscopies. We compare our measured total oscillator strengths in each region with intensities calculated using an anharmonic oscillator local mode model. We calculate intensities with 1D, 2D, and 3D Hamiltonians, including one or two CH stretches and two CH stretches with the HCH bending mode, respectively. The dipole moment function is calculated ab initio with self-consistent-field Hartree-Fock and density functional theories combined with double- and triple-zeta-quality basis sets. We find that the basis set choice affects the total intensity more than the choice of the Hamiltonian. We achieve agreement between the calculated and measured total intensities of approximately a factor of 2 or better for the fundamental and first five overtones.     

Fast phase unwrapping algorithm for interferometric applications

A wide range of interferometric techniques recover phase information that is mathematically wrapped on the interval (-pi, pi). Obtaining the true unwrapped phase is a longstanding problem. We present an algorithm that solves the phase unwrapping problem, using a combination of Fourier techniques. The execution time for our algorithm is equivalent to the computation time required for performing eight fast Fourier transforms and is stable against noise and residues present in the wrapped phase. We have extended the algorithm to handle data of arbitrary size. We expect the state of the art of existing interferometric applications, including the possibility for real-time phase recovery, to benefit from our algorithm.     

Iron‐Mediated Cleavage of C?C Bonds in Vicinal Tricarbonyl Compounds in Water

Three of a kind : Vicinal tricarbonyl compounds undergo C? C cleavage mediated by ferric ions (see scheme). The observed cleavage of ninhydrin and dehydroascorbic acid has relevance for amino acid detection and the metabolism of vitamin C.

     

Stable jets of viscoelastic fluids and self-assembled cylindrical capsules by hydrodynamic focusing

We demonstrate formation of long-lived cylindrical jets of a viscoelastic fluid using hydrodynamic focusing. A solution of polyacrylamide in water is driven coaxially with immiscible oil and subjected to strong extensional flow. At high flow rates, the aqueous phase forms jets that are 4-90 microm in diameter and several centimeters long. The liquid surfaces of these jets are then used as templates for assembly of microspheres into novel rigid and hollow cylinders.     

Assessing the reproducibility of an analytical method

The reproducibility of a validated analytical method may require reassessment because of various reasons, such as the transfer between laboratories or companies, changes in the instruments or software platforms (or both), or changes in critical reagents, among others. This paper is a demonstration of an assay bridging study in evaluating reproducibility. The approach is simple but very informative and offers many advantages over existing approaches.     

Rewritable DNA microarrays

Thiol-terminated single-stranded deoxyribonucleic acids (ssDNA) can be immobilized onto pulsed plasma deposited poly(allylmercaptan) surfaces via disulfide bridge chemistry and are found to readily undergo nucleic acid hybridization. Unlike other methods for oligonucleotide attachment to solid surfaces, this approach is shown to be independent of substrate material or geometry, and amenable to highly efficient rewriting.
