We have so far demonstrated that well-resolved and site-specifically assigned (13)C peaks as recorded by site-directed NMR study on (13)C-labeled membrane proteins can serve as a convenient probe to reveal their local conformation and dynamics. We attempted here to clarify the extent to which (13)C NMR spectra of (13)C-labeled fully hydrated bacteriorhodopsin (bR) as a typical membrane protein are visible or well resolved in the presence of inherent fluctuation motions with frequency of 10(2)-10(8) Hz, especially at the membrane surfaces. Accordingly, we estimated the relative proportion of (13)C NMR signals from the surface areas with and without peak suppression by the accelerated transverse relaxation effect by surface-bound Mn(2+) ions, which could be effective for residues within 8.7 angstroms of the membrane surface. It turned out that the experimental findings are consistent with the predicted amount of amino acid residues under consideration located within 8.7 angstroms of the surface for [1-(13)C]Val- and Ile-labeled bR and also [3-(13)C]Ala-bR. In contrast, (13)C NMR peaks from such surfaces area are almost completely or partially suppressed for [1-(13)C]Gly-, Ala-, Leu-, Phe- and Trp-labeled bR, as a result of plausible interference of the fluctuation frequency with frequency of magic angle spinning (10(4) Hz). We further assigned several (13)C NMR signals of [1-(13)C] Val-, Trp- and Ile-labeled bR on the basis of a variety of site-directed mutants with reference to those of the wild type. Further, we recorded the (13)C NMR of bR in lipid bilayers to search for the optimal conditions to be able to obtain signals with the highest peak intensities and spectral resolution. Backbone dynamics turn out to be essential for recording (13)C NMR spectra so as to escape from motional frequencies of the order of 10(4)-10(5) Hz, either in the direction of accelerated fluctuation or slowed motions in the direction of forming the 2D array.  相似文献

Two approaches for harvesting solar light energy effectively using oxide semiconductor materials are introduced. The one is water splitting using new types of oxide semiconductor photocatalyst systems. By Na2CO3 addition method, it was firstly demonstrated that water is decomposed to H2 and O2 steadily and stoichiometrically using NiO/TiO2 photocatalyst under the solar light. A new two-step water splitting system using photocatalysis is also introduced. The other is dyesensitized oxide semiconductor solar cells in order to convert visible light energy to electricity. Combinations of various types of oxide semiconductors and organic dyes, such as Eosin-Y, suggests the appearance of promising and cheap solar cells.  相似文献

Magnetically aligned bicelles are increasingly being used as model membranes in solution- and solid-state NMR studies of the structure, dynamics, topology, and interaction of membrane-associated peptides and proteins. These studies commonly utilize the PISEMA pulse sequence to measure dipolar coupling and chemical shift, the two key parameters used in subsequent structural analysis. In the present study, we demonstrate that the PISEMA and other rotating-frame pulse sequences are not suitable for the measurement of long-range heteronuclear dipolar couplings, and that they provide inaccurate values when multiple protons are coupled to a 13C nucleus. Furthermore, we demonstrate that a laboratory-frame separated-local-field experiment is capable of overcoming these difficulties in magnetically aligned bicelles. An extension of this approach to accurately measure 13C-31P and 1H-31P couplings from phospholipids, which are useful to understand the interaction of molecules with the membrane, is also described. In these 2D experiments, natural abundance 13C was observed from bicelles containing DMPC and DHPC lipid molecules. As a first application, these solid-state NMR approaches were utilized to probe the membrane interaction of an antidepressant molecule, desipramine, and its location in the membrane.  相似文献

The heat capacity of [Fe(2-pic)3]Cl2·C2H5OH Crystal (2-pic: 2-picolylamine) has been measured with an adiabatic calorimeter between 13 and 315 K. Two phase transitions centered at 114.04 and 122.21 K were observed. This finding accords with recent prediction of possible existence of two-step spin-conversion (H. Köppen et al., Chem. Phys. Lett., 91 (1982) 348). The total transition enthalpy and entropy amounted to ΔH = 6.14 kJ mol?1 and ΔS = 50.59 J K?1 mol?1. The transition entropy consists of the magnetic contribution (13.38 J K?1 mol?1), the orientational order-disorder phenomenon of the solvate ethanol molecule (8.97) and the change in the phonon system, in particular the change in stretching and deformation vibrations of the metal-ligand (28.24).  相似文献

A highly sensitive determination of fluorine in standard rocks by photon activation using the19F(,n)18F reaction combined with pyrohydrolysis for the separation of18F has been reported. The irradiation energy was operated at 20 MeV to avoid the interference from Na, because Na is one of the major element in rocks and18F is also produced from Na via23Na(,n)18F reaction above its threshold energy, 20.9 MeV. After irradiation, fluorine was extracted by pyrohydrolysis and separated as LaF3 precipitate. It was ascertained that the average recovery of fluorine in standard rocks was about 90% and the precipitate was of high radiochemical purity. This method was applied to the analysis of ten GSJ rock reference samples and two USGS standard rocks issued by the Geological Survey of Japan and the United States Geological Survey, respectively. The detection limit of this method was 0.02 g/g, and the results obtained by this method were in good agreement with the recommended values. This method was easily applied to the determination of a few ppm level of fluorine in rock samples, such as ultrabasic rock and feldspar.  相似文献
