A simple and inexpensive method for immobilizing alpha-amylase from Bacillus circulans GRS 313 on coconut fiber was developed. The immobilization conditions for highest efficiency were optimized with respect to immobilization pH of 5.5, 30 degrees C, contact time of 4 h, and enzyme to support a ratio of 1:1 containing 0.12 mg/mL of protein. The catalytic properties of the immobilized enzyme were compared with that of the free enzyme. The activity of amylase adsorbed on coconut fiber was 38.7 U/g of fiber at its optimum pH of 5.7 and 48 degrees C, compared with the maximum activity of 40.2 U/mL of free enzyme at the optimum pH of 4.9 and 48 degrees C. The reutilization capacity of the immobilized enzyme was up to three cycles.  相似文献

Summary Xylenol orange reacts very sensitively with gallium(III), indium (III) and thallium(III) to form reddish violet colored chelates having max 560 nm in case of Ga and In and max 590 nm in case of Ti at PH 4.0. The molar ratio for all the chelates is 1 1 (metal reagent). Optimum conditions including the range for adherence to Beer's law, effect of PH on the color intensity, effect of excess reagent, and sensitivity are reported for the photometric determination of these metal ions using Xylenol orange.
Zusammenfassung Xylenolorange reagiert sehr empfindlich mit Gallium(III), Indium(III) und Thallium(III) unter Bildung rötlich-violetter Chelate mit einem Absorptionsmaximum bei 560 nm im Falle von Ga und In bzw. 590 nm für Tl bei pH 4,0. Das Molverhältnis ist in jedem Fall 11. Die besten Arbeitsbedingungen, der Gültigkeitsbereich des Beerschen Gesetzes, der Einfluß des PH auf die Farbintensität und des Reagensüberschusses sowie die Empfindlichkeit werden für die photometrische Bestimmung der genannten Ionen angegeben.

We have reported herein the synthesis of three new Cu(II) complexes of tri- and tetradentate Schiff base ligands containing N3 or N4 donor set along with terminal NNN or SCN ligands: [L1Cu(NCS)]ClO4 (1), [L2Cu(NCS)2] (2) and [L3Cu(NNN)]ClO4 (3) [L1 = NC5H4C(CH3)=N(CH2)3N=C(CH3)C5H4N, L2= Me2N–(CH2)3–N=C(CH3)C5H4N and L3 = NC5H4CH=N–(CH2)4–N=CHC5H4N]. The complexes have been systematically characterised by elemental, spectroscopic and electrochemical techniques. Antimicrobial activities of the Schiff base ligands and their metal complexes have been studied using the disc diffusion method on the strains of Candida tropicalis and Bacillus megaterium. Structures of all the complexes have been unequivocally established from single crystal X-ray diffraction analyses that show the monomeric units containing a five-coordinated copper center in highly distorted square pyramidal geometry with thiocyanate or azide anion coordinated as terminal ligand. The complexes 1 and 3 crystallise in monoclinic (P21/c) and 2 in triclinic (P-1) space group, respectively.  相似文献

The feasibility of remote sensing by means of two-photon excited molecular fluorescence was investigated by using a single 115-m multimode optical fibr.Because of modal dispersion and reflecton of Rayleigh scatter, time-resolved detection ws required to resolve the weak fluorescence from the relatively intense laser background. The bare-ended probe, which both delivered the excitation and collected the fluorescence emission, was employed to detect an analyte in an optically dense environment. For a fixed amount of fluorophore, the solvent was continuously changed from 90% (w/w) ethanol (non-absorbing) to 100% acetone (absorbing). The result was a flurorescence signal constant to within 5%. Quantitation via one-photon excitatation using a 50-cm optical fiber demonstrated the characteristic inner filter roll-off of optically dense systems. Because of th elarge attenuation of the waveguide in the ultraviolet range, remote sensing via one-photon excited fluorescence proved impossible. However, linear calibration plots for the nonlinear excitation process were obtained from micromolar to near saturation levels. Sources of interference were isolated, and the extension to a single-mode fiber was made.  相似文献

Summary [NiL2X2] (L =N,N-dimethyl-1,2-ethanediamine; X = Cl, CF3CO 2 , CC13CO 2 and CBr3CO 2 ), [NiL2C2O4] · H2O and [NiL2X2] · 2 H2O (X = Br, 0.5 SO 4 2– and 0.5 SeO 4 ) have been synthesised and their thermal studies carried out. Thermally induced phase transition phenomena are noticed in [NiL2X2] (X = CF3CO 2 and CCl3CO 2 ) and their probable mechanisms are described. [NiL2X2] (X = Br, 0.5 SO 4 2– and 0.5 SeO 4 2– ) and [NiLX2] (X = Cl, 0.5 C2O 4 2– and 0.5 SO 4 2– ) have been prepared by solid state pyrolysis from the respective parent diamine complexes. [NiL2X2] have been made in solid state by temperature arrest technique from [NiL2(CX3CO2)2] (X = Cl and Br).  相似文献

Summary The formation of ternary complexes of the MAL3– type [where M = CuII, NiII and ZnII ; A = nitrilotriacetic acid (NTA); L = 1-hydroxy-2-naphthoic acid (1,2 HNA) and 2-hydroxy-1-naphthoic acid (2,1 HNA)] have been studied potentiometrically in 50% v/v aqueous — ethanol (25° and µ = 0.1). Under identical conditions the binary complexes of the 1,2- and 2,1-HNA ligands have also been examined. The values of mixed ligand formation constants KMAL have been found to be lower than KML (first step formation constant of binary complexes) and even less than (second step formation constant of binary complexes).  相似文献

Summary Chrome Azurol S permits a simple colorimetric determination of palladium with a sensitivity of 0.4g/cm2 (practical) or 0.04g/cm2 (Sandell), respectively. Conditions and interferences are described in detail.
Zusammenfassung Palladium läßt sich mit Hilfe von Chromazurol S auf einfache Weise kolorimetrisch bestimmen. Die praktische Empfindlichkeit der Methode beträgt 0,4g/cm2 bzw. 0,04g/cm2 nachSandell. Die Arbeitsbedingungen und die durch Fremdionen verursachten Störungen werden angegeben.

Résumé Le chromazurol S permet un dosage colorimétrique simple du palladium, avec une sensibilité de 0,4g/cm2 (en pratique) ou 0,04g/cm2 (Sandell) respectivement. On décrit en détail les conditions opératoires et les interférences.

Reactions of e aq , H atom and OH radicals with 4-pyridinemethanol (4-PM) and 4-pyridinecarboxaldehyde (4-PCA) have been studied at various pH values using the pulse radiolysis technique. Reaction of e aq with 4-PM and 4-PCA leads to the formation of pyridinyl and ketyl radicals of 4-PM and 4-PCA, respectively. Ketyl radicals formed from 4-PCA react with the parent molecule to give a dimeric radical species. At pH 7, the equilibrium constant for the dimer formation was determined to be 13 500 M-1. At pH 13 also dimer radical formation was observed. Reaction of e aq with 4-PM was found to give highly reducing pyridinyl radicals. Reaction of OH radicals with 4-PM gives a mixture of species,viz., OH adducts and radicals formed by H-atom abstraction from the –CH2OH moiety. Radicals formed by H-atom abstraction reaction from 4-PM were found to be reducing in nature. O radicals were found to react with 4-PM exclusively by H-abstraction pathway.  相似文献

13C, 14N, 15N, 17O, and 35Cl NMR parameters, including chemical shift tensors and quadrupolar tensors for 14N, 17O, and 35Cl, are calculated for the crystalline forms of various amino acids under periodic boundary conditions and complemented by experiment where necessary. The 13C shift tensors and 14N electric field gradient (EFG) tensors are in excellent agreement with experiment. Similarly, static 17O NMR spectra could be precisely simulated using the calculation of the full chemical shift (CS) tensors and their relative orientation with the EFG tensors. This study allows correlations to be found between hydrogen bonding in the crystal structures and the 17O NMR shielding parameters and the 35Cl quadrupolar parameters, respectively. Calculations using the two experimental structures for L-alanine have shown that, while the calculated isotropic chemical shift values of 13C and 15N are relatively insensitive to small differences in the experimental structure, the 17O shift is markedly affected.  相似文献

Summary A nitrate-selective electrode based on precipitated nitron nitrate in an epoxy resin matrix has been prepared. The lower detection limit is 10–5 M, the optimum pH range is 2.0–8.5, and the response time is 1 min. The electrode is selective in presence of CH3COO, SO4 2–, C2O4 2–, PO4 3–, NO2 , Cl and Br, but I interferes. Results for nitrate in soil samples were in agreement with values obtained spectrophotometrically.

Zusammenfassung Eine nitrat-spezifische Elektrode auf der Basis gefällten Nitronnitrats in einer Epoxyharz-Matrix wurde hergestellt. Die untere Nachweisgrenze ist 10–5 M, das pH-Optimum liegt zwischen 2,0 und 8,5, die Reaktionszeit beträgt 1 Minute. Die Elektrode ist selektiv in Gegenwart von Acetat, Sulfat, Oxalat, Phosphat, Nitrit, Chlorid und Bromid; aber Jodid stört. Ergebnisse der Nitratbestimmung in Bodenproben stimmen mit spektrophotometrischen Resultaten überein.
