A novel core structure among bacterial lipopolysaccharides (LPS) that belong to the genus Halomonas has been characterized. H. stevensii is a moderately halophilic microorganism, as are the majority of the Halomonadaceae. It brought to light the pathogenic potential of this genus. On account of their role in immune system elicitation, elucidation of LPS structure is the mandatory starting point for a deeper understanding of the interaction mechanisms between host and pathogen. In this paper we report the structure of the complete saccharidic portion of the LPS from H. stevensii. In contrast to the finding that the O‐antigen is usually covalently linked to the outer core oligosaccharide, we could demonstrate that the O‐polysaccharide of H. stevensii is linked to the inner core of an LPS. By means of high‐performance anion‐exchange chromatography with pulsed amperometric detection we were able to isolate the core decasaccharide as well as a tridecasaccharide constituted by the core region plus one O‐repeating unit after alkaline degradation of the LPS. The structure was elucidated by one‐ and two‐dimensional NMR spectroscopy, ESI Fourier transform ion cyclotron resonance (FT‐ICR) mass spectrometry, and chemical analysis.  相似文献

A constructive definition of the continuum based on formal topology is given and its basic properties studied. A natural notion of Cauchy sequence is introduced and Cauchy completeness is proved. Other results include elementary proofs of the Baire and Cantor theorems. From a classical standpoint, formal reals are seen to be equivalent to the usual reals. The implementation of a quality system based on the ISO/IEC 17025:1999 standard is a growing necessity for analytical laboratories to demonstrate their technical competence. In 2001, the Nuclear Analytical Techniques Group of the Argentine Atomic Energy Commission obtained the recognition of the International Atomic Energy Agency in the application of neutron activation analysis and the accreditation by the national accreditation body. The importance of the participation of the group in the Agency's Regional Programme for Latin America, ARCAL XXVI on Quality Assurance in Analytical laboratories is discussed, as well as the activities performed to attain these objectives. Some improvements worth mentioning resulted from the implementation of the quality system and, following the premise of continuous improvement, changes were introduced aiming at the laboratory re-accreditation.  相似文献

Ingestion of relatively small amounts of lead is now recognized to cause significant neurological and cognitive effects in humans. Large quantities may be fatal, yet lead poisoning, especially of children, is still a major public health concern in many parts of the world. In rural Mexican communities lead oxide (PbO) is added to ceramic glaze as a fluxing agent, lowering starting firing temperatures to 500 °C. The purpose of this study is to characterize the lead chemical forms in ceramic glazes from the Solis Valley, Mexico, to investigate lead leaching properties of these ceramics, and to demonstrate the applicability of lead isotope signatures as a means of tracing lead source origins. Ceramics were collected from the rural village of Santa Maria de Canchesda, State of Mexico, Mexico. Dried liquid glazes, post-fired glaze material, and pure PbO were analyzed by diffuse reflectance infrared Fourier transform spectroscopy (DRIFT). Results from DRIFT analysis indicate that PbO (1429 cm−1 band) is the active form of lead found in liquid glazes and ceramics. Some shifting of 1429 cm−1 PbO peak to lower wavenumbers occurs in post-fired ceramics, and this may be due to the formation of lead bisilicate during firing. Ceramics samples were leached in 0.02 M citric acid solution for 1 min, and leached lead concentrations were measured using inductively coupled plasma-mass spectrometry (ICP-MS). Lead concentrations in these leachates varied from 0.4-80.4 μg ml−1, while the control pottery from the US leached only 0.1 μg ml−1 lead. Elemental distributions on glaze surfaces were identified by laser ablation (LA)-ICP-MS. Nitric acid extracts of soils, teeth, and ceramic glazes were analyzed for lead isotope ratios (207Pb/206Pb vs. 208Pb/206Pb) using ICP-MS. Similarities of tooth and ceramic lead isotope ratios indicate that ceramics may be a substantial source of body lead burden in the Solis Valley. This study demonstrates the applicability of lead isotope ratios for lead source identification, and it identifies potential health risks from ceramic use induced lead toxicity within the Solis Valley.  相似文献

In the free-energy landscape picture of glassy systems, their slow dynamics is due to a complicated free-energy landscape with many local minima. We show that for a colloidal glassy material multiple paths can be taken through the free-energy landscape. The evolution of the nonergodicity parameter shows two distinct master curves that we identify as gels and glasses. We show that for a range of colloid concentrations, the transition to nonergodicity can occur in either direction (gel or glass), accompanied by "hesitations" between the two. Thus, colloidal gels and glasses are merely global free-energy minima in the same free-energy landscape, and the paths leading to these minima can be complicated.  相似文献
