Cervical cancer is known to be highly associated with viral oncogene E6 and E7 of human papilloma virus. Down-regulation of oncogene expression by antisense-based gene therapy has been extensively studied. To investigate the effect of HPV 16 E6 antisense nucleic acid (AS) on cervical cancer cells, human cervical cancer cell lines, CaSki and SiHa cells harboring HPV 16 genome were transfected with plasmid containing E6(AS). The decreased viability and the apoptotic morphology were observed in E6(AS)-transfected cervical cancer cell lines. By 6 h after transfection, inhibition of E6 splicing, rapid upregulations of p53 and a p53-responsive protein, GADD45, were displayed in E6(AS)-transfected CaSki cells. Furthermore, E6(AS) induced loss of mitochondrial transmembrane potential, release of mitochondrial cytochrome c into the cytoplasm, and subsequent activation of caspase-9 and caspase-3. These results indicate that HPV 16 E6(AS) induces apoptosis in CaSki cells via upregulation of p53 and release of cytochrome c into cytoplasm, consequently activating procaspase-9 and procaspase-3.  相似文献

Osteosarcoma is the most common primary bone tumor, but the pathogenesis is not well understood. While cyclooxygeanse-2 (COX-2) is known to be closely associated with tumor growth and metastasis in several kinds of human tumors, the function of COX-2 in osteosarcoma is unclear. Therefore, to investigate the function of COX-2 in osteosarcoma, we established stable cell lines overexpressing COX-2 in U2OS human osteosarcoma cells. COX-2 overexpression as well as prostaglandin E2 treatment promoted proliferation of U2OS cells. In addition, COX-2 overexpression enhanced mobility and invasiveness of U2OS cells, which was accompanied by increases of matrix metalloproteinase-2 and -9 (MMP-2 and -9) activities. Selective COX-2 inhibitors, NS-398 and celecoxib, inhibited cell proliferation and abrogated the enhanced mobility, invasiveness and MMP activities induced by COX-2 overexpression. These results suggest that COX-2 is directly associated with cell proliferation, migration and invasion in human osteosarcoma cells, and the therapeutic value of COX-2 inhibitors should be evaluated continuously.  相似文献

Circadian clocks are the endogenous oscillators that harmonize a variety of physiological processes within the body. Although many urinary functions exhibit clear daily or circadian variation in diurnal humans and nocturnal rodents, the precise mechanisms of these variations are as yet unclear. In the present study, we demonstrate that Per2 promoter activity clearly oscillates in neonate and adult bladders cultured ex vivo from Per2::Luc knock-in mice. In subsequent experiments, we show that multiple local oscillators are operating in all the bladder tissues (detrusor, sphincter and urothelim) and the lumbar spinal cord (L4–5) but not in the pontine micturition center or the ventrolateral periaqueductal gray of the brain. Accordingly, the water intake and urine volume exhibited daily and circadian variations in young adult wild-type mice but not in Per1−/−Per2−/− mice, suggesting a functional clock-dependent nature of the micturition rhythm. Particularly in PDK mice, the water intake and urinary excretion displayed an arrhythmic pattern under constant darkness, and the amount of water consumed and excreted significantly increased compared with those of WT mice. These results suggest that local circadian clocks reside in three types of bladder tissue and the lumbar spinal cord and may have important roles in the circadian control of micturition function.  相似文献

As typical emission spectroscopy involves chord integration along the line of sight, a local measurement with high spatial resolution is attempted using simple lens optics in this work. In the experiment, chord integrated optical plasma emission profile was measured by moving a scanning lens located outside the plasma. The measured emission intensities were spatially reconstructed by employing a geometry-dependent system function, and the local (i.e., only from the lens focal point) emission intensities were obtained with all out-focused emissions subtracted. The 34 different Ar I emission lines spatially reconstructed in this way were used to determine excitation temperature (Texc) of the argon plasma by the Boltzmann plot method. Being different from the plasma driven at 13.56 MHz where a rather uniform profile was obtained, the spatial profile of Texc from the plasma driven at 90 MHz showed a hollow profile, which is similar to that of the electron temperature (Te) measured by a Langmuir probe. This hollow profile is attributed from the electromagnetic phenomena such as skin effect and standing wave effect. The similar spatial tendency of Texc and Te implies that Texc can be a representative of Te. This is particularly useful for the cases in which conventional Langmuir probe measurements are limited, such as in large size plasmas.  相似文献

Light-emitting electrochemical cells (LECs) are a promising type of electroluminescent device for display and lighting applications. In this study, LECs based on ionic iridium complexes utilizing a tetrazole based ancillary ligand were fabricated and their electrical properties were investigated. Two new iridium(III) complexes with tetrazole based ancillary ligands, namely, [Ir(ppy)2(tetrazole)]PF6 (complex 1) and [Ir(dfppy)2(tetrazole)]PF6 (complex 2) (where ppy is 2-phenylpyridine, dfppy is 2-(2,4-difluorophenyl)pyridine, tetrazole is 5-bromo-2-(2-methyl-2H-tetrazol-5-yl)-pyridine and PF6 is hexafluorophosphate), have been synthesized and characterized. These synthesized complexes were used for the fabrication of LEC devices. LECs based on complex 1 result in orange light emission (576 nm) with the Commission Internationale de l’Eclairage (CIE) coordinates of (0.45, 0.49), while complex 2 emits green (518 nm) electroluminescence with the CIE coordinates of (0.33, 0.49). Our work suggests that the light emission of cationic iridium complexes can easily be tuned by the substituents on the cyclometalated ligands.  相似文献

The semihydrogenation of alkynes into alkenes rather than alkanes is of great importance in the chemical industry. Unfortunately, state‐of‐the‐art heterogeneous catalysts hardly achieve high turnover frequencies (TOFs) simultaneously with almost full conversion, excellent selectivity, and good stability. Here, we used metal–organic frameworks (MOFs) containing Zr metal nodes (“UiO”) with tunable wettability and electron‐withdrawing ability as activity accelerators for the semihydrogenation of alkynes catalyzed by sandwiched palladium nanoparticles (Pd NPs). Impressively, the porous hydrophobic UiO support not only leads to an enrichment of phenylacetylene around the Pd NPs but also renders the Pd surfaces more electron‐deficient, which leads to a remarkable catalysis performance, including an exceptionally high TOF of 13835 h?1, 100 % phenylacetylene conversion 93.1 % selectivity towards styrene, and no activity decay after successive catalytic cycles. The strategy of using molecularly tailored supports is universal for boosting the selective semihydrogenation of various terminal and internal alkynes.  相似文献

trans-Resveratrol (t-RVT), a naturally occurring polyphenol found in Polygonum cuspidatum, grape, and red wine, has been reported to have anti-inflammatory, cardioprotective, and cancer chemopreventive properties. However antidiabetic effect of t-RVT has not yet been reported. In this study, we show that t-RVT increases glucose uptake in C2C12 myotubes by activating AMP-activated protein kinase (AMPK), uncovering an antidiabetic potential of t-RVT for the first time. AMPK plays a central role in the regulation of glucose and lipid metabolism, and hence it is considered a novel therapeutic target for metabolic syndrome such as type 2 diabetes. t-RVT significantly induced glucose uptake in C2C12 cells, via AMPK activation, but not a phosphatidylinositol-3 kinase (PI-3 kinase) signal pathway. The induced glucose uptake was attenuated by pretreatment with a pharmacological inhibitor for AMPK, indicating that the effect of t-RVT primarily depends on AMPK activation. However, in the presence of insulin, t-RVT also potentiated the effect of insulin on glucose uptake via AMPK activation, which led to further activation of PI-3 kinase/Akt signal pathway.  相似文献

In the setting of the Fock space over the complex plane, Bauer and Lee have recently characterized commutants of Toeplitz operators with radial symbols, under the assumption that symbols have at most polynomial growth at infinity. Their characterization states: If one of the symbols of two commuting Toeplitz operators is nonconstant and radial, then the other must be also radial.
