Cancer stem cells are a subpopulation of cancer cells characterized by self-renewal ability, tumorigenesis and drug resistance. The aim of this study was to investigate the role of HMGA1, a chromatin remodeling factor abundantly expressed in many different cancers, in the regulation of cancer stem cells in ovarian cancer. Spheroid-forming cancer stem cells were isolated from A2780, SKOV3 and PA1 ovarian cancer cells by three-dimensional spheroid culture. Elevated expression of HMGA1 was observed in spheroid cells along with increased expression of stemness-related genes, such as SOX2, KLF4, ALDH, ABCB1 and ABCG2. Furthermore, spheroid A2780 cells, compared with adherent cells, showed higher resistance to chemotherapeutic agents such as paclitaxel and doxorubicin. HMGA1 knockdown in spheroid cells reduced the proliferative advantage and spheroid-forming efficiency of the cells and the expression of stemness-related genes. HMGA1 overexpression in adherent A2780 cells increased cancer stem cell properties, including proliferation, spheroid-forming efficiency and the expression of stemness-related genes. In addition, HMGA1 regulated ABCG2 promoter activity through HMGA1-binding sites. Knockdown of HMGA1 in spheroid cells reduced resistance to chemotherapeutic agents, whereas the overexpression of HMGA1 in adherent ovarian cancer cells increased resistance to chemotherapeutic agents in vitro. Furthermore, HMGA1-overexpressing A2780 cells showed a significant survival advantage after chemotherapeutic agent treatment in a xenograft tumorigenicity assay. Together, our results provide novel insights regarding the critical role of HMGA1 in the regulation of the cancer stem cell characteristics of ovarian cancer cells, thus suggesting that HMGA1 may be an important target in the development of therapeutics for ovarian cancer patients.  相似文献

In recent years, fluorescent probes for the detection of environmentally and biologically important metal cations have received extensive attention for designing and development of fluorescent chemosensors. Herein, we report the photophysical results of 2-(2-fluorobenzoyl)-N-(2-methoxyphenyl) hydrazinecarbothioamide (4) functionalized as Ni (II) sensor in micromolar concentration level. Through fluorescence titration at 488 nm, we were confirmed that ligand 4 showed the remarkable emission by complexation between 4 and Ni (II) while it appeared no emission in case of the competitive ions (Cr3+, Fe2+, Co2+, Ba2+, Cu2+, Ca2+, Na+, K+, Cu+, Cs+). Furthermore, ligand 4 exhibited no toxicity with precise cell permeability toward normal living cells using L929 cell lines in bio imaging experiment investigated through confocal fluorescence microscope. The non-toxic behavior of ligand 4 (assessed by MTT assay) and its ability to track the Ni2+ in living cells suggest its possibility to use in biological system as nickel sensor.

Sn-based thin films as new buffer layer for Cd-free Cu(In,Ga)Se2 (CIGS) solar cells were developed. The Sn(O,S)2 films were formed on CIGS substrates by chemical bath deposition from an alkaline ammonia solution by reacting tin(IV) chloride with thiourea. Optimization of the growth process allowed the smooth and conformal coverage of the films on the CIGS substrates with a thickness of 20 nm that was a self-limited thickness in the chemical bath deposition process. XPS analysis revealed that the as-deposited films contained Sn–O, Sn–OH, and Sn–S bondings and the ratio of Sn–S bonding to Sn–O bonding was 0.3. The CIGS solar cell fabricated with a 20-nm thick Sn(O,S)2 buffer layer had the best efficiency of 11.5% without AR coating. The open circuit voltage, short circuit current, and fill factor were 0.55 V, 34.4 mA/cm2, and FF = 0.61, respectively. The open circuit voltage and fill factor were low compared to the conventional CIGS solar cell with a 50-nm thick CdS buffer due to too thin Sn(O,S)2 buffer layer.  相似文献

In this paper we introduce two types of q‐deformed Wigner algebras. One is q‐deformation of Wigner algebra with q‐reflection symmetry and another is q‐deformation of Wigner algebra with reflection symmetry. For two types of q‐deformed Wigner algebras, we investigate the representation and the eigenvalue of the position operator. Like q‐calculus, we introduce the (q; ν)‐ numbers, (q; ν)‐derivatives and (q; ν)‐Hermite polynomials for two algebras. For the deformation parameter q = 1 ‐ ϵ with small ϵ, we discuss the thermodynamics of the particle obeying the q‐deformed Wigner algebra.  相似文献

In this paper, we consider the problem of automatic face recognition with limited manually labeled training data. We propose a new semi-supervised self-training approach which is used to automatically augment the manually labeled training set with new unlabeled data. Semi-supervised Discriminant Analysis is used in each iteration of self-training for discriminative dimensionality reduction by making use of both labeled and unlabeled training data. Sparse representation is applied for classification. Experimental results on four independent databases show that our algorithm outperforms other face recognition methods under 3 different configurations, namely transductive, semi-supervised and single training image.  相似文献

In this paper, we present a novel method to detect license plates in video sequences automatically. The framework mainly integrates the cascade detectors method and the Tracking Learning Detection (TLD) algorithm. The cascade detectors are used to detect license plates, and the TLD algorithm is adopted to track the license plate regions. The license plates in the first frame image are detected by the cascade detectors to build the original tracking list, the tracking results and the detection results in following frames will be compared, and the newly appearing license plate information will be added to the tracking list. Meanwhile, the tracking results existing in the current tracking list would be replaced by the corresponding detection results with higher degree of confidence. We demonstrate the effectiveness of our algorithm for license plate detection task on a road intersection dataset, and the experimental evaluation shows the detection performance has been greatly improved by synthetically using the detection and tracking strategy.  相似文献

Gain and Noise figure (NF) characteristics in dual-pump parametric amplifier based on silicon on insulator (SOI) Rib waveguides are numerically investigated in the presence of nonlinear losses. The impact of structure parameters of the silicon optical parametric amplifiers (SOPAs) on the gain and the NF are also analyzed. The results show that both the height and the width of the silicon on insulator (SOI) can affect the gain and the NF of SOPAs. 354 nm bandwidth (3 dB) and 8.135 maximum gain can be achieved by tailoring the structure parameters of the SOI rib waveguides. Moreover, the dispersion and the effective mode area of SOI are also analyzed.  相似文献

We present a photodiode-based Chua's chaotic circuit that is controllable by light. The proposed circuit consists of an inductor, two passive capacitors, a photodiode-based variable resistor, and a positive feedback trans- conductor with negative nonlinearity. The chaotic dynamics of the circuit were verified by using the simulation program with integrated circuit emphasis analysis using the 0.35 #m complementary metal-oxide-semiconductor process parameters. The gain results (such as the time waveform~ frequency analysis, three-dimensional attractor, bifurcation and Lyapunov exponents diagrams) confirm that the chaotic behavior of the circuit could be controlled by light intensity via the photodiode-based variable resistor.  相似文献

A compact, highly efficient, and passively assembled parallel optical-electrical convertor module (POECM) for active optical cable application is proposed. This paper presents our POECM structure, optical design simulation results, fabrication process, and data transmission test results, in sequence. The POECM has a compact size of \(18.5\hbox {mm} \times 10\hbox {mm} \times 2.8\hbox {mm}\) . We confirm a data rate of total throughput at 21.6 Gbps ( \(5.4\hbox {Gbps} \times 4\) channels) with a bit error rate of less than \(10^{-12}\) .  相似文献

Purpose

To describe the patterns of bile distribution in the biliary tree, duodenum, jejunum, and stomach, and to determine the gallbladder ejection fraction (GBEF) by using functional magnetic resonance cholangiography (MRC) with gadolinium-ethoxybenzyl-diethylenetriamine penta-acetic acid (Gd-EOB-DTPA) in healthy volunteers.

Materials and Methods

Forty subjects were included in this study. After conventional MRC, pre-fatty meal MRC (PRFM) was obtained at 30, 40, 50, and 60 min after contrast agent injection. Then, post-fatty meal MRC (POFM) was obtained every 10 min for 1 h. We assessed the PRFM and POFM for opacification of contrast agent in the first- and second-order intrahepatic ducts (IHDs) and the common bile duct (CBD). Contrast agent opacification in the cystic duct was assessed, and the percentage volume of contrast agent filling in the gallbladder (GB) was calculated on PRFM. We calculated the GBEF and assessed the presence of contrast agent in the GB, duodenum, jejunum, and stomach.

Results

Thirty-six (90%) subjects showed grade 3 CBD opacification (visible contrast and well-defined bile duct border) on 60-min PRFM. Thirty-four (85%) subjects showed grade 3 first-order IHD opacification on 60-min PRFM. All (100%) subjects showed cystic duct opacification of contrast agent, and the average percentage volume of contrast agent filling in the GB was 68.81% ± 16.84% on 60-min PRFM. The GBEF at 30-min POFM was 35.00% ± 18.26%. Ten (25%) subjects had no contrast agent in the stomach and small bowel on all PRFMs. Twelve (30%) subjects had contrast medium in the stomach on PRFM and/or POFM.

Conclusions

Functional MRC with Gd-EOB-DTPA can allow determining the distribution of bile in the biliary tree and small intestine, as well as the GBEF.  相似文献
