The authors describe a method for the colorimetric determination of unamplified microRNA. It is based on the use of citrate-capped gold nanoparticles (AuNPs) and, alternatively, a microRNA-probe hybrid or a magnetically extracted microRNA that serve as stabilizers against the salt-induced aggregation of AuNPs. The absorbance ratios A525/A625 of the reacted AuNP solutions were used to quantify the amount of microRNA. The assay works in the range of 5–25 pmol microRNA. The lower limit of detection (LOD) is 10 pmol. The performance of the method was tested by detection of microRNA-210-3p in totally extracted urinary microRNA from normal, benign, and bladder cancer subjects. The sensitivity and specificity for qualitative detection of urinary microRNA-210-3p using the assay are 74% and 88% respectively, which is consistent with real time PCR based assays. The assay was applied to the determination of specific microRNA by using its specific oligo targeter or following magnetic isolation of the desired microRNA. The method is simple, cost-efficient, has a short turn-around time and requires minimal equipment and personnel.

Cancer is a multifactorial disease and the second leading cause of death worldwide. Diverse factors induce carcinogenesis, such as diet, smoking, radiation, and genetic defects. The phosphatidylinositol 3-kinase (PI3Kα) has emerged as an attractive target for anticancer drug design. Eighteen derivatives of N-phenyl-6-chloro-4-hydroxy-2-quinolone-3-carboxamide were synthesized and characterized using FT-IR, NMR (1H and 13C), and high-resolution mass spectra (HRMS). The series exhibited distinct antiproliferative activity (IC50 µM) against human epithelial colorectal adenocarcinoma (Caco-2) and colon carcinoma (HCT-116) cell lines, respectively: compounds 16 (37.4, 8.9 µM), 18 (50.9, 3.3 µM), 19 (17.0, 5.3 µM), and 21 (18.9, 4.9 µM). The induced-fit docking (IFD) studies against PI3Kαs showed that the derivatives occupy the PI3Kα binding site and engage with key binding residues.  相似文献

Fe was introduced in mordenite zeolite by means of ion exchange either in solid or in liquid state. The iron loading (50--200 wt%), iron precursor (FeSO4.7H2O and FeCl3), and mordenite starting material (NH4M, HM, and NaM) were varied during the exchange processes. The Fe species were characterized by N2 adsorption measurements as well as by XRD and M?ssbauer spectroscopies. The Fe-mordenite samples prepared by liquid-state ion exchange attained remarkable Fe dispersion and surface areas higher than those of the parent. It was found that Fe3+ ions, which substituted the framework Al and accordingly occupied tetrahedral sites, were decreased with Fe loadings with concomitant increase in Fe3+-occupied octahedral sites. The latter sites disappeared at 20 K to provoke the superparamagnetic alpha-Fe2O3 in different particles size. The acid leaching (0.1 M HCl, 333 K, 3 h) of the samples showed the disappearance of the most highly distorted extra-framework Fe3+ species, providing an indication of their presence on the external surface. On the other hand, a hematite phase was detected in the solid-state ion exchange of FeCl3 with either HM or NH4M at the loading of 100% Fe. More correlations between M?ssbauer data on one hand and XRD and texturing properties on the other hand were evaluated and discussed.  相似文献

Thioglycosides derivatives of N-phenylmaleimide have been prepared by the reaction of derivatives of 1-thio-d-glucopyranose, d-galactopyranose, d-lactose, and d-maltose with 3,4-dichloro-N-phenylmaleimide. The reaction of 3,4-dichloro-N-phenyl maleimide with sugar thiols (protected or unprotected) took place by displacement of both chlorine atoms by sulfide nucleophile giving the corresponding bis-thioglycoside products.  相似文献

A micelle-mediated extraction (CPE) procedure has been developed to remove trace amounts of uranium from wastewater using a non-ionic surfactant (Triton (X-100)) and lipophilic chelating extracting agent (D2EHPA) in acetate medium. The methodology used is based on the formation of metal complexes soluble in a micellar phase of a non-ionic surfactant. The uranyl ions complexes are then extracted into the surfactant-rich phase at a ambient temperature. The effects of different operating parameters such as the concentrations of Triton (X-100), D2EHPA and metal ions, temperature, sodium acetate rate and pH on the cloud point extraction of uranyl ions were studied in details and a set of optimum conditions were obtained. The results showed, without contribution of energy (ambient temperature), that up to 1000?ppm of uranyl ions can quantitatively be removed (>97?%) in a single CPE extraction using optimum conditions.  相似文献

2-Amino-1.1.3-tricyanoprop-l-ene (1) reacted with thioglycollic acid to afford the key compound for this study namely: 2-amino-I,I-dicyano-3(2-thiazolin-4-on-2-yl)prop-1-ene 2. 2 reacted with hydrazines to afford the thiazolylmethylaminopyrazole derivatives 3 and 4. The reaction of 2 with diazotised primary aromatic amines afforded coupling products which were cyclised to the hydrazothiazolonylpyridazine derivatives 7. The reaction with aromatic aldehydes gave the his-ylidene derivatives 8. Structures are assigned based on elemental and spectroscopic analysis.  相似文献

This paper presents a study of the non-linear response of a simple rigid disk-rotor, supported by active magnetic bearings (AMB), without gyroscopic effects. The case of primary resonance is examined under multi-excitation forces. The rotating shaft is described by a coupled second order non-linear ordinary differential equations. Approximate solutions are sought applying the method of multiple scales. Numerical simulations are carried out to illustrate the steady-state response and the stability of the solutions for various parameters using the frequency response function method. It is shown that the system parameters have different effects on the non-linear response of the rotor. For steady-state response, however, multiple-valued solutions and jump phenomenon occur. Results are compared to previously published work.  相似文献

The vibration of a ship pitch-roll motion described by a non-linear spring pendulum system (two degrees of freedom) subjected to multi external and parametric excitations can be reduced using a longitudinal absorber. The method of multiple scale perturbation technique (MSPT) is applied to analyze the response of this system near the simultaneous primary, sub-harmonic and internal resonance. The steady state solution near this resonance case is determined and studied applying Lyapunov’s first method. The stability of the system is investigated using frequency response equations. Numerical simulations are extensive investigations to illustrate the effects of the absorber and some system parameters at selected values on the vibrating system. The simulation results are achieved using MATLAB 7.0 programs. Results are compared to previously published work.  相似文献

Dental caries results from the bacterial pathogen Streptococcus mutans (S. mutans) and is the maximum critical reason for caries formation. Consequently, the present study aims to evaluate the antibacterial activity of a newly synthesized nanoantibiotic–Biodentine formulation. The silver nanoparticles (ROE-AgNPs) were biosynthesized from the usage of Rosmarinus officinalis L. extract (ROE) and conjugated with cefuroxime to form Cefuroxime-ROE-AgNPs. Using Biodentine™ (BIOD), five groups of dental materials were prepared, in which Group A included conventional BIOD, Group B included BIOD with ROE-AgNPs, Groups C and D included BIOD with Cefuroxime-ROE-AgNPs at concentrations of 0.5% and 1.5% cefuroxime, respectively, and Group E included BIOD with 1.5% cefuroxime. The synthesized ROE-AgNPs or Cefuroxime-ROE-AgNPs were characterized for conjugating efficiency, morphology, particle size, and in vitro release. Minimum inhibitory concentration (MIC) of the cefuroxime, ROE-AgNPs, and Cefuroxime-ROE-AgNPs were additionally evaluated against cefuroxime resistant S. mutans, which furthered antibacterial efficacy of the five groups of dental materials. The UV-Visible spectrum showed the ROE-AgNPs or Cefuroxime-ROE-AgNPs peaks and their formation displayed through transmission electron microscopy (TEM), X-ray diffraction (XRD) pattern, and Fourier transforms infrared (FTIR) analysis. The end result of Cefuroxime-ROE-AgNPs showed conjugating efficiency up to 79%. Cefuroxime-ROE-AgNPs displayed the highest antibacterial efficacy against S. mutans as compared to cefuroxime or ROE-AgNPs alone. Moreover, the MIC of ROE-AgNPs and Cefuroxime-ROE-AgNPs was detected against S. mutans to be 25 and 8.5 μg/mL, respectively. Consequently, Cefuroxime-ROE-AgNPs displayed that a decrease in the MIC reached to more than three-fold less than MIC of ROE-AgNPs on the tested strain. Moreover, Cefuroxime-ROE-AgNPs/BIOD was employed as a novel dental material that showed maximum antimicrobial activity. Groups C and D of novel materials showed inhibitory zones of 19 and 26 mm, respectively, against S. mutans and showed high antimicrobial rates of 85.78% and 91.17%, respectively. These data reinforce the utility of conjugating cefuroxime with ROE-AgNPs to retrieve its efficiency against resistant S. mutant. Moreover, the nanoantibiotic delivered an advantageous antibacterial effect to BIOD, and this may open the door for future conjugation therapy of dental materials against bacteria that cause dental caries.  相似文献

Abstract

Cyanothioacetamide (1) reacted with α- and β-naphthaldehyde 2a,b to afford the corresponding 3-naphthyl-2-thiocarboxamidopropenonitriles 3a,b. Compounds 3a,b structures could be elucidated via their reactions with acrylonitrile, ethyl acrylate (4a,b). N-arylmaleimides 6a-c and ethyl acetoacetate (8). The isolated products could be represented as the thiopyran, thiopyranopyrrolidine and pyridinethione derivatives 5a-d, 7a-f and 9a,b respectively. Pyridinethiones 9a,b had been used as the starting materials in the present study in addition to the next ones to synthesize several new thienopyridines, pyridothienotriazine and pyridothienopyrimidines 12a-f, 15a,b, 16b, 17–19a,b respectively through their reactions with the corresponding reagents.

All structures of the newly synthesized heterocyclic compounds were established on the basis of the data of IR, 1H NMR and elemental analyses.  相似文献
