A high-performance liquid chromatography protocol for the analysis of brevetoxins has been developed using a silica hydride-based cholesterol column. Brevetoxins are neurotoxins produced by harmful algae that have additional potential as drugs for a number of illnesses/diseases. To develop the optimum conditions, a number of different experimental approaches were tested. These include isocratic and gradient elution, different organic mobile phase components, and temperature variations. A separate protocol was developed for the compounds brevenal and brevenol, also produced by the same algae that make brevetoxins. Brevenal is a natural product under investigation as a therapy for chronic respiratory diseases, such as cystic fibrosis or asthma. The goal of this study was to provide a protocol for the analysis of these compounds that could be further developed into a validated method depending on a particular laboratory's capabilities and to highlight some of the unique features of the cholesterol stationary phase.  相似文献

The coenzyme A (CoA) biosynthesis pathway has attracted attention as a potential target for much-needed novel antimicrobial drugs, including for the treatment of tuberculosis (TB), the lethal disease caused by Mycobacterium tuberculosis (Mtb). Seeking to identify inhibitors of Mtb phosphopantetheine adenylyltransferase (MtbPPAT), the enzyme that catalyses the penultimate step in CoA biosynthesis, we performed a fragment screen. In doing so, we discovered three series of fragments that occupy distinct regions of the MtbPPAT active site, presenting a unique opportunity for fragment linking. Here we show how, guided by X-ray crystal structures, we could link weakly-binding fragments to produce an active site binder with a KD <20 μM and on-target anti-Mtb activity, as demonstrated using CRISPR interference. This study represents a big step toward validating MtbPPAT as a potential drug target and designing a MtbPPAT-targeting anti-TB drug.  相似文献

The new quaternary thiosilicate, Li2PbSiS4 (dilithium lead silicon tetrasulfide), was prepared in an evacuated fused‐silica tube via high‐temperature, solid‐state synthesis at 800 °C, followed by slow cooling. The crystal structure was solved and refined using single‐crystal X‐ray diffraction data. By strict definition, the title compound crystallizes in the stannite structure type; however, this type of structure can also be described as a compressed chalcopyrite‐like structure. The Li+ cation lies on a crystallographic fourfold rotoinversion axis, while the Pb2+ and Si4+ cations reside at the intersection of the fourfold rotoinversion axis with a twofold axis and a mirror plane. The Li+ and Si4+ cations in this structure are tetrahedrally coordinated, while the larger Pb2+ cation adopts a distorted eight‐coordinate dodecahedral coordination. These units join together via corner‐ and edge‐sharing to create a dense, three‐dimensional structure. Powder X‐ray diffraction indicates that the title compound is the major phase of the reaction product. Electronic structure calculations, performed using the full potential linearized augmented plane wave method within density functional theory (DFT), indicate that Li2PbSiS4 is a semiconductor with an indirect bandgap of 2.22 eV, which compares well with the measured optical bandgap of 2.51 eV. The noncentrosymmetric crystal structure and relatively wide bandgap designate this compound to be of interest for IR nonlinear optics.  相似文献

Secondary metabolome mining efforts in the myxobacterial multiproducer of natural products, Chondromyces crocatus Cm c5, resulted in the isolation and structure elucidation of crocagins, which are novel polycyclic peptides containing a tetrahydropyrrolo[2,3-b]indole core. The gene cluster was identified through an approach combining genome analysis, targeted gene inactivation in the producer, and in vitro experiments. Based on our findings, we developed a biosynthetic scheme for crocagin biosynthesis. These natural products are formed from the three C-terminal amino acids of a precursor peptide and thus belong to a novel class of ribosomally synthesized and post-translationally modified peptides (RiPPs). We demonstrate that crocagin A binds to the carbon storage regulator protein CsrA, thereby inhibiting the ability of CsrA to bind to its cognate RNA target.  相似文献

Cananodine was synthesized using a remarkable intramolecular opening of a trisubstituted epoxide as the key step in construction of the seven-membered carbocycle of the target. Abstract— Breaks induced in purified DNA by 334-nm ultraviolet (UV) radiation are enhanced 30 times when 2-thiouracil (s2Ura) is present during aerobic irradiation. This enhancement by s2Ura is maximally effective at a concentration of about 1 m M. Anoxic irradiation reduces the s2Ura-enhanced breakage by 90%, indicating a Type II photosensitization. Benzoate, glycerol, diazabicyclo[2.2.2.]octane (DABCO) and histidine all inhibit formation of s2Ura photosensitized breaks, unlike diethylenetriaminepenta-acetic acid (DETAPAC) and catalase, which do not. The relationships between the concentration of DABCO. benzoate and histidine and their protection against induction of single strand breaks (SSBs) were similar, with little inhibition below 10 m M and maximal inhibition near 0.1 M for all compounds. Irradiation of the DNA-s2Ura mixture dissolved in D2O instead of H2O enhanced the rate of induction of SSBs in DNA by 334-nm light almost five times. Addition of superoxide dismutase (40, 80 and 200 μg/ml) decreased the rate of induction of breaks in DNA by 334-nm radiation plus s2Ura (in H2O) by about 40%. Boiled superoxide dismutase had no effect.  相似文献

Hydrogallation of Me3Si–C≡C–NR'2 with R2Ga–H (R = tBu, CH2tBu, iBu) yielded Ga/N‐based active Lewis pairs, R2Ga–C(SiMe3)=C(H)–NR'2 ( 7 ). The Ga and N atoms adopt cis‐positions at the C=C bonds and show weak Ga–N interactions. tBu2GaH and Me3Si–C≡C–N(C2H4)2NMe afforded under exposure of daylight the trifunctional digallium(II) compound [MeN(C2H4)2N](H)C=C(SiMe3)Ga(tBu)–Ga(tBu)C(SiMe3)=C(H)[N(C2H4)2NMe] ( 8 ), which results from elimination of isobutene and H2 and Ga–Ga bond formation. 8 was selectively obtained from the ynamine and [tBu(H)Ga–Ga(H)tBu]2[HGatBu2]2. 7a (R = tBu; NR'2 = 2,6‐Me2NC5H8) and H8C4N–C≡N afforded the adduct tBu2Ga‐C(SiMe3)=C(H)(2,6‐Me2NC5H8) · N≡C–NC4H8 ( 11 ) with the nitrile bound to gallium. The analogous ALP with harder Al atoms yielded an adduct of the nitrile dimer or oligomers of the nitrile at room temperature. The reaction of 7a with Ph–N=C=O led to the insertion of two NCO groups into the Ga–Cvinyl bond to yield a GaOCNCN heterocycle with Ga bound to O and N atoms ( 12 ).  相似文献
