Curvularia lunata is a dark pigmented fungus that is the causative agent of several diseases in plants and in both immunodeficient and immunocompetent patients. 1,8-Dihydroxynaphthalene-melanin is found in the cell wall of C. lunata and is believed to be the important virulence factor of dematiaceous fungi. Trihydroxynaphthalene reductase is an enzyme of the 1,8-dihydroxynaphthalene-melanin biosynthetic pathway, and it thus represents an emerging target for the development of novel fungicides and antimycotics. In the present study, we describe novel inhibitors of trihydroxynaphthalene reductase from C. lunata. These inhibitors were identified by ligand-based three-dimensional similarity searching and docking to a homology-built model and by subsequent biochemical and antifungal evaluation. Discovery of competitive inhibitors with K(i) values in low micromolar and even nanomolar concentration range proves the aplicability of homology-built model of 3HNR for hit finding by virtual screening methods. 相似文献

An improved method for the isolation and purification of pyrrolizidine alkaloids from comfrey (Symphytum officinale L.) roots was developed, introducing very fast, selective and ion residue-free reduction of N-oxides followed by ion-exchange chromatography giving a non-aqueous solution of alkaloids, from which solvents can be easily removed. With this procedure the use of large volumes of organic solvents, very slow reduction of N-oxides and input of additional impurities was avoided. Lycopsamine, which proved to be the major alkaloid, was additionally purified by preparative layer chromatography (PLC) and high performance liquid chromatography (HPLC). The identity of the alkaloid was confirmed by (I)H NMR spectroscopy and mass spectrometry. 相似文献

Polymer molar mass distributions critically affect macroscopic characteristics and performance of polymeric materials. While multi-detector methods coupled to size exclusion chromatography (SEC) are widely used to measure endproduct mass distributions, less progress has been made in simultaneously controlling and verifying the evolution of these distributions during synthesis. This work focuses on quantitative predictions and online verification of conversion kinetics and of molecular weight during free radical homopolymerization of acrylamide, where reagents were fed into the reactor during the reaction. The central task is to establish and experimentally test a formalism combining free radical polymerization kinetics with time dependent processes related to flows of material into and out of the reactor. Monomer feed experiments were performed that alternately hold molecular weight constant and ramp the weight up, in contrast to batch reactions, where molecular weight decreases. Three types of initiator feed ‘tapers’ were also used to produce predictable conversion kinetics and mass distributions: (i) constant initiator feed, (ii) linearly stepped feed to produce Gaussian conversion kinetics, and (iii) booster shots to produce multi-modal masses. Automatic Continuous Online Monitoring of Polymerization reactions (ACOMP) was used to follow the conversion and evolution of the average mass distribution, and multi-detector SEC was used to cross-check results and measure full distributions of endproducts. In general, there was good agreement between the predictions and results. In future work this approach can be used as an Ansatz for reaction trajectory prediction, and the online monitoring signals exploited to make feedback controlled corrections to the reagent flows and other reaction conditions. 相似文献

The versatile novel instrument for rotation planar extraction and rotation planar chromatography was exploited for the investigation of oak bark (Quercus robur L.). The same instrument enabled extraction of the bark, analytical proof of (+)-catechin directly in the crude extract and also its fractionation. Additionally, epimeric flavan-3-ols, (+)-catechin and (-)-epicatechin were separated by analytical ultra-micro rotation planar chromatography on cellulose plates with pure water as developing solvent. A comparison of the extraction of oak bark with 80% aqueous methanol by rotation planar extraction and medium pressure solid-liquid extraction was carried out and both techniques were shown to be suitable for the efficient extraction of oak bark. The raw extracts and fractions on thin-layer chromatography showed many compounds that possessed antioxidant activity after spraying with 1,1-diphenyl-2-picrylhydrazyl. Rotation planar fractionation of 840 mg of crude oak bark extract on silica gel gave 6.7 mg of pure (+)-catechin in one run. 相似文献

Genetic engineering allows modification of bacterial and bacteriophage genes, which code for surface proteins, enabling display
of random peptides on the surface of these microbial vectors. Biologic peptide libraries thus formed are used for high-throughput
screening of clones bearing peptides with high affinity for target proteins. There are reports of many successful affinity
selections performed with phage display libraries and substantially fewer cases describing the use of bacterial display systems.
In theory, bacterial display has some advantages over phage display, but the two systems have never been experimentally compared.
We tested both techniques in selecting streptavidin-binding peptides from two commercially available libraries. Under similar

ABSTRACTWe report on high-resolution calorimetry and small-angle X-ray scattering measurements along the Smectic-A to chiral Smectic-C* phase transition of the liquid crystal 4-(2-methyl butyl) phenyl 4-n-octylbiphenyl-4-carboxylate with dispersed, CoPt nanoparticle-coated reduced-graphene oxide nanosheets. The temperature dependence of heat capacity and smectic layer spacing are obtained in the vicinity of Smectic-A to chiral Smectic-C* phase transition. Though no remarkable pretransitional effects are present, the critical fits show a crossover from mean-field near a tricritical point to classical mean-field compared to pure liquid crystal. The X-ray data yield a dilation of smectic layer thickness, indicating the assembly of graphene oxide nanosheets between the smectic layers. 相似文献

Novel electrochemical microsensor based on mixed iron‐ruthenium hexacyanoferrate (FeRuHCF) modified carbon fiber microelectrode (CFME) is presented for voltammetric and amperometric measurement of hydrogen peroxide at physiological pH. The FeRuHCF coating was electrochemically deposited using a one step procedure onto the substrate carbon fiber microelectrode by cycling the potential between 0.0 and +1.0 V (vs. Ag/AgCl) in a solution containing all precursor salts. The microsensor displayed good stability in neutral and alkaline media and had a nonstop working lifetime of up to 12 hours. The amperometric response time varied from 5 to 15 s depending on the hydrogen peroxide concentration level. The newly developed electrochemical microsensor exhibited a highly linear behavior in the examined concentration range from 5 to 1000 μmol L?1 (R2=0.999), an LoD (3σ) of 0.9 μmol L?1, and a favorable reproducibility with a calculated RSD of 2.9% (n= 6) for 100 μmol L?1 hydrogen peroxide, thus holding great promise for its further application in real samples and its exploitation in combination with biorecognition elements in advanced microbiosensor design. 相似文献

The application of protective overoxidized poly‐1‐naphtylamine membrane (ONAP) is demonstrated in combination with bismuth film microelectrode (ONAP‐BiFME) for anodic stripping voltammetric measurement of trace heavy metals in the presence of some selected surfactants. The ONAP membrane was electrochemically deposited on the surface of bare single carbon fiber microelectrode followed by the in situ or ex situ preparation of the bismuth film. The key operational parameters influencing the stripping performance of the ONAP‐BiFME were optimized and its electroanalytical performance was examined in the model solution containing Cd(II) and Pb(II) as test metal ions. The ONAP‐BiFME exhibited significantly enhanced stripping voltammetric response (approximately 70% for Cd(II) and 45% for Pb(II)) in comparison with unmodified BiFME in the absence of surfactants. In the presence of high concentrations, e.g., 20 mg L?1, of anionic or cationic surfactants, the stripping signal for, e.g., Cd(II) decreased for less than 6% at the ONAP‐BiFME, whereas at the unmodified BiFME the signal attenuated considerably (approximately 38%). Moreover, in the presence of 10 mg L?1 of nonionic surfactant Triton X‐100, the stripping signals at the bare BiFME were almost completely suppressed, whereas at the ONAP‐BiFME exhibited linear concentration behavior in the examined concentration range from 10 to 120 μg L?1, with the calculated limit of detection of 5.0 μg L?1 and 3.4 μg L?1 for Cd(II) and Pb(II), respectively in connection with 60 s accumulation time. The attractive behavior of ONAP‐modified BiFME expands the applicability of bismuth‐based electrodes for measurement of trace heavy metals in real environments, where the presence of more complex matrix can be expected. 相似文献

Cytotoxic effects of micrometer-sized polymer capsules composed out of alternating layers of polystyrenesulfonate (PSS) and polyallylamine hydrochloride (PAH) on a fibroblast cell line have been investigated with an adhesion assay. For the purpose of visualization with fluorescence nanometer-sized CdTe nanoparticles have been embedded in the walls of the capsules. Similar to free CdTe nanoparticles, toxic Cd-ions are also released from CdTe nanoparticles that have been embedded in capsules. At high capsule concentrations, the capsules start to sediment on top of the cells and thus impair cell viability. 相似文献
