Heterofunctional azide and alkyne PEG-linkers have been synthesized and site specifically conjugated to scFv via a reactive thiol functionality; two scFv were coupled by copper catalyzed 1,3-dipolar cycloaddition to make divalent scFv (di-scFv) with an inter-scFv distance defined to provide divalent binding; antigen binding was maintained for the di-scFv construct and increased several times compared to that of the parent scFv; the cycloaddition reaction reported herein represents an important ligation strategy to covalently link macromolecular proteins and retain sensitive structural conformations.  相似文献

The non-symmetric imide ligand Hpypzca (N-(2-pyrazylcarbonyl)-2-pyridinecarboxamide) has been deliberately synthesised and used to produce nine first row transition metal complexes: [M(II)(pypzca)(2)], M = Zn, Cu, Ni, Co, Fe; [M(III)(pypzca)(2)]Y, M = Co and Y = BF(4), M = Fe and Y = ClO(4); [Cu(II)(pypzca)(H(2)O)(2)]BF(4), [Mn(II)(pypzca)(Cl)(2)]HNEt(3). These are the first deliberately prepared complexes of a non-symmetric imide ligand. X-ray crystal structures of [Cu(II)(pypzca)(2)]·H(2)O, [Co(II)(pypzca)(2)], [Co(III)(pypzca)(2)]BF(4), [Cu(II)(pypzca)(H(2)O)(2)]BF(4)·H(2)O and [Mn(II)(pypzca)Cl(2)]HNEt(3) show that each of the (pypzca)(-) ligands binds in a meridional fashion via the N(3) donors. In the first three complexes, two such ligands are bound such that the 'spare' pyrazine nitrogen atoms are positioned approximately orthogonally to one another and also to the imide oxygen atoms. In MeCN the [M(II/III)(pypzca)(2)](0/+) complexes, where M = Ni, Co or Fe, exhibit one reversible metal based M(II/III) process and two distinct, quasi-reversible ligand based reduction processes, the latter also observed for M(II) = Zn. [Mn(II)(pypzca)Cl(2)]HNEt(3) displays a quasi-reversible oxidation process in MeCN, along with several irreversible processes. Both copper(II) complexes show only irreversible processes. Variable temperature magnetic measurements show that [Fe(III)(pypzca)(2)]ClO(4) undergoes a gradual spin crossover from partially high spin at 298 K (3.00 BM) to fully low spin at 2 K (1.96 BM), and that [Co(II)(pypzca)(2)] remains high spin from 298 to 4 K. All of the complexes are weakly coloured, other than [Fe(II)(pypzca)(2)] which is dark purple and absorbs strongly in the visible region.  相似文献

Attempts to use alkylation to introduce a positive charge at the nitrogen atom of the 4‐pyridyl ring in the bis(bidentate) triazole ligand N4‐(4‐pyridyl)‐3,5‐di(2‐pyridyl)‐1,2,4‐triazole ( pydpt ) were made to ascertain what effect a strongly electron‐withdrawing group would have on the magnetic properties of any subsequent iron(II) complexes. Alkylation of pydpt under relatively mild conditions led in some cases to unexpected rearrangement products. Specifically, when benzyl bromide is used as the alkylating agent, and the reaction is carried out in refluxing acetonitrile, the N4 substituent moves to the N1 position. However, when the same reaction is performed in dichloromethane at room temperature, the rearrangement does not occur and the desired product containing an alkylated N4 substituent is obtained. Heating a pure sample of N4‐Bzpydpt?Br to reflux in MeCN resulted in clean conversion to N1Bzpydpt.Br . This is consistent with N4‐Bzpydpt.Br being the kinetic product whereas N1Bzpydpt.Br is the thermodynamic product. When methyl iodide is used as the alkylating agent, the N4 to N1 rearrangement occurs even at room temperature, and at reflux pydpt is doubly alkylated. The observation of the lowest reported temperatures for an N4 to N1 rearrangement is due to this particular rearrangement involving nucleophilic aromatic substitution: a possible mechanism for this transformation is suggested.  相似文献

Two new methods based on FT–Raman spectroscopy, one simple, based on band intensity ratio, and the other using a partial least squares (PLS) regression model, are proposed to determine cellulose I crystallinity. In the simple method, crystallinity in cellulose I samples was determined based on univariate regression that was first developed using the Raman band intensity ratio of the 380 and 1,096 cm?1 bands. For calibration purposes, 80.5% crystalline and 120-min milled (0% crystalline) Whatman CC31 and six cellulose mixtures produced with crystallinities in the range 10.9–64% were used. When intensity ratios were plotted against crystallinities of the calibration set samples, the plot showed a linear correlation (coefficient of determination R 2 = 0.992). Average standard error calculated from replicate Raman acquisitions indicated that the cellulose Raman crystallinity model was reliable. Crystallinities of the cellulose mixtures samples were also calculated from X-ray diffractograms using the amorphous contribution subtraction (Segal) method and it was found that the Raman model was better. Additionally, using both Raman and X-ray techniques, sample crystallinities were determined from partially crystalline cellulose samples that were generated by grinding Whatman CC31 in a vibratory mill. The two techniques showed significant differences. In the second approach, successful Raman PLS regression models for crystallinity, covering the 0–80.5% range, were generated from the ten calibration set Raman spectra. Both univariate-Raman and WAXS determined crystallinities were used as references. The calibration models had strong relationships between determined and predicted crystallinity values (R 2 = 0.998 and 0.984, for univariate-Raman and WAXS referenced models, respectively). Compared to WAXS, univariate-Raman referenced model was found to be better (root mean square error of calibration (RMSEC) and root mean square error of prediction (RMSEP) values of 6.1 and 7.9% vs. 1.8 and 3.3%, respectively). It was concluded that either of the two Raman methods could be used for cellulose I crystallinity determination in cellulose samples.  相似文献

The ability of 2'-alpha-C-carboxyalkyl nucleosides to undergo an unusual two-step stereocontrolled nucleobase exchange process has been investigated. Upon silylation a protected 2'-deoxy-2'-alpha-C-(carboxymethyl)uridine derivative can undergo intramolecular displacement of the uracil base, by the 2'-carboxylic acid group, to form a pentofuranosyl gamma-lactone. Under identical conditions the homologous 2'-deoxy-2'-alpha-C-(carboxyethyl)uridine derivative does not yield the corresponding delta-lactone, but undergoes elimination of uracil to give the corresponding glycal. The pentofuranosyl gamma-lactone is a good substrate for nucleoside synthesis by the Vorbrüggen procedures and undergoes completely stereoselective ring opening with either pyrimidine or purine silylated nucleobases to give novel 2'-C-carboxymethyl beta-nucleosides in moderate to high yield.  相似文献

Switchable surfaces are highly useful materials with surface properties that change in response to external stimuli. These surfaces can be employed in both research and industrial applications, where the ability to actively control surface properties can be used to develop smart materials and intelligent surfaces. Herein, we review a range of surfaces in which hydrophobicity can be controlled. We present the principal ideas of surface switching, discuss recent developments, explore experimental issues and examine factors that influence surface switching, including the nature of the stimuli, the underlying material, the morphology of the surface and the surrounding environment. We have categorised switchable surfaces according to the stimuli that trigger changes in surface hydrophobicity. These are electrically, electrochemically, thermally, mechanically, photo- and environmentally inducible surfaces. In addition, we review the use of chemical reactions to modify the properties of switchable surfaces and produce changes in the molecular structure and nanoscale features of the surface.  相似文献

Energy distributions in the products of the ion-molecule reaction Cl? + HBr → HC1 + Br? have been studied using quasiclassical trajectories on a semi-empirical collinear potential energy surface. Vibrational energy is favored in the products. While some trajectories are long-lived, the kinematic factor of the light central atom prevents effective energy redistribution.  相似文献

A novel heterobimetallic alkynyl-bridged complex, [Re(bpy)(CO)(3)(C[triple bond]C[bond]C(6)H(4)[bond]C[triple bond]C)Fe(C(5)Me(5))(dppe)], 1, and its oxidized species, [Re(bpy)(CO)(3)(C[triple bond]C[bond]C(6)H(4)[bond]C[triple bond]C)Fe(C(5)Me(5))(dppe)][PF(6)], 2, have been synthesized and their X-ray crystal structures determined. A related vinylidene complex, [Re(bpy)(CO)(3)(C[triple bond]C[bond]C(6)H(4)[bond](H)C[double bond]C)Fe(C(5)Me(5))(dppe)][PF(6)], 3, has also been synthesized and characterized. The cyclic voltammogram of 1 shows a quasireversible reduction couple at -1.49 V (vs SCE), a fully reversible oxidation at -0.19 V, and a quasireversible oxidation at +0.88 V. In accord with the electrochemical results, density-functional theory calculations on the hydrogen-substituted model complex Re(bpy)(CO)(3)(C[triple bond]C[bond]C(6)H(4)[bond]C[triple bond]C)Fe(C(5)H(5))(dHpe) (Cp = C(5)H(5), dHpe = H(2)P[bond](CH(2))(2)[bond]PH(2)) (1-H) show that the LUMO is mainly bipyridine ligand pi* in character while the HOMO is largely iron(II) d orbital in character. The electronic absorption spectrum of 1 shows low-energy absorption at 390 nm with a 420 nm shoulder in CH(2)Cl(2), while that of 2 exhibits less intense low-energy bands at 432 and 474 nm and additional low-energy bands in the NIR at ca. 830, 1389, and 1773 nm. Unlike the related luminescent rhenium(I)-alkynyl complex [Re(bpy)(CO)(3)(C[triple bond]C[bond]C(6)H(4)[bond]C[triple bond]C[bond]H)], 4, complex 1 is found to be nonemissive, and such a phenomenon is attributed to an intramolecular quenching of the emissive d pi(Re) --> pi*(bpy) (3)MLCT state by the low-lying MLCT and LF excited states of the iron moiety. Interestingly, switching on of the luminescence property derived from the d pi(Re) --> pi*(bpy) (3)MLCT state can be demonstrated in the oxidized species 2 and the related vinylidene analogue 3 due to the absence of the quenching pathway.  相似文献

Four bis-tetradentate N(4)-substituted-3,5-{bis[bis-N-(2-pyridinemethyl)]aminomethyl}-4H-1,2,4-triazole ligands, L(Tz1)-L(Tz4), differing only in the triazole N(4) substituent R (where R is amino, pyrrolyl, phenyl, or 4-tertbutylphenyl, respectively) have been synthesized, characterized, and reacted with M(II)(BF(4))(2)·6H(2)O (M(II) = Cu, Ni or Co) and Co(SCN)(2). Experiments using all 16 possible combinations of metal salt and L(TzR) were carried out: 14 pure complexes were obtained, 11 of which are dinuclear, while the other three are tetranuclear. The dinuclear complexes include two copper(II) complexes, [Cu(II)(2)(L(Tz2))(H(2)O)(4)](BF(4))(4) (2), [Cu(II)(2)(L(Tz4))(BF(4))(2)](BF(4))(2) (4); two nickel(II) complexes, [Ni(II)(2)(L(Tz1))(H(2)O)(3)(CH(3)CN)](BF(4))(4)·0.5(CH(3)CN) (5) and [Ni(II)(2)(L(Tz4))(H(2)O)(4)](BF(4))(4)·H(2)O (8); and seven cobalt(II) complexes, [Co(II)(2)(L(Tz1))(μ-BF(4))](BF(4))(3)·H(2)O (9), [Co(II)(2)(L(Tz2))(μ-BF(4))](BF(4))(3)·2H(2)O (10), [Co(II)(2)(L(Tz3))(H(2)O)(2)](BF(4))(4) (11), [Co(II)(2)(L(Tz4))(μ-BF(4))](BF(4))(3)·3H(2)O (12), [Co(II)(2)(L(Tz1))(SCN)(4)]·3H(2)O (13), [Co(II)(2)(L(Tz2))(SCN)(4)]·2H(2)O (14), and [Co(II)(2)(L(Tz3))(SCN)(4)]·H(2)O (15). The tetranuclear complexes are [Cu(II)(4)(L(Tz1))(2)(H(2)O)(2)(BF(4))(2)](BF(4))(6) (1), [Cu(II)(4)(L(Tz3))(2)(H(2)O)(2)(μ-F)(2)](BF(4))(6)·0.5H(2)O (3), and [Ni(II)(4)(L(Tz3))(2)(H(2)O)(4)(μ-F(2))](BF(4))(6)·6.5H(2)O (7). Single crystal X-ray structure determinations revealed different solvent content from that found by microanalysis of the bulk sample after drying under a vacuum and confirmed that 5', 8', 9', 11', 12', and 15' are dinuclear while 1' and 7' are tetranuclear. As expected, magnetic measurements showed that weak antiferromagnetic intracomplex interactions are present in 1, 2, 4, 7, and 8, stabilizing a singlet spin ground state. All seven of the dinuclear cobalt(II) complexes, 9-15, have similar magnetic behavior and remain in the [HS-HS] state between 300 and 1.8 K.  相似文献

We consider the existence of simple closed geodesics or “geodesic knots” in finite volume orientable hyperbolic 3-manifolds. Every such manifold contains at least one geodesic knot by results of Adams, Hass and Scott in (Adams et al. Bull. London Math. Soc. 31: 81–86, 1999). In (Kuhlmann Algebr. Geom. Topol. 6: 2151–2162, 2006) we showed that every cusped orientable hyperbolic 3-manifold in fact contains infinitely many geodesic knots. In this paper we consider the closed manifold case, and show that if a closed orientable hyperbolic 3-manifold satisfies certain geometric and arithmetic conditions, then it contains infinitely many geodesic knots. The conditions on the manifold can be checked computationally, and have been verified for many manifolds in the Hodgson-Weeks census of closed hyperbolic 3-manifolds. Our proof is constructive, and the infinite family of geodesic knots spiral around a short simple closed geodesic in the manifold.   相似文献
