Investigation of the influence of cell density of human fibroblasts cryopreserved inside collagen sponges at various cooling rates

The influence of cell density of cells cryopreserved inside a collagen matrix at various cooling rates was investigated. Human fibroblasts were three-dimensionally cultured for 2 days in a collagen sponge (20 mm in diameter and 1 mm in thickness) as an extracellular matrix to imitate biological tissue (artificial tissue). Different cell densities for the artificial tissue were used, from 10(5) to 10(7) cells/cm(3). Four artificial tissues were first stacked in a test chamber, frozen at a cooling rate of 0.3 to 50 degrees C/min in a solution of Dulbecco's Modified Eagle Medium, 20% fetal bovine serum and 10% dimethylsulfoxide, kept frozen below -185 degrees C for 2 hours, and then finally thawed. Membrane integrity of fibroblasts using a trypan blue exclusion assay was evaluated as an index for post-thaw cellular viability. Results show that with increasing cell density, the post-thaw membrane integrity decreased. Therefore, in the cryopreservation of biological tissue, it seems high cell density is one factor which causes a decline in viability.     

Rapid and Efficient Copper‐Catalyzed Finkelstein Reaction of (Hetero)Aromatics under Continuous‐Flow Conditions

A general, rapid, and efficient method for the copper‐catalyzed Finkelstein reaction of (hetero)aromatics has been developed using continuous flow to generate a variety of aryl iodides. The described method can tolerate a broad spectrum of functional groups, including N‐H and O‐H groups. Additionally, in lieu of isolation, the aryl iodide solutions were used in two distinct multistep continuous‐flow processes (amidation and Mg–I exchange/nucleophilic addition) to demonstrate the flexibility of this method.     

Asymmetric Total Synthesis of ent‐Pyripyropene A

An asymmetric total synthesis of ent‐pyripyropene A was achieved by a convergent synthetic route. We used our originally developed Ti^III‐catalyzed radical cyclization to construct an AB‐ring portion that consisted of a trans‐decalin skeleton with five contiguous stereogenic centers. The coupling between the AB‐ring and the DE‐ring portions, and a subsequent C‐ring cyclization, led to the total synthesis of ent‐pyripyropene A. An evaluation of the insecticidal activity of ent‐pyripyropene A against two aphid species revealed that ent‐pyripyropene A was 35–175 times less active than naturally occurring pyripyropene A. This result indicated that the biological target of pyripyropene A recognizes the absolute configuration of pyripyropene A.     

A general extension theorem for cohomology classes on non reduced analytic subspaces

The main purpose of this paper is to generalize the celebrated L~2 extension theorem of Ohsawa and Takegoshi in several directions: The holomorphic sections to extend are taken in a possibly singular hermitian line bundle, the subvariety from which the extension is performed may be non reduced, the ambient manifold is K¨ahler and holomorphically convex, but not necessarily compact.     

Rational development of caged-biotin protein-labeling agents and some applications in live cells

Biotin-(strept)avidin complex is widely used in biotechnology because of its extremely high binding constant, but there is no report describing spatiotemporally controlled formation of the complex in live cells. Here, based on X-ray crystal structure analysis and calorimetric data, we designed and synthesized photoreleasable biotins, which show greatly reduced affinity for (strept)avidin, but recover native affinity after UV irradiation. For application at the cell surface, we introduced an amine-reactive moiety into these "caged" biotin molecules. Specific fluorescence imaging of live cells that had been labeled with these agents and then UV-irradiated, was accomplished by addition of streptavidin conjugated with a fluorophore. We also demonstrated the applicability of these compounds for UV-irradiated-cell-specific drug delivery by using caged-biotin-labeled cells, a prodrug, and streptavidin conjugated with a prodrug-activating enzyme.     

H(x)TeV9O28]((5-x)-) (x=1 and 2): vanadotellurates with decavanadate structure

Two new vanadotellurates, [HTeV(9)O(28)](4-) and [H(2)TeV(9)O(28)](3-) have been synthesized and structurally characterized as tetra-n-butylammonium (TBA) salts: TBA(4)[HTeV(9)O(28)]·2CH(3)CN [triclinic, space group P ?1, a = 16.7102(6) ?, b = 17.4680(7) ?, c = 17.9634(7) ?, α = 74.412(1)°, β = 67.494(1)°, γ = 74.160(2)°, Z = 2] and TBA(3)[H(2)TeV(9)O(28)] [monoclinic, space group P2(1)/c, a = 13.0013(5) ?, b = 19.157(1) ?, c = 28.453(1) ?, β = 97.222(2)°, Z = 4]. The results of the structural analyses indicate that the four O atoms that bridge two V atoms on the Te side are the most basic ones in the structure. The results of density-functional theory (DFT) calculations support this view.     

Thermomechanical properties and phase structure of epoxy/silica nano‐hybrid materials constructed from a linear silicone oligomer

Epoxy‐grafted silicone oligomer (ESO), which has a linear silicone chain in the backbone moiety, was synthesized from a trifunctional alkoxysilane via a sol–gel reaction. Characterization of ESO was performed with ¹H and ²⁹Si NMR, Fourier transform infrared, and gel permeation chromatography. The number‐average molecular weight of ESO was 3300. By adding the silicone oligomer as the inorganic source in the curing process of the epoxy resin, novel epoxy/silica hybrid materials were prepared. It was observed by transmission electron microscope that fine silica‐rich domains of about 5‐nm diameter were uniformly dispersed in the cured epoxy matrix. Thermomechanical properties of the hybrid materials were also investigated. The storage modulus in the rubbery region and the peak area of the tan δ curve at the glass‐transition region increased and decreased, respectively, with the hybridization of the silica network. The mobility of the epoxy network chains should be considerably suppressed by the hybridization with the silica network. © 2005 Wiley Periodicals, Inc. Synthesis,p-Nitrophenolate Complexation and Competitive Anion Signaling of Novel Calixpyrrole Dimer

A novel dimeric calixpyrrole has been synthesized. The dimer forms stable complexes with p-nitrophenolate ion with a concomitant reduction in extinction coefficient. The chromogenic anion is displaced by the addition of various anions like fluoride and acetate. Effective optical sensing of these anions is accomplished using the calixpyrrole dimer.     

A new synthesis of 7-substituted 2,3-bis(alkylthio)naphth[1,8-bc]-azepines using the tris(alkylthio)cyclopropenyl cation

7-Substituted 2,3-bis(isopropylthio)naphth[1,8-bc]azepines 3a-c were synthesized in good yields by the reactions of 5-substituted 1-naphthylamines 2a-c with tris(isopropylthio)cyclopropenylium perchlorate ( 1 ) in acetonitrile under reflux. This reaction proceeds through the facile ring opening of 1 , followed by the intermediary formation of iminium salts 5a-c and then intramolecular cyclization.
