The membrane properties of the ganglioside GM1 (GM1)/dioleoylphosphatidylcholine (DOPC) binary system and GM1/dipalmitoylphosphatidylcholine (DPPC)/DOPC ternary system were investigated using surface pressure measurements and atomic force microscopy (AFM), and the effect of surface pressure on the properties of the membranes was examined. Mixed GM1/DPPC/DOPC monolayers were deposited on mica using the Langmuir-Blodgett technique for AFM. GM1 and DOPC were immiscible and phase-separated. The AFM image of the GM1/DOPC (1:1) monolayer showed island-like GM1 domains embedded in the DOPC matrix. There was no morphological change on varying surface pressure. The surface pressure-area isotherm of the GM1/DPPC/DOPC (2:9:9) monolayer showed a two-step collapse as in the DPPC/DOPC (1:1) monolayer. The AFM image for the GM1/DPPC/DOPC monolayer showed DPPC and GM1 domains in the DOPC matrix, and the DPPC-rich phase containing GM1 showed a percolation pattern the same as the GM1/DPPC (1:9) monolayer. The percolation pattern in the GM1/DPPC/DOPC monolayer changed as the surface pressure was varied. The surface pressure-responsive change in morphology of GM1 was affected by the surrounding environment, suggesting that the GM1 localized in each organ has a specific role.  相似文献

A cell culture-based in vitro bioassay was developed to measure the biological activity of recombinant human interleukin-11 (rhIL-11). The bioassay measures induced proliferation of T10 cells, derived from the T1165 murine plasmacytoma line. A colorimetrically detectable formazan product, obtained by cellular reduction of the tetrazolium compound, 4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulfonate (WST-1), was used as an endpoint for response of clone T10 to added rhIL-11. Positions of the samples and the standards in 96-well microplates affected the precision of this bioassay, which was improved by using 2 microplates where serially diluted sample and standard lines were interleaved and their positions were alternated. The coefficient of variation for this bioassay was less than 8%. This method is suitable for quality control of rhIL-11 because of its simplicity, reproducibility, and accuracy.  相似文献

Four new halogenated chamigrenes have been isolated from the sea hare, the (E)- and (Z)-9-(bromomethylidene)-1,2,5-trimethylspiro[5.5]undeca-1,7-dien-3-ones ( 11 and 10 , resp.) the structures of which were deduced by NMR spectroscopy and by comparison with a known relative, the (8R,9R)-8-bromo-9-chloro-5,5,9-trimethyl-1-methylidenespiro[5,5]undec-3-en-2-one ( 12 ), and its (11 R)-11-acetoxy-substituted derivative 13 . Their structures and absolute configurations were determined and deduced by X-ray using the absolute-structure parameter. The configurations were remarkable in being enantiomeric to those of the tertiary chlorochamigrenes isolated so far. Consequently, a more general scheme is proposed to account for their biogenesis.  相似文献

In order to investigate the physical and chemical effects of neutron capture reaction, a neutron in-beam Mössbauer spectroscopic study on two isomorphs of iron disulfide: pyrite and marcasite, were carried out with a parallel plate avalanche counter at room temperature. In both compounds only two major products accounted for the obtained spectrum: one with Mössbauer parameters close to the parent compound and the other one considered to be a new product. The yield of the parent-like species was different in the two isomorphs.  相似文献

Boiling-water extract of Korean-Saiko (Bupleuri Radix, from South Korea, Bupleurum falcatum L.) enhanced the mutagenic activity of Trp-P-1, Trp-P-2 and benzo[a]pyrene with S9mix. The boiling-water extract was fractionated with ether and then n-BuOH. Both the ether and the n-BuOH fractions also enhanced mutagenicity of Trp-P-1, respectively. The n-BuOH fraction was separated into seven fractions by silica gel chromatography and the chloroform eluate had the strongest enhancing effect on the mutagenic activity of Trp-P-1 with S9mix. The chloroform eluate fraction was further separated into five spots by thin-layer chromatography. Two of the spots had the strongest enhancing effect on the mutagenic activity of Trp-P-1. Since saikosaponin is a well known component in Bupleuri Radix, the effects of its existence were tested and saikosaponins a and c were found. The enhancement activity of saikosaponin a was very weak. The effective components are now being studied.  相似文献

Rh-Catalyzed cyclization was applied to the formation of a chiral quaternary carbon. It has become clear that the Rh-complex can discriminate between isopropenyl and 2-isopentenyl (or isopentyl) substituents, and the cyclization afforded 3,3,4-trisubstituted cyclopentanones with a chiral quaternary carbon in a stereoselective manner. The cyclization of 4-pentenals 6a, b by an achiral neutral Rh(PPh3)3Cl afforded 3,3,4-cis-trisubstituted cyclopentanones (+/-)-7a,b in 86-96%, and the cyclization by a cationic Rh[(R)-BINAP]CIO4 afforded 3,3,4-trans-trisubstituted cyclopentanones (-)-8a, b of 82-86% ee in 88-98% yields. The mechanism of stereoselection by Rh-complexes is also discussed.  相似文献
