A rapid and sensitive immunoassay based on a sequential injection analysis (SIA) using magnetic microbeads for the determination of alkylphenol polyethoxylates (APnEOs) is described. An SIA system was constructed from a syringe pump, a switching valve, a flow-through type immunoreaction cell equipped with a photon counting unit and a neodymium magnet. Magnetic beads, to which an anti-APnEOs monoclonal antibody was immobilized, were used as a solid support in an immunoassay. The introduction, trapping and release of the magnetic beads in and from the immunoreaction cell were controlled by means of a neodymium magnet and adjusting the flow of a carrier solution. The immunoassay was based on an indirect competitive immunoreaction of an anti-APnEOs monoclonal antibody immobilized on the magnetic beads with a sample APnEOs and a horseradish peroxidase (HRP)-labeled APnEOs in the same sample solution, and was based on the subsequent chemiluminscence reaction of HRP on the magnetic microbeads with a luminol solution containing hydrogen peroxide and p-iodophenol. The anti-APnEOs antibody was immobilized on the magnetic microbeads by coupling the antibody with the magnetic beads after activation of a carboxylate moiety on the surface of the magnetic beads that had been coated with a polylactic acid film. The antibody immobilized magnetic beads were introduced in the immunoreaction cell and trapped in it by the neodymium magnet, which was equipped beneath the immunoreaction cell. An APnEOs sample solution containing the HRP-labeled APnEOs at a constant concentration, and a luminol solution containing hydrogen peroxide and p-iodophenol were sequentially introduced into the immunoreaction cell, according to an SIA programmed sequence. Chemiluminescence emission was monitored by means of a photon counting unit located at the upper side of the immunoreaction cell by collecting the emitted light with a lens. A typical sigmoidal calibration curve was obtained, when the logarithm of the concentration of APnEOs was plotted against the chemiluminescence intensity as the number of photons in 100 ms using standard APnEOs sample solutions at various concentrations (0–1000 ppb) under optimum conditions. The lower detection limit defined as IC80 is ca 10 ppb. The time required for analysis is less than 15 min per a sample. The present method was successfully applied to the determination of APnEOs in river water.  相似文献

A duplex surface enhanced Raman scattering (SERS)-based lateral flow immunosensor was established for the simultaneous detection of two common antibiotic residues including tetracycline and penicillin in milk. The newly synthesized Au@Ag nanoparticles were labeled with different Raman molecules including 5,5-dithiobis-2-nitrobenzoic acid (DTNB) or 4-mercaptobenzoic acid (MBA), followed by the conjugation of anti-tetracycline monoclonal antibody or anti-penicillin receptor, forming two kinds of SERS nanoprobes. The two nanoprobes can recognize tetracycline-BSA and ampicillin-BSA, respectively, which facilitates the simultaneous detection of the two types of antibiotics on a single test line. After optimization, detection limits of tetracycline and penicillin as low as 0.015 ng/mL and 0.010 ng/mL, respectively, were achieved. These values were far below those of most of other documented bio-analytical approaches. Moreover, the spiking test demonstrates an excellent assay accuracy with recoveries of 88.8% to 111.3%, and satisfactory assay precision with relative standard deviation below 16%. Consequently, the results demonstrate that the SERS-based lateral flow immunosensor developed in this study has the advantages of excellent assay sensitivity and remarkable multiplexing capability, thus it will have great application potential in food safety monitoring.  相似文献

一些缩氨硫脲及其衍生物对结核、麻风、风湿、疟疾、天花、真菌和某些肿瘤有一定的药理活性。缩氨硫脲的真菌活性是由于它的配位原子在生物体的新陈代谢中和真菌所需的金属离子螯合。缩氨硫脲具有与氨基酸、肽、蛋白质、酶、核糖核酸等生物配体相同的配位原子、被看作研究生物配体和痕量金属离子配位的优良模型。因为聚乙二醇醚链上的氧原子能与金属离子配位,所以在缩氨硫脲分子中连接上聚乙二醇醚链,增加缩氨硫脲分子中的配位原子种类和数目,也许能够增强螯合剂的配位能力,扩展其与金属离子的螯合围范。为此,我们将缩乙二醇二氯化物和苯环上带酰基的苯酚反应,制成化合物1_(a-h),然后再与硫代氨基脲缩合,得到醚链桥接的芳香醛缩氨硫脲2_(a-g)。它们的合成路线和主要结构如下:  相似文献

Limiting nitrogen supply has been routinely used as the master regulator to direct lipid biosynthesis. However, this strategy does not work with nitrogen-rich substrates, such as Jerusalem artichoke (JA), a fructose-based biomass, while it is difficult to obtain a high carbon-to-nitrogen (C/N) molar ratio. In this study, an alternative strategy to promote lipid accumulation by the oleaginous yeast Trichosporon fermentans CICC 1368 was developed by limiting phosphorous supply, and this strategy was implemented with JA hydrolysate as substrate. We showed that lipid accumulation was directly correlated with the C/P ratio of the culture media for T. fermentans. The time course of cell growth and lipid production was analyzed in a media with an initial C/P ratio of 6342, and the cellular lipid content could reach up to 48.5% of dry biomass. Moreover, JA hydrolysates were used as substrate for microbial lipid accumulation, under high C/P molar ratio condition, lipid yield, lipid content, and lipid coefficient increased by 10, 30, and 34%, respectively. It showed that by limiting phosphorus, the conversion of sugar into lipids can be improved effectively. Limiting phosphorus provides a promising solution to the problem of microbial lipid production with nitrogen-rich natural materials.  相似文献
