Low-frequency stimuli (40- to 1000-Hz tones) have been used to correlate the motion of the 8-to 9-kHz place of the chinchilla basilar membrane with the cochlear microphonics recorded at the round window and with the responses of auditory nerve fibers with appropriate characteristic frequency. At the lowest stimulus frequencies, maximum displacement of the basilar membrane toward scala tympani occurs in near synchrony with maximum rarefaction at the eardrum and maximum negativity at the round window; at higher frequencies, the mechanical and microphonic response phases progressively lag rarefaction, reaching - 240 deg at 1000 Hz. At most frequencies (40-1000 Hz) near-threshold neural responses, once corrected for neural travel-time and synaptic delays, somewhat lead (by some 40 deg) maximal scala tympani displacement and maximal negativity of the round window microphonics. The variation of sensitivity with frequency is similar for basilar membrane displacement and microphonic responses: Under open-bulla conditions, sensitivity is constant for frequencies between 100 and 1000 Hz; below 100 Hz, sensitivity decreases at rates close to 12 dB/oct toward lower frequencies. Neural response sensitivity matches BM displacement more closely than BM velocity.  相似文献

Reported here is the correlation between the transfection efficiency of cationic liposome/DNA complexes (lipoplexes) and the structural evolution that they undergo when interacting with anionic membrane lipids. Multicomponent lipoplexes, incorporating from three to six lipid species simultaneously, presented a much higher transfection efficiency than binary lipoplexes, which are more commonly used for gene-delivery purposes. The discovery that a high transfection efficiency can be achieved by employing multicomponent complexes at a lower-than-ever-before membrane charge density of lipoplexes was of primary significance. Synchrotron small-angle X-ray diffraction (SAXD) experiments showed that anionic liposomes made of dioleoylphosphatidylglycerol (DOPG) disintegrated the lamellar phase of lipoplexes. DNA unbinding was measured by electrophoresis on agarose gels. Most importantly, structural changes induced by anionic lipids strictly depended on the lipid composition of lipoplexes. We found evidence of the existence of three different regimes of stability related to the interaction between complexes and anionic membranes. Both unstable (with low membrane charge density, sigmaM) and highly stable lipoplexes (with high sigmaM) exhibited low transfection efficiency whereas highly efficient multicomponent lipoplexes exhibited an "optimal stability". This intermediate regime reflects a compromise between two opposing constraints: protection of DNA in the cytosol and endosomal escape. Here we advance the concept that structural stability, upon interaction with cellular anionic lipids, is a key factor governing the transfection efficiency of lipoplexes. Possible molecular mechanisms underlying experimental observations are also discussed.  相似文献

Contemporary theories of antimatter have a number of insufficiencies which stimulated the recent construction of the new isodual theory based on a certain anti-isomorphic map of all (classical and quantum) formulations of matter called isoduality. In this note we show that the isodual theory predicts that antimatter emits a new light, called isodual light, which can be distinguished from the ordinary light emitted by matter via gravitational interactions (only). In particular, the isodual theory predicts that all stable antiparticles such as the isodual photon, the positron and the antiproton experience antigravity in the field of matter (defined as the reversal of the sign of the curvature tensor). The antihydrogen atom is therefore predicted to: experience antigravity in the field of Earth; emit the isodual photon; and have the same spectroscopy of the hydrogen atom, although subjected to an anti-isomorphic isodual map. In this note we also show that the isodual theory predicts that bound states of elementary particles and antiparticles (such as the positronium) experience ordinary gravitation in both fields of matter and antimatter, thus bypassing known objections against antigravity. A number of intriguing and fundamental, open theoretical and experimental problems of “the new physics of antimatter” are pointed out. In this paper we identify some of the most significant references on the inverse problem of the calculus of variations for single integrals and initiate the study of the generalization of the underlying methodology to classical field theories. We first classify Lorentz-covariant tensorial field equations into nonlinear, quasi-linear, and semilinear forms, and then introduce their systems of equations of variation and adjoint systems. The necessary and sufficient conditions for the self-adjointness of class C2, regular, tensorial, nonlinear, quasi-linear and semilinear forms are worked out. We study the Lagrange equations, their system of equations of variations (Jacobi equations) and their adjoint system by proving that, for class C4 and regular Lagrangian densities, they are always self-adjoint. We then introduce a concept of analytic representation which occurs when the Lagrange equations coincide with the field equations up to equivalence transformations and refine the definition by particularizing it as direct or indirect and ordered or nonordered. Some of the conventional cases of tensorial fields are considered and we prove, in particular, that the conventional representation of the complex scalar field in interaction with the electromagnetic field is of the ordered indirect type. For the objective of identifying our program we recall the two classes of equivalence transformations of the Lagrangian densities which are primarily used nowadays, namely, the Lorentz (coordinate) transformations and the gauge transformations (transformations of fields within a fixed coordinate system), and postulate the existence of a third class, which we term isotopic transformations of the Lagrangian density and which consist of equivalence transformations within a fixed coordinate system and gauge. We finally outline the objectives of our program, which essentially consist of the identification of the necessary and sufficient conditions for the existence of a Lagrangian in field theories and their first application to the transformation theory within the framework of our variational approach to self-adjointness.  相似文献

We propose a mechanism for DNA release from lipoplexes in cells that accounts for various observations of lipoplex-anionic lipid interactions. We examined the structural evolution of lipoplexes upon interaction with cellular lipids by synchrotron small-angle X-ray diffraction (SAXD), and the extent of DNA release from lipoplexes was determined by gel electrophoresis. We find that the interaction of lipoplexes with anionic cellular lipids is a two-stage process. In the first step, anionic lipids laterally diffuse into the complex and neutralize the cationic lipids. As a result, the membrane charge density of lipoplexes decreases and interactions between cationic lipids and DNA become weaker, but DNA is extremely poorly released. Only after the cationic charge of lipoplex membranes is completely neutralized by anionic lipids does DNA starts to be released significantly.  相似文献

Microemulsions have been widely used as microreactors for the synthesis of nanoparticles and mesoporous materials. The correlation between the microstructure of a microemulsion and the features of the obtained materials is the most intriguing problem. On this point, many investigations have their ground on the structure of the precursor microemulsion, i.e., the system before the reaction takes place. Nevertheless, any reactions usually involve the formation of byproducts (aside from the nanoparticles). Several of these byproducts (e.g., ions, amphiphilic molecules) could modify the microemulsion structure during the course of the reaction. Here we examine the hydrolysis of tetraethoxysilane (TEOS) in the water-in-oil microemulsion hexadecyl-trimethylammonium bromide (CTAB)/pentanol/hexane/water. Conductivity and NMR measurements performed during the course of the reaction, in combination with dynamic light scattering and pulsed field gradient spin-echo NMR investigation performed on the microemulsion upon the addition of ethanol, indicate that a byproduct (ethanol) modifies the microreactor structure. The ethanol produced by the TEOS hydrolysis drives the microemulsion structure from small disconnected reverse micelles toward large connected aggregates until (for high enough ethanol loading) the system phase separates into two coexisting liquid phases (a dense interconnected network and a dilute reverse micellar phase).  相似文献

We present an analysis of the motion of a simple torsion pendulum and we describe how, with straightforward extensions to the usual basic dynamical model, we succeed in explaining some unexpected features we found in our data, like the modulation of the torsion mode at a higher frequency and the frequency splitting of the swinging motion. Comparison with observed values yields estimates for the misalignment angles and other parameters of the model.  相似文献

The aim of this work was to investigate the surface structure of aqueous hematite dispersions characterized by a large variability of morphology and particle size combining structural investigations obtained from Atomic Force Microscopy (AFM) and Transmission Electron Microscopy (TEM) techniques with in vitro particle size distributions and zeta potential measurements from Dynamic Light Scattering (DLS) technique, and we achieved a self-consistent and detailed characterization of hematite particles whose sizes and morphologies could be correlated to the synthesis conditions (type of added anion, Al substitution and pH). Surface AFM characterization provided an accurate analysis of particle microstructure and also indicated that the growth of microcrystals followed different surface roughness. DLS, AFM, and TEM techniques furnished complementary information on the average particle dimensions, whose variation could be attributed to the morphological difference of hematites, ranging from platy to regular or irregular hexagonal or ellipsoidal shape. Finally, a correlation between the average particle dimensions and the measured zeta potential was also been found in aqueous dilute suspensions characterized by neither pH nor-ionic-strength-control, for which a drop of zeta potential from positive to negative values was detected for hematite particle dimensions larger than a threshold size of ~150 nm.  相似文献
