In these studies, we investigated the antioxidant activity of three ruthenium cyclopentadienyl complexes bearing different imidato ligands: (η5-cyclopentadienyl)Ru(CO)2-N-methoxysuccinimidato (1), (η5-cyclopentadienyl)Ru(CO)2-N-ethoxysuccinimidato (2), and (η5-cyclopentadienyl)Ru(CO)2-N-phthalimidato (3). We studied the effects of ruthenium complexes 1–3 at a low concentration of 50 µM on the viability and the cell cycle of peripheral blood mononuclear cells (PBMCs) and HL-60 leukemic cells exposed to oxidative stress induced by hydrogen peroxide (H2O2). Moreover, we examined the influence of these complexes on DNA oxidative damage, the level of reactive oxygen species (ROS), and superoxide dismutase (SOD) activity. We have observed that ruthenium complexes 1–3 increase the viability of both normal and cancer cells decreased by H2O2 and also alter the HL-60 cell cycle arrested by H2O2 in the sub-G1 phase. In addition, we have shown that ruthenium complexes reduce the levels of ROS and oxidative DNA damage in both cell types. They also restore SOD activity reduced by H2O2. Our results indicate that ruthenium complexes 1–3 bearing succinimidato and phthalimidato ligands have antioxidant activity without cytotoxic effect at low concentrations. For this reason, the ruthenium complexes studied by us should be considered interesting molecules with clinical potential that require further detailed research.  相似文献

Traditionally evolution is seen as a process where from a pool of possible variations of a population (e.g. biological species or industrial goods) a few variations get selected which survive and proliferate, whereas the others vanish. Survival probabilities and proliferation rates are typically associated with the ‘fitness’ of particular variations. In this paper we argue that the notion of fitness is an a posteriori concept, in the sense that one can assign higher fitness to species that survive but one can generally not derive or predict fitness per se. Proliferation rates can be measured, whereas fitness landscapes, i.e. the inter-dependence of proliferation rates, cannot. For this reason we think that in a physical theory of evolution such notions should be avoided. In this spirit, here we propose a random matrix model of evolution where selection mechanisms are encoded in interaction matrices of species, thereby extending the previous work of ours by a control parameter describing suppressors in the system. We are able to recover some key facts of evolution dynamics endogenously, such as punctuated equilibrium, i.e. the existence of intrinsic large extinction events, and, at the same time, periods of dramatic diversification, as known e.g. from the fossil record. Further, we comment on two fundamental technical problems of a ‘physics of evolution’, the non-closedness of its phase space and the problem of co-evolving boundary conditions, apparent in all systems subject to evolution.  相似文献

Until now, the potential effects of titanium dioxide (TiO2) nanoparticles on endothelial cells are not well understood, despite their already wide usage. Therefore, the present work characterizes six TiO2 nanoparticle samples in the size range of 19 × 17 to 87 × 13 nm, which are commonly present in sun protection agents with respect to their physicochemical properties (size, shape, ζ-potential, agglomeration, sedimentation, surface coating, and surface area), their interactions with serum proteins and biological impact on human microvascular endothelial cells (relative cellular dehydrogenase activity, adenosine triphosphate content, and monocyte chemoattractant protein-1 release). We observed no association of nanoparticle morphology with the agglomeration and sedimentation behavior and no variations of the ζ-potential (?14 to ?19 mV) in dependence on the surface coating. In general, the impact on endothelial cells was low and only detectable at concentrations of 100 μg/ml. Particles containing a rutile core and having rod-like shape had a stronger effect on cell metabolism than those with anatase core and elliptical shape (relative cellular dehydrogenase activity after 72 h: 60 vs. 90 %). Besides the morphology, the nanoparticle shell constitution was found to influence the metabolic activity of the cells. Upon cellular uptake, the nanoparticles were localized perinuclearly. Considering that in the in vivo situation endothelial cells would come in contact with considerably lower nanoparticle amounts than the lowest-observable adverse effects level (100 μg/ml), TiO2 nanoparticles can be considered as rather harmless to humans under the investigated conditions.  相似文献

The key role of the pentacene kinetic energy (Ek) in the early stages of growth on SiOx/Si is demonstrated: islands with smooth borders and increased coalescence differ remarkably from fractal-like thermal growth. Increasing Ek to 6.4 eV, the morphology evolves towards higher density of smaller islands. At higher coverage, coalescence grows with Ek up to a much more uniform, less defected monolayer. The growth, interpreted by the diffusion mediated model, shows the critical nucleus changing from 3 to 2 pentacene for Ek>5-6 eV. Optimal conditions to produce single crystalline films are envisaged.  相似文献

Experimental data are presented for the angular dependence of the relative flux, the mean energy and the speed ratio of deuterium molecules desorbing from a Ni(111) crystal surface at a surface temperature of Ts = 1143 K and at sulphur coverages ranging between 30% and less than 2% of a monolayer.The angular flux distribution is sharply peaked in the forward direction (cosdθwith 3 ? d ? 5) and the mean energy 〈E〉 of the desorbate depends strongly on the desorption angle θ. For normal desorption (θ = 0°) 〈E〉2k is about 700 K higher than Ts and for glancing angles (θ = 80°) it decreases to about 400 K below Ts The results obtained on sulphur free and sulphur covered Ni(111) surfaces are compared with our former data on polycrystalline nickel. The main differences in the kinetic features can be ascribed to the surface roughness. Accordingly, the angular distributions of flux, mean energy, and speed ratio, which deviate strongly from the Knudson and Maxwellian law, do not seem to depend considerably on sulphur coverage and surface structure. A qualitative explanation for these deviations is presented using the principle of detailed balancing.  相似文献

Photoemission was observed when the samples were irradiated with photons in the energy range from 2.5 to 3.3 eV from a tunable dye laser with an intensity of 108Wcm?2. The emission shows a quadratic intensity dependence. The variation with angle of incidence and polarization is different for the two surfaces. The result obtained from the (1 1 0) surface is discussed with help of the band structure as a two-photon surface photoelectric effect.  相似文献

We show that lim n → ∞ $ \frac{{c\left( n \right)}}{{2^n }} $ > $\frac{1}{2}$ , but can't decide whether the limit equals 1. Furthermore, for integers 1 < k < ? ≤ n we prove first results about c n (k, ?) ? max{|A| : AP n (k, ?)}, where P n (k, ?) = {A : A ? V n k and V n ? is cone independent of A} and V n k equals the set of binary sequences of length n and Hamming weight k.
