[reaction: see text] The three-carbon alpha,alpha'-annulation of a sterically hindered cyclic beta-keto ester can be achieved by alkynylation with 3,3-diethoxypropyne, syn reduction of the alkyne with Co(2)(CO)(8) and Et(3)SiH, and an intramolecular aldol reaction. The method is potentially useful for the synthesis of nemorosone, hyperforin, and other polycyclic polyprenylated acylphloroglucinols.  相似文献

Chromatographic separation plays a key role in the identification, quantification, and characterization of protein variants. Here we describe separation of species containing two post-translational modifications (glycosylation and methionine oxidation) in the Fc fragment of a monoclonal antibody. The method is based on cation-exchange chromatography under mildly acidic conditions that destabilize mainly the CH2 domain. Our data suggest that the separation is not mediated by the chemical modification itself, but rather by subtle structural changes induced by the chemical modification in the domain-decoupled conformation that monoclonal antibodies adopt around pH 4. Compared to other procedures already described in the literature, this method demonstrates an improved separation and allows purification of species in the native fold for additional functional characterization. This approach of separation under conditions where the protein assumes an alternative conformation could find a more general utility for the separation of chemical modifications in proteins.  相似文献

Grace-Davidson silica pellets (SMR-057-015) are found to be optimal for surface grafting of (RO)3Si(CH2)3FG reagents. While loadings of up to 2.1 mmol g−1 can be attained (ca. 0.05 mmol per pellet) access of further reagents to the graft sites is problematic above loadings of 0.8 mmol g−1. 13C CPMAS NMR studies may be carried out on individual pellets (using natural abundance substrates) and the resulting spectra are diagnostic in identifying successful subsequent coupling reactions.  相似文献

Background  

Olfactory systems create representations of the chemical world in the animal brain. Recordings of odour-evoked activity in the primary olfactory centres of vertebrates and insects have suggested similar rules for odour processing, in particular through spatial organization of chemical information in their functional units, the glomeruli. Similarity between odour representations can be extracted from across-glomerulus patterns in a wide range of species, from insects to vertebrates, but comparison of odour similarity in such diverse taxa has not been addressed. In the present study, we asked how 11 aliphatic odorants previously tested in honeybees and rats are represented in the antennal lobe of the ant Camponotus fellah, a social insect that relies on olfaction for food search and social communication.  相似文献

Abstract

Hyperspectral imaging is an optical method that provides a large amount of information about the investigated object. Its medical applications are reviewed in this article, including tumor delimitation and identification, assessing tissue perfusion and its pathological conditions (including some complications like diabetic foot ulceration), making accurate surgical decisions, evaluating the health of dental structures, etc. Many of the articles show very promising results that required brief comments by the authors. It is clear that choosing the appropriate hyperspectral imaging system for each medical field, together with the most reliable hyperspectral image processing methods, are the main goals of future studies, before hyperspectral imaging becomes a widely applicable evaluation method in medicine. The authors try to answer some questions on this topic and set up some directions for future research.  相似文献

Hexaalkoxy‐substituted azamacrocyclic metal complexes 1 · MXn [MXn = FeCl3, CuCl2, SrCl2, Ln(NO3)3; Ln = La, Pr, Eu, Ho, Er] with various chain lengths were prepared by a convergent approach using 1,2‐bisalkoxy‐4,5‐diamines 3 and 4‐alkoxy‐pyridine‐2,6‐dicarbaldehydes 4 as key building blocks for template‐assisted cyclocondensation. Metal complexes 1 · MXn were considered as potential metallomesogens. However, differential scanning calorimetry and optical polarizing microscopy of 1 · MXn did not reveal any mesomorphism. The magnetic susceptibility shows deviation from Curie‐like behavior. Due to ligand field effects the effective magnetic moments are a function of the temperature. In order to obtain structural informations on the free ligand 1 , which is synthetically not accessible, theoretical calculations were carried out. For the free azamacrocycle 2a and the free hexamethoxy‐substituted azamacrocycle 1a the IR spectra were computed at the level of density functional theory. A planar and a saddle‐shaped conformation was considered. The IR spectra and especially the dependence of the C=N vibration on the structural parameters and the charge distribution are discussed.  相似文献

A Systematic Study on the Coordination Properties of the Guanidine Ligand N1,N2‐Bis(1,3‐dimethylimidazolidin‐2‐ylidene)‐ethane‐1,2‐diamine with the Metals Mn, Co, Ni, Ag and Cu The syntheses and characterization of the compounds [Mn(DMEG2e)Cl2] ( 1 ), [Co(DMEG2e)Cl2] ( 2 ), [Ni(DMEG2e)2]I2 ( 3 ), [Cu(DMEG2e)I] ( 4 ) and {[Ag(DMEG2e)]BF4}n ( 5 ) with the bisguanidine ligand N1,N2‐bis(1,3‐dimethylimidazolidin‐2‐ylidene)ethane‐1,2‐diamine (DMEG2e) are described. All complexes are synthesized by the reaction of the corresponding metal salt with the DMEG2e ligand in MeCN or THF. The coordination of the metal atoms vary from a distorted tetrahedron in 1 and 2 , a distorted trigonal planar coordination in 4 to linear coordination in 5 . Contrasting to the compounds 1 , 2 , 4 and 5 which exhibit a 1:1 ratio of metal to ligand, two DMEG2e ligands are bound to the Ni atom in the case of 3 resulting in a coordination polyhedron which represents the stage exactly in the middle between the square‐planar and the tetrahedral geometry. Whereas crystals of 1 , 2 , 3 and 4 contain discrete molecules, in 5 the Ag atoms are alternately linked by two different DMEG2e ligands to form a chain structure. The comparative discussion of several DMEG2e containing complexes with the compounds reported herein supplements this systematic study.  相似文献
