Protein expression, aggregation, and triggered release from polymersomes as artificial cell-like structures

Bringing droplets to life: A cytoskeletal protein (red dots, see scheme) is expressed in artificial cells composed of biocompatible polymersomes, which encapsulate expression machinery and amino acid building blocks. Release of the expressed proteins can be triggered by a negative osmotic shock.     

Syntheses and degradations of fluorinated heterocyclics. IV. Imidoylamidoximes, 1,2,4-oxadiazole precursors

Perfluoroalkylether amidoximes free from amide-contamination were prepared from imidate esters. The amidoximes were stable at 110°C; at ～170°C partial decomposition to 1,2,4-oxadiazoles, admixed with other compounds, took place. Interactions between nitriles and amidoximes at 50°C resulted in the formation of imidoylamidoximes; these dissociated readily into their constituents when subjected to higher temperatures. At 70°C and above, in the presence of excess nitrile or other ammonia acceptors, the imidoylamidoximes afforded high yields of the corresponding 1,2,4- oxadiazoles.

Alkyldimesitylboranes, yield anions¹, Mes₂B$\text{C}$HR, that on reaction with oxiranes followed by oxidation give 1,3-diols. These anions are thus the operational equivalent of R$\text{C}$HOH. The scope and limitations of the new process are delineated.     

cis- and trans-stereoselective epoxidation of N-protected 2-cyclohexen-1-ylamines

The first systematic study of the cis and trans stereoselectivity in the m-CPBA epoxidation of N-protected cyclic allylic amines has been completed. Mono-N-protected systems gave epoxides with cis stereochemistry (amides are better cis directors than sulfonamides or carbamates) whereas di-N-protected systems gave trans-epoxides (TsNBoc protection gave complete trans stereoselectivity). Direct one-index transformations in multiconfiguration response calculations

The evaluation of gradient vectors of a general operator with one-index, double-one-index, or higher-order one-index transformed integrals is a key operation in multiconfiguration response theory calculations. We describe an integral-driven direct algorithm that carries out this operation efficiently without pretransforming the integrals. The use of this algorithm leads to a considerable reduction in disk space and timing. © 1994 by John Wiley & Sons, Inc.     

Design,synthesis of novel quinazolin-4-one derivatives and biological evaluation against human MCF-7 breast cancer cell line

A new series of 6,8-dibromo-2-(4-chlorophenyl)quinazolin-4(3H)-one derivatives VI–XIII were synthesized. Their chemical structures were confirmed by spectral and elemental analysis. The cytotoxic effect of the newly synthesized compounds was tested in vitro against human breast cancer cell line (MCF-7). Most of the tested compounds have shown promising cytotoxic activity. Compounds X and XIIIb exerted a powerful cytotoxic effect against MCF-7 with a very low IC50 (0.0015 and 0.0047 µmol/ml), while compounds VI, VII, VIII, XIIb, XI, XIIIc and IX exerted a moderate cytotoxic effect (IC50 0.01523, 0.0213, 0.031, 0.0478, 0.049, 0.068 and 0.079 µmol/ml respectively), compared to doxorubicin (0.0025 µmol/ml). Exploring their apoptotic effect; interestingly,all compounds activated apoptotic cascade in MCF-7. Compounds VI, XIIIb, XIIb, XI, XIIa, VII, V and VIII showed potent effect even much more than doxorubicin by 12.87–5.91 folds, while compounds XIIIc, IX, XIIIa, XIIc and X showed moderate increase in CASP3 activity by 4.96–3.22 folds relative to untreated cells more or less similar to doxorubicin (5.57 folds).     

Ultra-low-frequency dust-electromagnetic modes in self-gravitating magnetized dusty plasmas

Obliquely propagating altra-low-frequency dust-electromagnetic waves in a self-gravitating, warm, magnetized, two fluid dusty plasma system have been investigated. Two special cases, namely, dust-Alfvén mode propagating parallel to the external magnetic field and dustmagnetosonic mode propagating perpendicular to the external magnetic field have also been considered. It has been shown that effects of self-gravitational field, dust fluid temperature, and obliqueness significantly modify the dispersion properties of these ultra-low-frequency dust-electromagnetic modes. It is also found that in parallel propagating dust-Alfvén mode these effects play no role, but in obliquely propagating dust-Alfvén mode or perpendicular propagating dust-magnetosonic mode the effect of self-gravitational field plays destabilizing role whereas the effect of dust/ion fluid temperature plays stabilizing role.     

An integrated workflow for extraction and solubilization of intermediate filaments from colorectal biopsies for proteomic analysis

We report a technique for isolation and solubilization of intermediate filament (IF) proteins from colonic biopsies compatible with both gel electrophoresis and liquid chromatography "shotgun" proteomics using mass spectrometry (MS). This is important because changes in the IF proteome, particularly in keratin expression and modification, are noted in colonic mucosa of patients with colorectal cancer. Though keratins have traditionally been dissolved in high concentration of urea, the latter solvent precludes efficient proteolytic digestion by trypsin prior to gel-free LC-MS/MS approaches. The extraction of cytoskeletal proteins was initially evaluated using MCF-7 cancer cell lines using a published, differential detergent solubilization protocol. IF proteins were extracted from colonic biopsies using a combination of homogenization and sonication. Since comparable efficiency of solubilization was noted on the extracted IF from cell lines between urea and guanidine hydrochloride (GuHCl) in triethylammonium bicarbonate buffer, isolated proteins from endoscopic biopsies were solubilized in GuHCl. Using immunoblotting techniques, we successfully demonstrated isolation of keratins and preservation of posttranslational modifications (phosphorylation, acetylation). Dissolved proteins were tryptically digested and peptides analyzed by MS, showing the functionality of the workflow in shotgun proteomic applications, specifically compatibility of the workflow for isobaric tagging relative and absolute quantification based quantitation approaches.
