Among possible approaches proposed in the literature on this topic, we will focus on the electrosynthesis of MIPs and on less

In the last decade, the improved performance and versatility of the mass spectrometers together with the increasing availability of gene and genomic sequence database, led the mass spectrometry to become an indispensable tool for either protein and proteome analyses in cereals. Mass spectrometric works on prolamins have rapidly evolved from the determination of the molecular masses of proteins to the proteomic approaches aimed to a large-scale protein identification and study of functional and regulatory aspects of proteins. Mass spectrometry coupled with electrophoresis, chromatographic methods, and bioinformatics tools is currently making significant contributions to a better knowledge of the composition and structure of the cereal proteins and their structure-function relationships. Results obtained using mass spectrometry, including characterization of prolamins, investigation of the gluten toxicity for coeliac patients, identification of proteins responsible of cereal allergies, determination of the protein pattern and its modification under environmental or stress effects, investigation of genetically modified varieties by proteomic approaches, are summarized here, to illustrate current trends, analytical troubles and challenges, and suggest possible future perspectives. 相似文献

This article presents a multi-analytical approach to investigating the drying, polymerisation and oxidative degradation of
linseed oil, which had undergone various treatments known to be undertaken during the nineteenth century in preparation for
painting. The oil was mechanically extracted from the same seed lot then processed by different methods: water washing, heat
treatments, and the addition of driers, with and without heat. The oil was prepared in 1999 within the framework of the MOLART
project. We compared thermogravimetric analysis (TG), which yields macromolecular information, with gas-chromatography mass-spectrometry
(GC/MS) and direct exposure mass spectrometry (DE-MS), which provide molecular information. This comparison enabled us to
elucidate the role of pre-treatment on the composition of the oil. TG and oxygen uptake curves registered at a constant temperature
helped us to identify the different physical behaviour of the oil samples, thus highlighting the presence of hydrolysed, oxidised
and crosslinked fractions, as a consequence of the different pre-treatments. GC/MS was used to characterise the soluble and
non-polymeric fraction of the oil, to calculate the ratios of palmitic to stearic acid (P/S), and azelaic to palmitic acid
(A/P), and to further evaluate the effects of oil pre-treatments. DE-MS using chemical ionisation with CH4, enabled us to establish the chemical composition of the oil in different stages of ageing. DE-MS proved to be a useful tool
for a simultaneous semi-quantitative characterisation of the free fatty acids, monoglycerids, diglycerides and triglycerides
present in each sample. The combination of thermal analysis with GC/MS and DE-MS enabled a model to be developed, which unravelled

Diastereomers of antiinflammatory/analgesic and antihistaminic 3,3′[(1,2‐ethanediyl)bis(2‐aryl‐4‐thiazo‐lidinone)] derivatives possessing two stereogenic centers (indicated as BIS 2*C) have been widely investigated in recent years. The 5,5′‐dimethyl analogues (BIS 4*C), now reported, have been synthesized by reaction of (±) α‐mercaptopropionic acid and N,N'‐di(3‐fluorobenzyliden)ethylenediamine. Because the 2 and 2′carbons bear the same groups and similarly the 5 and 5′ carbons, and the latter groups are different from the former, four enantiomeric pairs and two meso forms exist in this situation. Tacle® is a citrus fruit obtained from the crossbreeding of Clementine and Tarocco cultivars. This fruit retains a promising nutraceutical potential most likely due to a high content in polyphenols, among which the main constituents are the two glycosides naringin and hesperidin. Herein, we evaluated, through an in vitro assay, the capability of Tacle extracts to inhibit the hydroxymethylglutaryl-CoA reductase enzyme, which plays a key role in cholesterol biosynthesis. The results obtained spurred us to investigate whether the anti-enzymatic activity observed may be due to a direct interaction of aglycones naringenin and hesperetin with the enzyme catalytic site. Molecular docking simulations indicated that these two compounds are able to anchor to the protein with binding modes and affinities similar to those found for statins, which represent mainstream medications against hypercholesterolemia. The overall results showed an interesting nutraceutical potential of Tacle, suggesting that its extract could be used for dietary supplementation in the treatment of moderate hypercholesterolemia. 相似文献

In a previous paper, we demonstrated the synergistic action of the anti-ischemic lubeluzole (Lube S) on the cytotoxic activity of doxorubicin (Dox) and paclitaxel in human ovarian cancer A2780 and lung cancer A549 cells. In the present paper, we extended in vitro the study to the multi-drug-resistant A2780/DX3 cell line to verify the hypothesis that the Dox and Lube S drug association may potentiate the antitumor activity of this anticancer compound also in the context of drug resistance. We also evaluated some possible mechanisms underlying this activity. We analyzed the antiproliferative activity in different cancer cell lines. Furthermore, apoptosis, Dox accumulation, MDR1 downregulation, ROS, and NO production in A2780/DX3 cells were also evaluated. Our results confirm that Lube S improves Dox antiproliferative and apoptotic activities through different mechanisms of action, all of which may contribute to the final antitumor effect. Moderate stereoselectivity was found, with Lube S significantly more effective than its enantiomer (Lube R) and the corresponding racemate (Lube S/R). Docking simulation studies on the ABCB1 Cryo-EM structure supported the hypothesis that Lube S forms a stable MDR1-Dox-Lube S complex, which hampers the protein transmembrane domain flipping and blocks the efflux of Dox from resistant A2780/DX3 cells. In conclusion, our in vitro studies reinforce our previous hypothesis for repositioning the anti-ischemic Lube S as a potentiating agent in anticancer chemotherapy. 相似文献
