We present a centrifugal process for the extraction of plasma from sediment by a decanting structure, terminating with metered plasma which is readily available for subsequent on-disk processing. Our technique supplies 2 microl plasma from 5 microl of whole blood at moderate spinning frequencies of 40 Hz within 20 s, only. The residual cell concentration in the purified plasma amounts to less than 0.11%, independent of the frequency of rotation. A capillary duct connects the extracted plasma to subsequent on-disk processing units. 相似文献

A chemical probe was developed for the detection of the emerging cancer marker galectin-3. The probe contains a benzophenone moiety which covalently attaches itself to the protein upon binding and irradiation. Introduction of a fluorescent label via'click' chemistry allows the labelled proteins to be visualized in a gel. Novel propranolol analogues have been designed and synthesised and their enantioselective binding to the cellulose degrading enzyme, Cel7A, has been evaluated. Affinity and enantioselectivity have been determined by capillary electrophoresis experiments. Ligands with significantly improved affinity and selectivity have been obtained and an analysis of the results has led to insights concerning the relation between the changes in ligand structure and selectivity as well as affinity to the protein. 相似文献

A general approach is proposed to analyze electrical conductivities in aqueous solutions of polybasic organic acids. Experimental conductivities are examined in the context of dissociation and hydrolysis reactions by applying the Quint-Viallard conductivity equations and the Debye-Hückel equations for activity coefficients. The proposed numerical procedure is illustrated by the case of benzenehexacarboxylic (mellitic) acid and its neutral and acidic salts. From conductivity measurements of mellitic acid and its salts, performed in dilute aqueous solutions in the 278.15-308.15 K temperature range, the limiting conductances of mellitic anions, lambda(0)(1/jH(6-j)Mel(-j), T), j = 1, 2, 3, 4, 5, 6 are determined. 相似文献

Site-differentiated solvated clusters of the general formula [Re(6)(mu(3)-Se)(8)(PEt(3))(n)(MeCN)(6)(-)(n)](SbF(6))(2) (n = 4, cis and trans; n = 5) undergo ligand substitution reaction with isonicotinamide to afford the corresponding amide derivatives, [Re(6)(mu(3)-Se)(8)(PEt(3))(n)(isonicotinamide)(6)(-)(n)](2+) [1 (n = 5); 2 (n = 4, trans); 3 (n = 4, cis)]. Retention of stereochemistry in each case was confirmed by (1)H and (31)P NMR. The solid-state structures of all three compounds were established crystallographically, which revealed self-complementary hydrogen-bonding interactions between adjacent cluster units. While complex 1 exists as hydrogen-bonded dimers in the solid state, compounds 2 and 3 form one-dimensional chains of clusters bridged by paired hydrogen bonds. It is the rigid stereochemistry of the cluster, combined with the classic crystal engineering motif of complementary N-H.O amide hydrogen bonding, that affords the predictable solid-state structures and dimensionality. 相似文献

A combined temperature‐ and pressure‐dependent study was employed to reveal the conformational and free‐energy landscape of phenylalanine transfer RNA (tRNAPhe), a known model for RNA function, to elucidate the features that are essential in determining its stability. These studies also help explore its structural properties under extreme environmental conditions, such as low/high temperatures and high pressures. To this end, fluorescence and FTIR spectroscopies, calorimetric and small‐angle scattering measurements were carried out at different ion concentrations over a wide range of temperatures and pressures up to several hundred MPa. Compared with the pronounced temperature effect, the pressure‐dependent structural changes of tRNAPhe are small. A maximum of only 15 % unpaired bases is observed upon pressurization up to 1 GPa. RNA unfolding differs not only from protein unfolding, but also from DNA melting. Its pressure stability seems to be similar to that of noncanonical DNA structures. 相似文献
